反相高效液相色谱法研究大黄配伍附子后化学成分含量的变化

摘    要:目的建立反相高效液相色谱法同时测定大黄-附子药对中5个蒽醌成分——大黄酚、大黄素、大黄酸、芦荟大黄素和大黄素甲醚的含量,探讨在不同配伍比例下大黄中化学成分的总体变化。方法采用反相高效液相色谱法,DiamonsibC18色谱柱(4.6mm×200mm,5μm),流动相:甲醇-水(含1.0%醋酸)梯度洗脱,流速1.0mL·min^-1,检测波长:430nm。结果大黄酚、大黄素、大黄酸、芦荟大黄素和大黄素甲醚的线性范围分别为0.0106~0.2120、0.00272~0.0544、0.002520~0.05040、0.002560~0.05120、0.002760~0.05520mg·mL^-1,井日关系数分别为0.9995、0.9993、0.9996、0.9997、0.9993。平均加样回收率(n=5)分别为:(101.1%±1.8%)、(102.8%±2.6%)、(96.7%±1.5%)、(100.9%±2.1%)、(99.6%±2.4%)。通过欧式距离(又称二阶Minkowski)比较,大黄附子配伍比例1:1时与单味大黄的化学成分含量总体差异最大。结论该方法快速、准确、重复性好,为研究大黄配伍附子增效减毒提供实验基础。

Abstract:Objective To develop an RP-HPLC method for simultaneous determination of chrysophanol, emodin, rhein, aloeemodin and physcion and to investigate the change in chemical constituents in the herb couples. Methods Diamonsil C18 column (4.6 mm×200 mm, 5 μm) was used, and methanol-1.0% acetic acid was used as the mobile phase by gradient elution with the flow rate of 1.0 mL·min^-1 and detected at 430 nm. Results The linear range of chrysophanol, emodin, rhein, aloe-emodin, and physcion was 0. 010 60 ~ 0. 212 0 (r=0. 999 5), 0. 002 720 0.054 40 (r= 0.999 3), 0.002 520 ~ 0.05040 (r: 0.999 6), 0.002 560 ~ 0.051 20 (r= 0.999 7), and 0.002 760 ~ 0. 055 20 mg· mL^-1 (r=0. 999 3), respectively. The average recoveries (n=5) of the 5 anthraquinones were (101.1%±1.8%), (102.8%±2.6%), (96.7%±1.5%), (100.9%±2.1%), and (99. 6%±2. 4%). The unitary content diversity of the ration 1 : 1 in Rheum palmatum L. after combining with Aconitum carrnichaeli Debx. de. at different rations was the greatest via the Minkowski. Conclusion The method is rapid, accurate, credible and repeatable, and can provide bases for the efficiency increase and toxicity reduction in Rheum palrnatum L. compatible with Aconitum carmichaeli Debx. de.
