小鼠实验性肿瘤冷刺激后复温规律及其机制探讨

摘    要:目的 探讨肿瘤组织冷刺激后复温规律及其与组织微血管密度、增殖细胞核抗原表达、局部组织血流量之间的关系。方法 将实验小鼠分为肿瘤组、肿瘤对照组和空白组。在接种后第 1、4、7、10、13、16、19、2 2、2 5天时分别对实验各组进行冷刺激 (将装有 12℃水的塑料袋敷于动物后肢背侧 2 min,立即用红外热图观察局部温度改变 ) ,同时在该时相点上测定局部组织血流量、组织微血管密度以及增殖细胞核抗原表达水平。结果 肿瘤在接种后局部温度均升高 ,在第十天时达到峰值 ,此后逐渐下降 ,冷刺激后 ,在肿瘤早期和中期复温速度明显加快 ,但在肿瘤生长的晚期 ,局部组织复温速度却明显减慢。肿瘤组织在各时相点上的 Gm ax值和Tm值与局部组织血流量、组织微血管密度以及增殖细胞核抗原表达呈正相关关系。结论 肿瘤组织各时相点上复温规律不同 ,其机制可能与局部组织血流量 ,组织微血管密度以及增殖细胞核抗原表达不同有关

Abstract:Objective To explore the rewarming regularity and its mechanism with infrared thermography, microvessel densities, PCNA and blood flow in local tissue. of mice suffered from tumor after cold provoation. Methods Mice were randomly divided into:(1) Tumor group(Tt), (2) Tumor control group(Tc),(3)Control group(Co). The hindlimb muscles of Kunming mice were inocutated with hepatic cancer cells (H 22 ) to establish the tumor model. After the hindlimbs were treated with cold water bag, their temperatures were monitored using the infrared thermography; in addition, local tissue blood flow, microvessel density, and the level of PCNA were examined. Results The maximum rate of rewarming (Gmax) and the time of the Gmax reaching (Tm) showed no significant differences between Tt and Tc group after cold provocation at day 1 post inoculation. Gmax and Tm reached the peak and valley respectively at day 10. At day 25 Gmax in Tt group was significantly lower than that in Tc or Co group. Tm in Tt group was significantly longer than that in Tc or Co group. Gmax in Tt group was positively correlated with microvessel density (r=0.9036, P <0 05), PCNA(r=0 9136, P <0 05) and local tissue blood flow (r=0 8976, P <0 05) respectively. Conclusion the temperature of local tissue was affected by inoculating cancer cells. The resulted differences of rewarming regularity at different time was probably due to changes in microvessel dentity, local blood flow and PCNA.
