胃癌中基质金属蛋白酶及其抑制物表达的临床意义

目的研究基质金属蛋白酶-2（MMP-2）、基质金属蛋白酶-9（MMP-9）和金属蛋白酶组织抑制剂（TIMP-2）在胃癌中的表达及与临床病理特征的关系。方法应用组织芯片技术。免疫组织化学SP法检测45例胃癌，45例癌旁正常黏膜组织中的基质金属蛋白酶及其抑制物的表达。结果胃癌中的MMP-2、MMP-9的阳性表达显著高于癌旁正常黏膜组织（P〈0．05）；MMP-2、MMP-9的阳性表达与胃癌浸润深度、淋巴结转移及分期呈正相关（P〈0．05）；淋巴结转移的胃癌中MMP-2、MMP-9呈高表达，而TIMP-2显著低表达；胃癌中MMP-2与TIMP-2的表达呈负相关（P〈0．05）。结论胃癌中MMP-2、MMP-9表达显著高于癌旁正常黏膜组织，结果提示可作为胃癌预后判断的指标之一。

Study the Expression of MMPs and TIMP Protein in the Colon and Rectal Cancer and Their Clinical Significances

Objective To investigate the expressions of MMP -2,MMP-9 and TIMP- 2 in human gastric carcinoma and their correlation with the clinicopathological characteristics. Methods .The expressions of MMPs and its inhibitors were detected using tissue microarray and immunohistochemistry S - P method, in 45 cases of gastric carcinoma and 45 cases of normal adjacent gastric mucosa. Results In gastric carcinoma, the positive expression rates of MMP - 2 and MMP - 9 proteins were significantly higher than in normal mucosa （ P 〈 0.05 ）. The expressions of MMP - 2 and MMP - 9 proteins were positively correlated with infiltrating depth, lymph node metastasis and clinical stages （ P 〈 0.05 ）. MMP - 2 and MMP- 9 in lymph node metastasis showed high expression, but TIMP- 2 showed low expression; the expression of MMP - 2 and TIMP - 2 were negatively related （ P 〈 0.05 ）. Conclusion The expression of MMP - 2 and MMP - 9 in gastric carcinoma was significantly higher than the normal mucosa, which could be the prognostic marker.
