抗角蛋白自身抗体的免疫组织化学研究

作者对人体液 IgG抗角蛋白自身抗体(AK auto Ab)进行检测,结果显示:IgG AK auto Ab很难通过血脑屏障,极易穿透血管壁进入上皮组织。免疫电镜观察表明,AK auto Ab与角朊细胞的反应主要定位于张力细丝和桥粒,而与其它细胞器无关。家兔硫化钠皮炎皮损及血清内AK auto Ab的动态观察提示,皮损角朊细胞胞浆内AK auto Ab的沉积和血清AK auto Ab一时性降低,主要与角朊细胞的损伤有关。

Immunohistochemical Studies on Antikeratin Autoantibody

On the basis of the detection of IgG AK auto Ab in human body fluids, the positive rate of AK auto Ab ir cerebrospinal fluid is only 6.25%. It suggests that AK aito Ab can hardly penetrate the blood-brain barrier . On the contrary , the positive rates of AK auto Ab in pleura! fluid ,exudate of auris media , and blister fluid of second-degree bum are close to normal serum , which shows that IgG AK auto Ab can easily pass through the capillary walls into epithelial tissues . Using sections of normal skin as an antigen , the AK auto Ab reaction of normal pooled sera was observed by immunogold -silver technique . All of the 4 samples were positive , specifically located in the endoplasma of pickle cells and granularcells . Further observations showed that the site of reaction was mainly in the tonofilament of the endoplasma of the keratinocytes with immunogold staining under TEM . Dynamic observations of AK auto Ab suggest that both the accumulation of AK auto Ab in keratino-cytoplasm and transitorydecrease of IgG AK auto Ab in serum of rabbits with sodium sulfide dermatitis are related prodominently to keratinocytic injury .
