加味参附颗粒对慢性心力衰竭大鼠心肌超微结构的影响

摘    要:
目的建立大鼠心衰模型,观察加味参附颗粒对腹主动脉缩窄法造模成功的慢性心力衰竭大鼠模型的心肌线粒体Na+-K+-ATP酶、Ca址ATP酶活性、血清TNF-α及IL-6的表达和超微结构的影响。方法雄性SD大鼠50只,随机分为正常组(不给药)、模型组(灌胃生理盐水)、曲美他嗪组(灌胃曲美他嗪5.4mg/kg)、加味参附颗粒低剂量组(灌胃加味参附颗粒6.84g/kg)、加味参附颗粒高剂量组(灌胃加味参附颗粒13.68g/kg)各10只,采用腹主动脉缩窄法建立心衰模型。测定心肌线粒体Na+-K+-ATP酶、Ca2+-ATP酶活性、血清TNF-α及IL-6的表达和电镜下观察心肌超微结构形态。结果加味参附颗粒高、低剂量均能够显著提高心衰大鼠心肌线粒体Na+-K+-ATP酶、Ca2+-ATP酶活性。减轻心肌超微结构的损伤。结论加味参附颗粒对心衰大鼠超微结构有保护作用,其机制可能是通过慢性心衰心肌各细胞因子的表达具有重要的抑制效应.并通过调节心肌线粒体Na+-K+-ATP酶、Ca2+-ATP酶活性发挥作用。

Effect of Jiaweishenfu Granule on Ultrastructure of Myocardium in Rats with Chronic Heart Failure

Abstract:
Objective: To establish heart failure model of rats with chronic heart failure by abdominal aorta con- striction and to observe the effect of Jiaweishenfu granule on myocardial mitochondria Na+-K+-ATP enzyme,activ- ity of Ca2+-ATP enzyme,serum TNF-α and IL-6 expression,and ultrastructure. Methods: 50 male SD rats were randomly divided into normal control group,model group,Qumei trimetazidine group,low dose of Jiaweishenfu granule group,and high dose of Jiaweishenfu granule group, 10 rats in each group. Rats with heart failure model were established by abdominal aortic coarctation method. Myocardial mitoehondria Na+-K+-ATP enzyme,Ca2+- ATP enzyme activity,and expressions of serum TNF-α and IL-6 were determined. Ultrastrueture under electron microscopic was observed. Results: low dose of Jiaweishenfu granule group can significantly improve activity of myocardial mitochondria Na+-K+-ATP enzyme and Ca2~-ATP enzyme,and reduce myocardial ultrastructure dam- age. Conclusion: Jiaweishenfu granule has a protective effect on ultrastructure of heart failure rats. It works by inhibiting expression of chronic heart failure cytokines,and by regulating activity of myocardial mitochondria Na+- K+-ATP enzyme and Ca2+-ATP enzyme.
