Background:

Non-alcoholic fatty liver disease (NAFLD) is the most prevalent liver disorder in western countries and an important cause of liver cirrhosis, as well as liver failure. Up to now, 20‒40% of the individuals suffer from this disorder and its prevalence is estimated around 5‒30% in Asia. The NAFLD is one of the most prevalent causes for increases in liver enzymes and has a close relationship with obesity, dyslipidemia, hypertension, and type II diabetes. However, no definite treatment has been identified for it yet.

Objectives:

The present study aimed to investigate the effect of berberis vulgaris extract in inducing changes in liver enzymes levels.

Patients and Methods:

The present clinical trial was conducted on 80 patients, including 32 males (40%) and 48 females (60%), who were randomly assigned into two groups of case and control. All the patients had ultrasound evidence of lipid accumulation in the liver and increases in liver enzymes. The case group received two capsules (750 mg) containing berberis vulgaris extract every day for 3 months, while the control group was treated with placebo. The weight, liver transaminases levels and lipid profiles of the two groups were assessed before, during, and after the study.

Results:

In the case group, the mean serum levels of alanine transaminase (ALT) and aspartate transaminase (AST) decreased from 49 to 27.48 and 48.22 to 29.8 u/L, respectively, which was statistically significant compared to the control group (P < 0.001, P < 0.001). In the control group, the mean of ALT and AST decreased from 50.4 to 46.8 and 45.7 to 44.9 u/L, respectively. The difference was not statistically significant. In addition, a significant decrease was observed in weight, triglycerides, and cholesterol, while no significant change was found in fasting blood sugar, high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL).

Conclusions:

Considering the significant decrease in the liver enzymes, triglycerides and cholesterol after using berberis vulgaris extract, further studies with larger sample sizes will identify the accurate dose as well as duration of consumption for this extract, to recommend in the treatment of patients with NAFLD.  相似文献

The aim of this study was to analyse the relationship between insulin–glucose metabolism, nocturnal blood pressure dipping and nonalcoholic fatty liver disease (NAFLD) in obese adolescents without diabetes. One hundred one consecutive children, with biopsy-proven NAFLD, were included in this study. Blood samples were drawn for the analyses of liver function tests, insulin–glucose metabolism and lipid profile appraisal. An ambulatory blood pressure measurement (ABPM) was performed. Seventy-six children (75.3 %) were systolic nondippers, and 23 of them were diastolic nondippers (30.3 %). No differences were found in the anthropometric parameters between the two groups. When compared to the systolic dippers, the systolic nondippers had higher medians of mean nocturnal blood pressure, glucose at 0, 60 and 120 min in the oral glucose tolerance test (OGTT), OGTT insulin at all time points and insulin-resistance values. No correlation of histopathological features with dipping/nondipping statuses was found. Conclusions: We found an association between a nocturnal blood pressure fall and measures of insulin levels, independent of obesity, or daytime blood pressure levels, among the obese patients with NAFLD. Although no association between nondipping profiles and NAFLD was observed in our study, further studies with a longer term follow-up are needed, to better elucidate the complex link between these particular entities.  相似文献

The aim of this study was to investigate the magnitude of mandibular incisor retraction to be expected during full fixed appliance mechanotherapy in Class II Division 1 malocclusions treated with premolar extractions when mandibular incisor retraction was not part of the treatment plan. The material consisted of pretreatment and posttreatment cephalometric films and dental casts of 70 Class II Division 1 malocclusion patients (33 males and 37 females) treated with the preadjusted edgewise appliances. The mandibular first premolars were extracted in 31 subjects, and the mandibular second premolars were extracted in 39. The pretreatment factors that suggested a basis for the extraction choice in the mandibular arch in these patients were found to include the ratio between posterior and anterior facial heights, the maxillary-mandibular plane angle, and the residual space in the mandibular arch. The mandibular incisors were retracted in only 65% of the patients. On average, the mandibular incisors were retracted by 1.02 mm relative to the nasion-pogonion line; there was no significant difference between the 2 extraction groups. Multiple regression analysis showed that the change in the mandibular incisor position is significantly correlated with the residual space in the mandibular arch, the initial distance between the mandibular incisor and the nasion-pogonion line, maxillary-mandibular angle, and the duration of crown labial torque in the mandibular incisors. In combination, these 4 factors could explain 61% of the change in the mandibular incisor position.  相似文献

