摘    要:目的 研究NO在局灶性脑缺血再灌流过程中对病理改变的影响,并探讨其机制。方法 利用光、电镜及TUNEL法观察了NNLA干预后局灶脑缺血大鼠脑组织的病理学改变。结果 缺血、再灌流1d时光镜下皮质、海马区均可见神经细胞呈坏死样改变;电镜下半影区神经细胞的改变仅见于核染色质凝集(类似凋亡小体)和小胶质细胞核变形,以细胞凋亡为主;小剂量给予NNLA干预后坏死和凋亡的病理改变均较手术对照组轻。TUNEL显示与病理改变结果一致。结论 局灶性脑缺血后,调节NO生成量在适当范围,可以达到保护、防止神经细胞进一步损伤的目的。

Abstract:Objective To study the relation of nitric oxide (NO) and pathology changes in the focal cerebral ischemia. Methods Using of light microscope and electriomicroscope and TUNEL method the pathological changes were observed in the focal cerebral ischemia. Results In the 1d of focal cerebral ischemia and reperfusion for under light microscope,neuronal cell were necrostic in cortex region and hippocampus. Under electriomicroscope there were only chromatin dense stain (likely an apoptosis body) of neuronal cell nucleus and distortion of gliocyte nucleus in ischemic penumbra in the operation side of brain. They showed apoptosis. But the pathology changes of necrosis and apoptosis under the influence of nitric oxide synthase (NOS) inhibitor [ N nitro L arginine (NNLA)] were lighter than those in the focal cerebral ischemia. TUNEL results showed:The change of NO content influenced percent rate of apoptosis cells of DAB positive stain in infarction. Conclusion In the process of focal cerebral ischemia and reperfusion,regulating the quantity of NO in the propriety range may protect and avoid purpose of neuronal cell injury.
