摘    要: 目的:探讨影响庆大霉素毫微球(GM-NP)主要特性的制备工条件。方法:以聚氰基丙烯酸正丁酯为载体, 用乳化聚合法(一步法和二步法)制备不同配方的GM-NP ,考察制备工艺,表面活性剂种类,投葯量,添加剂和离子强度等对形态,包封率,粒度及分布等的影响。结果:表面活性剂种类,制备工艺对包封率和粒度影响明显,添加剂种类对包封率也有明显影响。离子强度与包封率成正比。投药量与载药量成正比,而对包封率的影响不大。投药量増加,粒径增大,分布展宽。表面活性剂种类影响GM-NP的形态。结论:对影响GM-NP主要特性的制备条件有了较好的了解。

Abstract:OBJECTIVE To investigate the factors affecting the main characteristics of gentamicin nanoparticles (GM-NP) in the preparation procedure. METHODS Several formulations of GM-NP were prepared by emulsion polymerization (one step and two step method) with the polybutylcyanoacrylate. The effects of preparation techniques (PT ), surfactants, additives, the amount of drug (AD) ,and ion strength (IS) on morphology associating ratio (AR), particle size (PS) and so on were studied. RESULTS The AR and PS were significantly affected by surfactants and PT,and AR was also affected by additives. The AR was directly proportional to IS,while the DLCdrug loading) was directly proportional to AD. As AD incresing, PS became bigger and its distribution became wider. The morphology was influenced by surfactants. CONCLUSION A better understanding of PT which may affect the major properties of GM-NP was achieved.
