摘    要: 目的建立大鼠脑脊液中石杉碱甲的高效液相色谱-荧光测定方法。方法采用ODS-C18柱(4.6mm×250mm,10μm),以甲醇-水-三乙醇胺(60:40:0.1)为流动相,流速1mL·min-1,荧光检测器于激发波长310nm,发射波长370nm处检测,同时紫外检测器于波长310nm处检测作为对照。结果石杉碱甲质量浓度在0.5~200μg·L-1内线性关系良好(r=0.9999),最低定量限为0.5μg·L-1,比紫外法降低了10倍。结论该方法准确,可靠,比高效液相-紫外法灵敏度更高。

Abstract:OBJECTIVE To establish a HPLC-Fluorescence method for the determination of the huperzine A in rat cerebrospinal fluid. METHODS The separation was performed on a ODS-C18 column(4.6mm×250mm, 10μm) with the mobile phase consisted of methanol-water-triethanol amine(60:40:0.1). The flow rate was 1mL·min-1. The excitation wavelength was the at 310nm and the emission wavelength was at 370 nm, at the same time UV wavelength was at 310 nm as a comparison. RFSULTS A good linear relationship of huperzine A was obtained over the range of 0.5~200μg·L-1, and the detection limit of HPLC-Fluorescence method was 0.5μg·L-1 which was 10 times lower than that of HPLC-UV method. CONCLUSION HPLC-Fluorescence method is suitable for huperzine A determination, which is more accurate and reliable than HPLC-UV method.
