摘    要:目的:研究大鼠脑缺血不同时间脑能量代谢的变化。方法:复制大鼠永久性局灶性大脑中动脉缺血模型,测定缺血6,24,72,168,336 h脑组织乳酸(Lac)和丙酮酸(Pyru)的含量,Na+,K+-ATPase和Ca2+,Mg2+-ATPase活性。结果:在各个不同时间点,各检测指标与假手术组比较均具有显著性,P<0.05,P<0.01。结论:在缺血336 h内观测脑组织中能量代谢相关指标,可以用于评价药物对脑缺血的保护作用。

Abstract:Objective :To investigate changes in brain energy metabolism for different durations of cerebral ischemia in rats. Method: Permanent cerebral ischemia was induced by permanent middle cerebral artery occlusion(pMCAO) in rats,and ischemic cortex Lac and Pyru content,Na+,K+-ATPase and Ca2+,Mg2+-ATPase activity were measured at 6,24,72,168,336 h after ischemia. Result :At different time points after ischemia,brain energy metabolism in vehicle-treated rats showed significant changes compared with sham-operated group(P<0.05,P<0.01). Conclusion :There were fluctuation changes in ischemic cortex Lac and Pyru content,Na+,K+-ATPase and Ca2+,Mg2+-ATPase activity at different time points after ischemia in rats in vehicle-treated groups.Changes of ischemic cortex Lac and Pyru content,Na+,K+-ATPase and Ca2+,Mg2+-ATPase activity may be used as important indexes for early diagnosis and treatment assessment of cerebral ischemia.
