滋阴清热方对系统性红斑狼疮阴虚内热证患者PBMC基因表达谱的影响

摘    要:目的观察滋阴清热方对系统性红斑狼疮(SLE)阴虚内热证患者PBMC基因表达谱的影响。方法用滋阴清热含药血清孵化阴虚内热型SLE患者的PBMC,提取总RNA,用基因芯片检测孵化后PBMC基因表达谱。结果研究发现滋阴清热方含药血清上调51个基因表达,下调92个基因表达,它们分属于21个基因功能簇,基因编码蛋白分布于细胞的8个部位。含药血清和地塞米松共同上调2个基因表达和下调12个基因表达;但是含药血清还下调12个基因表达和上调9个基因表达,而地塞米松对这21个基因无类似调控作用;同样,地塞米松下调11个基因表达和上调15个基因表达,而含药血清对这26个基因无类似调控作用。结论滋阴清热方对SLE阴虚内热型基因表达谱影响是广泛和复杂的,涉及基因数目较多,基因编码蛋白在细胞中分布广泛,具有上调和下调功能簇内基因表达的作用。

Abstract:Objective To observed the influence of nourishing yin clearing heat formula in gene expression profiling on PBMC of internal heat due to yin deficiency type SLE patients.
