离体异位子宫内膜细胞ki-67、钙粘素-E的表达及自发凋亡测定

摘    要:目的:检测原代培养异位内膜细胞ki-67、钙粘素-E的表达及自发凋亡率从而推测其增殖、凋亡特性及侵袭性。方法:对10例异位内膜组织(实验组)及10例正常内膜组织(对照组)进行原代培养,采用免疫细胞化学检测两组内膜ki-67及钙粘素-E表达水平,采用流式细胞术检测两组细胞的细胞凋亡指数。结果:1)腺上皮细胞和基质细胞的ki-67积分实验组与对照组均相当(P均 >0.05)。2)实验组的自发凋亡指数显著低于对照组(P <0.01)。3)实验组腺上皮细胞钙粘素-E表达积分(3.45±1.45)显著低于对照组(8.50±0.89)( P <0.01)。结论:1)异位内膜细胞并非高度增殖,而是表现为抗凋亡和抗侵袭性。2)异位内膜细胞的生物学特性在子宫内膜异位症的病理生理中可能起着重要作用。

Abstract:Objective: To detect ki-67 and E-cadherin expression and spontaneous apoptosis in primarycultured human endometriotic cells. Metholds: ki-67 and E-cadherin expression and the apoptotic index weredetected in primary cultured ectopic endometrial cells from women with endometriosis (experiment group, n=10)and normal euctopic endometrial cells (control group, n=10). The apoptic index was detected by flowcytometry.The ki-67 and E-cadherin expression were examined by s-p immunocytochemical stainings. Results:①The ki-67exprsssion in glandular epithelial cells and stromal cells of the experiment group did not have apparent differencewith that in control group (P>0.05). ②The apoptosis index was significantly lower in experiment group than incontrol group (P<0.01). ③ E-cadherin expression in endometirial epithelial cells of the experiment group wassignificantly lower than the control group (P<0.01). Conclution: ①The ectopic endometria cells are not over-proliferative, they are featured by anti-apoptosis and anti-invasion abilities. ②The unique biological characteristicsof ectopic endometrial cells might be involved in the molecular pathogenesis of endometriosis.
