人正常肝,新生儿肝,肝癌及癌旁肝组织内C／EBPmRNA丰度分析

ｃ／ＥＢＰ（ＣＣＡＡＴ／ＥｎｈａｎｃｅｒＢｉｎｄｉｎｇＰｒｏｔｅｉｎ）是一种热稳定蛋白，作为肝细胞核富含的转录调控因子，在促进和维持肝细胞分化状态中担负重要功能，本文通过Ｎｏｒｔｈｅｒｎ杂交检测３例正常肝组织，１例新生儿肝组织，１０例肝癌及８例癌旁肝组织中Ｃ／ＥＢＰｍＲＮＡ丰度，结果显示在正常肝、新生儿肝及癌旁肝组织中均有表达，在多数癌组织中的表达明显减弱。瘤旁肝组织和肝癌组织中的表达相差显著（Ｐ＜０．０５）．以上结果进一步提示Ｃ／ＥＢＰ在维持肝细胞分化状态中起重要作用，并可能和肝癌的发生存在一定关系。

DETERMINATION OF C/EBP mRNA IN HUMAN NORMAL ADULT LIVER, NEW BORN LIVER, HEPATOCARCINOMA AND ADJACENT HEPATIC TISSUE

C/EBP,a heat -stable DNA -binding protein, plays a very improtant role in estblishing and maintaining the differentiated state of liver cells as a transeription factor enriched in liver nuclei. In order to identify its distribution and function further.C/EBP mRNA was detected by using synthesized cDNA probe in 3 cases of human normal adult liver tissue, one cased of new born liver tissue, and 10 cases of hepatocarcinoma tissue (8 cases in connection with adjacent hepatic tissue).The result showed that C/EBP mRNA was detected in normal liver, new born liver and adjacent hepatic tissue.In most cancer tissues ,the positive signal was very weak. C/EBP mRNA abundance was obviously richer in adjacent hepatic tissue than in cancer tissue(P<0.05).This result suggested that C/EBP plays a very important role in establishing and maintaining the differentiated state of liver cells. The result together with other efforts,also suggests that C/EBP may have an anti-proliferative effect on liver neoplastic tissue.
