通络止痛胶囊对佐剂性关节炎大鼠血清细胞因子的调节

目的：观察通络止痛胶囊对佐剂性关节炎（AA）大鼠血清白细胞介素-1（IL-1β）、IL-4、肿瘤坏死因子α（TNF-α）的影响。方法：将50只Wistar大鼠随机分为正常对照组、空白模型组和通络止痛胶囊高、中、低剂量治疗组（简称高剂量组、中剂量组、低剂量组）,每组10只,用ELISA法测定AA大鼠外周血细胞因子IL-1β、IL-4和放射免疫法测定TNF-α的表达情况。结果：与正常对照组相比,AA大鼠外周血中IL-1β、TNF-α显著升高（P〈0.01）,IL-4显著降低（P〈0.01）;通络止痛胶囊低、中、高剂量组大鼠血清IL-1β、TNF-α水平显著低于空白模型组,尤以高剂量组最显著（P〈0.01）,IL-4水平显著高于空白模型组,尤以高剂量组最显著（P〈0.01）。结论：通络止痛胶囊可能通过对细胞因子网络的调节,降低促炎性的细胞因子IL-1β和TNF-α水平,上调抗炎性的细胞因子IL-4水平,抑制细胞因子的促炎效应,增加抗炎效应,从而减轻滑膜炎症。

Effect of Tongluozhitong Capsule on the Cytokine in Adjuvant Arthritis Rats

Objective： To observe the effect of Tongluozhitong Capsule （TLZTC） on cytokine of intedetukin （IL）- 1β, IL-4, TNF-α in adjuvant arthritis（AA） rats. Methods：Fifty Wistar rats were randomly divided into blank control group,model control group,TLZTC low dosage group,TLZTC middle dosage group,TLZTC high dosage group, ten in each group. The content of IL-1β,IL-4 was measured with ELISA and TNF-α with radio-immunity. Results：The content of IL-1β and TNF-α in AA rats increased siguificantly（P〈0.01 ）,IL-4 decreased significantly（P〈0.01）compared with those of blank con- trol group. The content of IL-1β and TNF-α in TLZTC low,middle and high dosage group were lower than those of model control group with that of high dosage group dropped significantly（P〈0.01 ）. IL-4 level was higher than that of model control group with those of high dosage group increased significantly （P〈0.01）. Conclusion ：TLZTC of different dosage could relieve synovial inflammation by regulating cytokine network. It reduced inflammation promoting cytokine IL-1β and TNF- α and increased anti-inflammation cytokine to inhibit inflammation reaction and reinforce anti-inflammation reaction.