The purpose of this study was to investigate the effects of oral administration of misoprostol, a prostaglandin E1 analog, on orthodontic tooth movement and root resorption in rats. Sixty-four male Sprague-Dawley rats that initially weighed 250 +/- 20 g were used in this study. The animals were randomly assigned to 1 of 6 experimental and 2 control (nonappliance and appliance) groups. The experimental groups received 2.5, 5.0, 10.0, 25.0, 50.0, and 100.0 microg/kg misoprostol by gastric lavage every 24 hours for 2 weeks. A fixed orthodontic appliance consisting of a nickel-titanium closed-coil spring, 5.0 mm long was ligated between the maxillary right incisor and the maxillary right first molar. The initial activating force was 60 g. For analysis of root resorption, 99 maxillary right and left first molars from 61 animals were chosen. Serial histologic sections of the mesial root of the maxillary first molars were made, and histologic analysis of root resorption on the mesial and distal surfaces was performed. The results showed that oral misoprostol did increase the amount of orthodontic tooth movement in all the experimental groups compared with the appliance control group. This increase was statistically significant in doses of 10.0, 25.0, 50.0, and 100.0 microg/kg (P <.001). However, there were no statistically significant differences among these 4 different doses. There were no statistically significant differences in the amount of root resorption among the groups. However, a trend toward more root resorption was registered. On the basis of these findings, oral misoprostol can be used to enhance orthodontic tooth movement with minimal root resorption.  相似文献

Colorectal cancer (CRC) is the second most common cancer in women and the third most common in men worldwide. Impaired cell cycle regulation leads to many cancers and is also approved in CRC. Therefore, cell cycle regulation is a critical therapeutic target for CRC. Furthermore, miRNAs have been discovered as regulators in a variety of cancer-related pathways. This study is designed to investigate how miRNAs and mRNAs interact to regulate the cell cycle in CRC patients. Utilizing the Kyoto Encyclopedia of Genes and Genomes (KEGG), Gene Expression Omnibus (GEO), and Therapeutic Target Database (TTD), cell cycle-associated genes were identified and evaluated. Seven of the 22 differentially expressed genes (DEGs) implicated in the cell cycle in three GSEs (GSE24514, GSE10950, and GSE74604) were identified as potential therapeutic targets. Then, using PyRx software, we performed docking proteins with selected drugs. The results demonstrated that these drugs are appropriate molecules for targeting cell cycle DEGs. Tarbase, miRTarbase, miRDIP, and miRCancer databases were used to find miRNAs that target the indicated genes. The ability of these six miRNAs to impact the cell cycle in colorectal cancer may be concluded. These miRNAs were found to be downregulated in SW480 cells when compared to the normal tissue. Our data imply that a precise selection of bioinformatics tools can facilitate the identification of miRNAs that impact mRNA translation at different stages of the cell cycle. The candidates can be investigated more as targets for cell cycle arrest in cancers.  相似文献

Three strains of nontypeable Haemophilus influenzae namely NTHi-I, NTHi-II and NTHi-III were isolated from the sputum of patients with bronchitis and identified by biochemical, serological and electron microscopy. The polypeptide patterns of isolates were compared and found to have similar sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) polypeptide patterns, although some of the bands were specific in some strains. A similar comparison was made on extracted outer membrane proteins (OMPs) on the above mentioned strains, using Triton X-100 and sodium dodecyle sulphate (SDS). It was found that the polypeptides with molecular weights of 70, 42, 33 and 27 KDa were identified as P1, P2, P4 and P5 respectively. The protein estimation of crude OMPs from the three strains were calculated, and OPM-I prepared from NTHi-I showed the highest amount of protein and was chosen for its immunogenicity in a rat respiratory model. The efficacy of immunization with OMP was determined by enhancement of pulmonary clearance of live bacteria in the rat lung. A significant protective immune response induced by OMP was observed by enhanced respiratory clearance of nontypeable H. influenzae following mucosal immunization.  相似文献
