微波提取法在板蓝根木脂素部位提取中的应用

摘    要:目的:优选微波提取板蓝根中木脂素部位的工艺。方法:以总木脂素含量及clemastanin B(CB)单体含量作为筛选指标,分别应用紫外分光光度法、HPLC对两者进行含量测定,采用正交实验法对提取过程中的微波辐照功率、提取时间、料液比、浸泡时间4个因素进行优选。结果:最佳提取条件为辐照功率700 W,加15倍量水提取2次,每次8 min,提取前浸泡15min,在此条件下所得总木脂素和CB的提取率分别达到3.39%,0.019 3%,与回流提取3.5 h的提取率无显著差异。结论:本方法具有快速、高效、方便、节能等优点,值得推广应用。

Abstract:Objective: To optimize the technology for the microwave extraction of lignanoids from Isatidis Radix . Method: With the comprehensive extraction ratio of total lignanoids and clemastanin B (CB for short) as indexes,whose content was separately determined by UV spectrophotometry and HPLC,four main factors including microwave power,extraction time,volume of solvent and soak period were optimized by orthogonal test. Result: The optimal extraction process was that 15 times amount of water was added,soaking for 15 minutes with the microwave power at 700 w for 8 minutes,twice in all.Under that condition,the extraction ratio of total lignanoids and CB was 3.39% and 0.019 3%,which was close to the yield achieved by water decoction for 3.5 hours. Conclusion: Microwave extraction of lignanoids from Isatidis Radix is characterized by rapid,efficient,simple,energy conservation,and should be popularized in the future.
