雌激素药物对大鼠子宫内膜异位症OPN、MMP-9表达的影响

摘    要:目的探讨雌激素药物(丙氨瑞林、米非司酮、孕三烯酮)对大鼠子宫内膜异位症(EMS)OPN、MMP-9表达的影响。方法将建模成功的36例实验用大鼠随机性地均分为四组,各9例.分别给予丙氨瑞林、米非司酮、孕三烯酮三种雌激素类药物干预,另外使用酵母片进行干预,1个月后将大鼠杀死.将其在位与异位内膜取出,运用RT—PCR法对OPN与MMP-9蛋白及基因表达进行分析、测定。结果三组雌激素药物干预组干预后在位内膜与异位内膜中的OPN与MMP-9的表达均要明显低于干预前,且干预前后差异存在高度统计学意义(P〈0.01);对照组干预后异位内膜中的OPN与MMP-9的表达要强于在位内膜;三组雌激素干预组干预后在位与异位内膜OPN表达无差异统计学意义(P〉0.05),但干预后雌激素干预组异位内膜中MMP-9表达明显强于在位内膜(P〈0.01);对照组干预后异位内膜中OPNmRNA的表达较干预前出现显著性降低(P〈0.01),且在位与异位内膜的表达差异无统计学意义(P〉0.05)。结论雌激素类药物能够降低大鼠在位及异位内膜中OPN与MMP-9的表达能力,且对异位内膜中的OPN表达能力的抑制作用要明显强于在位内膜,对在位内膜中的MMP-9表达能力的抑制性作用不亚于对异位内膜;雌激素类药物可能通过对局部病灶OPN及MMP-9表达能力的抑制作用进行抑制来减小细胞的侵袭及黏附性能。

The effect of estrogen drugs on expression of OPN and MMP-9 in endometriosis rats

Abstract:Objective To investigate the drug (estrogen Alarelin, mifepristone, pregnancy three alkene ketone) on rats with endometriosis EMS) OPN and its effects on MMP-9 expression. Methods The model 36 successful cases of ex- periments using rats at random divided into four groups, 9 cases in each group, were given Alarelin, mifepristone, pregnancy three alkene ketone three kinds of estrogen-like drug intervention, using yeast to intervene, January after rat was killed, its reign and ectopie cndometrium out, use the RT-PCR method to the OPN and MMP-9 protein and gene expression analysis, determination. Results The three groups of estrogen drug intervention group in eutopic and ectopie endometrium of OPN and MMP-9 expression were significantly lower than before intervention and after inter- vention, there was significant difference(P 〈 0.01); the control group after intervention of ectopic endometrium in OPN and the expression of MMP-9 was stronger than the cutopic endometrium; estrogen three group intervention group eu- topic: and ectopic endometrial expression of OPN there is no statistically significant difference (P 〉 0.05), but after the intervention of cctopic endomctrium estrogen intervention group MMP-9 expression was significantly stronger in eu- topic endometrium(P 〈 0.01); the control group after intervention of ectopic endometrium in OPNmRNA expression than those before intervention significantly decreased (P 〈 0.01), and reign and ectopic endometrial expression there is no statistically significant difference (P 〉 0.05). Conclusion Estrogen can reduce rat eutopic and ectopic endometri- um of OPN and MMP-9 expression ability, and the ability of OPN expression in endometriosis inhibition would be more obvious in the eutopic endometrium, in eutopic endometrium of MiVIP-9 expression ability of inhibitory role than on heterotopic endometrium; estrogen may pass for local lesions in OPN and MMP-9 expression inhibition was inhibit- ed to reduce cell invasion and adhesion properties.
