125I放射性粒子植入对兔肝癌微血管密度及血管内皮生长因子的影响

摘    要:目的 探讨125I放射性粒子植入对兔移植性肝癌微血管密度(microvascular density,MVD)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,MVD和VEGF的相关性及其与影像和临床病理的关系,评价125I放射性粒子植入对兔VX2肝移植瘤血管生成及其疗效的影响,为原发性肝癌的放射性粒子植入治疗提供实验数据及理论依据.方法 将36只荷瘤兔随机分成6组:对照组6只、粒子组30只.分别于接种后1周、2周、1月、2月、3月取材.测量肿瘤体积并行HE染色进行病理学检查,免疫组化染色检测肿瘤VEGF、CD31的表达.结果 对照组肿瘤体积较前增大,粒子组肿瘤体积明显缩小.两组间相互比较粒子组与对照组之间均有明显的差异性(P<0.01) 粒子组VEGF、MVD明显低于对照组(P<0.05),植入粒子不同时期VEGF、MVD有明显的差异(P<0.05).结论 125I放射性粒子植入可以明显降低肿瘤VEGF、MVD的表达,可以有效地破坏植入粒子边缘肿瘤组织的微血管,有效地抑制血管形成并减少肿瘤组织的血液供应,从而达到灭活植入粒子边缘肿瘤组织的目的 .

Abstract:Objective To investigate the effect on the expression ofmicrovascular density (MVD) and vascular endothelial growth factor(VEGF) in hepatic implantation tumor after 125I radioactive particles implanted, the correlation between MVD and VEGF, the relationship between them and the image and pathology characters in VX2 hepatic implantation tumor. To evaluate the therapeutic effect and angiogenesis of the rabbit VX2 hepatic implantation tumor. Method 36 rabbits with VX2 hepatic implantation tumor were collected and randomized into six groups, Group A: six rabbits, control group; group B: thirty rabbits, particle group. When lw, 2w, 1m, 2m, 3m after operation, the rabbits were sacrificed and the tumors were taken out. detect the size and the growth of the tumor, and then immunohistochemistry were performed to demonstrate the expression of VEGF and CD31 to find the change and effect of the tumor after 125I radioactive particles implanted. Result The size of the tumor decreased after the treatment in group B. The tumor size in group A enlarged obviously, and there was significant difference between this rate and the rates of the group B (P〈0.01). VEGF protein was detected by immunohistochemistry, the expression intensity in the group A is the highest. Compared with the data of the group B, it had obvious differences (P〈0.01). The right rate of the group B (13%) was lower than that of group A (15.4%), but there was significant difference between them (P〈0.05). Conclusion Rabbit VX2 hepatic implantation tumor is a tumor with sufficient blood supply. 125I radioactive particles implanted can obviously depress the expression of VEGF and lessen angiogenesis of the tumor. And the treatments can inhibit the growth of the tumor implanted on the rabbits.
