摘 要: | ![]() 目的 探讨姜黄素对单侧输尿管梗阻(UUO)大鼠模型的影响及其可能机制.方法 将30只大鼠随机分为3组,每组10只:假手术组、模型组、姜黄素组.模型组和姜黄素组行右侧输尿管接扎术,假手术组只游离不接扎.术后第14天处死各组中的大鼠,股动脉取血,检测血清肌酐、尿素氮;留取梗阻侧肾脏,Maason染色观察肾间质纤维化程度,免疫组织化学方法测定TGF-β1、CTGF的表达情况,RT-PCR技术榆测肾组织TGF-β1 mRNA、CTGF mRNA表达.结果 姜黄素降低了BUN、Scr的含量,同时姜黄素显著减少了大鼠肾间质TGF-β1、CTGF的表达,并有效改善了肾脏的病理学损伤.结论 姜黄素对单侧输尿管梗阻大鼠有较明显的保护作用,这可能与其能减少大鼠肾间质TGF-β1、CTGF的表达有关.

Effect of curcumin on renal interstitial fibrosis after unilateral ureteral obstruction in rats |

Abstract: | ![]() Objective To explore the therapeutic efficacy and the mechanism of Curcnmin in rats with the unilateral ureteral obstruction (UUO). Methods The experiment was successfully conducted from May 2007 to August 2007 in Union Hospital, Tongji Medical College, HUST (China). Thirty SpragueDawley rats were randomly divided into three groups:sham operated group, UUO model group, Curcumintreated group (n = 10 in each group). The rats in the UUO model group and Curcumin-treated group underwent UUO surgery. The rats were killed at 14th day after surgery. Blood sample was collected from each mouse for the measurement BUN and Scr levels. The kidneys of the rats were sliced to make pathological sections, and histological changes were observed by Masson stain. By using immunohistochemistry and RTPCR,the expression of TGF-β1 and CTGF protein and mRNA was detected,respectively. Results In the obstructed kidneys,there were significant renal interstitial fibrosis and interstitial space expansion. Immunohistochemical staining showed that the expression of TGF-β1 and CTGF was significantly increased in the obstructed kidneys. In comparison with the model group, Curcumin could significantly decrease interstitial volume and the expression of TGF-β1 and CTGF, and effectively improve the histological changes of the UUO renal tissues. Conclusion Curcumin has certain protecting effect on UUO rats probably by down-regulating the expression of TGF-131 and CTGF in UUO rats. |
