摘    要:目的:研究穿心莲内酯的微乳提取方法及其对有效成分含量、生物活性和生物利用度的影响。方法:试验一,分别用微乳液和乙醇溶液从穿心莲中药饮片提取穿心莲内酯,用HPLC法分别测量两种提取物中穿心莲内酯的含量;试验二,选取二甲苯致小鼠耳肿胀模型,分为空白组、微乳组和乙醇组分别计算二甲苯致小鼠耳肿胀度和抑制率;试验三,选取大耳白兔随机分为两组,分别灌胃给微乳提取液和乙醇提取液,测血药浓度。结果:穿心莲的微乳提取液和乙醇提取液中的穿心莲内酯含量差异不显著,但微乳提取液的抗炎活性明显高于乙醇提取液,其穿心莲内酯在体内的生物利用度也明显提高。结论:利用微乳液提取中药不仅可以提高有效成分的提取效率,而且可以显著提高有效成分的生物活性和生物利用度,是一种具有良好应用前景的中药提取技术。

Abstract:Objective :To study the extraction of andrographolide by microemulsion method as well as its influence on the content of the active component,biological activity and bioavailability.Methods:1.Respectively extract andrographolide by way of microemulsion and ethanol solution from the Chinese Herbal Medicine Andrographis drinking pills,further by using the method of HPLC to measure respective content of andrographolide.2.Select the models of xylene-induced swollen ears of mice and then divide them into blank group,microemulsion group and the alcohol group to calculate the xylene-induced degrees of swollen as well as inhibition rate.3.Select big-eared rabbits and divide them randomly into two groups,then respectively gavage them the microemulsion extracts and ethanol extracts,after that,measure the drug concentrations in serum.Results:The content of andrographolide by way of microemulsion-extraction and ethanol-extraction was not significantly different,however,the anti-inflammatory activity by means of microemulsion-extraction was much higher than that of ethanol-extraction,so does bioavailability in vivo.Conclusion:Extracting Chinese herb medicine by use of microemulsion can not only enhance extraction efficiency of the active components but also significantly improve the biological activity and bioavailability of the active component.It is also possible to form an microemulsion-carrier drug system by one step of extraction and this extraction technology is of great prospect.
