流式细胞仪检测HLA-B_(27)抗原初探

目的　建立一种快速、准确的HLA B2 7检测技术。方法　利用流式细胞仪 (FCM)检测HLA B2 7抗原 ,并将结果与基因定型法比较 ,74例临床样本分别用FCM法、PCR ASP基因定型法检测HLA B2 7抗原和基因。结果　FCM法有 2 2例HLA B2 7阳性 ,5 2例阴性 ,与PCR ASP法结果比较 ,阳性符合率 88% ,阴性符合率 10 0 % ,总观察一致率 95 .9% ,二种检测方法无显著性差异 (P >0 .0 5 )。结论　可以认为 ,FCM法检测HLA B2 7具有快速灵敏、多参数检测等优点 ,针对不同的样本来源和临床要求 ,与PCR ASP法配合使用 ,能在临床强直性脊椎炎等疾病诊断中发挥重要作用

Detection of HLA-B27 antigens by flow cytometry

Objective The aim of the present study was to construct a rapid and accurate method for HLA B 27 antigens detection by flow cytometry (FCM).Methods Whole blood samples from 74 patients was directly immunoflurescence stained by FITC labeled monoclonal anti HLA B 27 and then analyzed by FCM for HLA B 27 phenotyping.Simultaneously,genomic DNA was extracted for HLA B 27 genotyping by PCR ASP..Results 22 samples were HLA B 27 antigen positive whilst 52 samples were negative using FCM assay,and the results showed no significant difference( P >0.05) compared with the results of PCR ASP genotyping.The consistency between FCM and PCR ASP genotyping was 88% for positive results,100% for negative results,with a total of 95.9%.Conclusion Flow cytometric HLA B 27 typing is rapid and sensitive and can be used together with PCR technique for diagnosis of some diseases such as ankylosing spondylitis.
