独活寄生汤对兔膝骨性关节炎关节液白细胞介素-1和肿瘤坏死因子的影响

目的观察独活寄生汤对兔膝骨性关节炎中白细胞介素-1(IL-1)和肿瘤坏死因子(TNF)表达水平的影响,探讨其治疗骨性关节炎的机制。方法将24只兔随机分为2组,即对照组和治疗组,各12只,24个膝关节,采用兔膝关节腔内注入木瓜蛋白酶造成骨关节炎的病理模型,以独活寄生汤浓缩液喂食饲养,于治疗后1、3、5周分别抽取关节液,采用双抗体夹心酶联免疫吸附法(ELISA),分别测定兔膝关节液中IL-1及TNF表达水平。结果治疗后1周,膝关节关节液中即可检测到IL-1及TNF的表达,对照组在3周时达到高峰,5周时略有回落。治疗组数值与对照组比较均见下降,1周时下降差异无统计学意义(P>0.05),3、5周时下降差异有统计学意义(P<0.01,P<0.05)。结论独活寄生汤能降低骨性关节炎中IL-1及TNF的表达水平。

Effect of Duhuojisheng decoction on IL - 1 and TNF from synovial fluid in rabbit with osteoarthritis

Objective To observe the effect of Duhuojisheng decoction on the expression of IL-1 and TNF from synovial fluid in rabbit with osteoarthritis. Methods 24 rabbits were randomly divided into control and treatment groups. There were 12 rabbits (24 articular genus) in every group. Osteoarthritis model was established by injection of caroid in articular genu intracavitary. Rabbits with osteoarthritis were interfered by Duhuojisheng decoction. The expression of IL-1 and TNF from synovial fluid in rabbit with osteoarthritis were measured by double antibody sandwich ELISA 1, 3, 5 weeks after treatment. Results There were expression of IL-1 and TNF from synovial fluid a week after treatment. Following weeks expression of IL-1 and TNF raised in control group. In 3 weeks peak appeared. Expression of IL-1 and TNF raised in treatment group decreased compared with those in control group (P<0.01, P<0.05). Conclusion Duhuojisheng decoction can depress the expression of IL-1 and TNF in osteoarthritis.
