穴位注射复方丹参液对大鼠跟腱病组织修复的实验研究

摘    要:
目的:通过观察穴位注射复方丹参注射液对跟腱病大鼠步态改善情况,探讨穴位注射治疗跟腱病跟腱组织修复的影响,为跟腱病的临床应用提供理论依据。方法:从60只2月龄雄性SD大鼠中随机选出10只.设为空白对照组(I组),对其余大鼠行跟腱病造模后随机分为阳性对照组(生理盐水对照组,Ⅱ组),复方丹参注射液治疗组(Ⅲ组),每组各25只,造模3周后Ⅱ组每只大鼠同样取阳陵泉、承山、昆仑三穴,注射生理盐水0.1ml/穴.1次/2d;Ⅲ组每只大鼠同样取阳陵泉、承山、昆仑三穴,行复方丹参注射液穴位注射,0.1ml/穴,1次/2d;I组大鼠不予任何干预。于治疗20d后,处死大鼠行步态检测,分别计算步长差比值(TOFF)、足长差比值(PLF)、全趾宽比值(TSF)和中间趾宽差比值(ITF),进行统计学分析,以评价跟腱病功能恢复情况。用HE染色观察组织学结构改变,作跟腱内成纤维细胞,纤维细胞总数计数后行统计分析,以评价跟腱病组织学恢复情况。结果:步态,TOFF,I、Ⅱ组之间差异有统计学意义(P=0.024〈0.05),Ⅲ、I组之间差异无统计学意义(P=0.150〉0.05);TSF,I、Ⅱ组之间差异有统计学意义(P=0.015〈0.05),Ⅲ、I组之间差异无统计学意义(P=0.070〉0.05);ITF,I、Ⅱ组之间差异有统计学意义(P=0.024〈0.05),Ⅲ、I组之间差异无统计学意义(P=0.150〉0.05)。细胞计数,Ⅲ组与I组相比,差异有统计学意义(P=0.035〈0.05);Ⅱ组与I组比较,差异有统计学意义(P=0.007〈0.01)。结论:复方丹参注射液能有效改善跟腱病大鼠步态。根据多项指标统计比较并结合形态学观察.廊用复方丹参注射液穴位沣射能有效缩短治疗时间并取得良好疗效.

Abstract:
Objective: To observe the efficacy of composie Salvia milyiorrhizae acupoint-injection on achilles tendinopathy by investigating suffering rats' gaits and histological changes of their achilles tendon, and to provide information for acupoint-injection therapy of achilles tendinopathy. Methods: 10 male SD rats (2 months old) were drawn out from sixty ones as blank control group (group I, n=10) randomly. The models of rats' achilles tendinopathy were set up by 1% Collagenase Injection divided into the control group (group II, n=25) and point-injection group (group Ⅲ, n=25). Treatments start three weeks after the model was set up. Each rat of group I1 was point-injected (Yanglingquan, Chengshan, Kunlun) with sodium chloride injection (0.1 ml/P) and group Ⅲ miltiorrhizae once two days. Rats' gaits were analyzed by the accounting of TOFF, PLF, TSF and ITF after treatment 20 days and estimated the functional recovery before rats were executed. All the specimens were given HE coloration, and histological examination were performed. The number of the fibroblasts and fibrocytes was counted and statistically analyzed to estimate the histological recovery of this disease. Results: Gait, TOFF, group II compared with group I had statistical difference (P=0.024〈0.05), group Ⅲ compared with group I had no statistical difference (P=0.150〉0.05); TSF, group II compared with group I had statistical difference (P=0.015〈0.05), group Ⅲ compared with group I had no statistical difference (P=0.070〉0.05); ITF, group II compared with group I had statistical difference (P=0.024〈0.05), group Ⅲ compared with group I had no statistical difference (P=0.150〉0.05). Cell counts, the fibroblast counts of group Ⅲ compared with group I had statistical difference (P=0.035〈0.05), group II compared with group I had significantly statistical difference (P=0.007〈0.01). But the fibroblast of group -Ⅲ was slightly lower than that of group I . Conclusion: Salviae mihiorrhizae acupoint-injecton can better the suffering rats' gaits. Composite Salviae miltiorrhizae acupoint-injection can shorten therapy time and acquire favorable curative efficacy.
