While beta2‐adrenoceptor stimulation has been shown to increase lean mass and to alter metabolic properties of skeletal muscle, adaptations in muscle oxidative enzymes and maximal oxygen uptake (O2max) in response to beta2‐adrenergic agonist treatment are inadequately explored in humans, particularly in association with resistance training. Herein, we investigated beta2‐adrenergic‐induced changes in O2max, leg and arm composition, and muscle content of oxidative enzymes in response to treatment with the selective beta2‐adrenergic agonist terbutaline with and without concurrent resistance training in young men. Forty‐six subjects were randomized to 4 weeks of lifestyle maintenance (n = 23) or resistance training (n = 23). Within the lifestyle maintenance and resistance training group, subjects received daily terbutaline (8 × 0.5 mg) (n = 13) or placebo (n = 10) treatment. No apparent treatment by training interactions was observed during the study period. Terbutaline increased leg and arm lean mass with the intervention, whereas no treatment differences were observed in absolute O2max and incremental peak power output (iPPO). Treatment main effects were observed for O2‐reserve (P < .05), O2max relative to body mass (P < .05), O2max relative to leg lean mass (P < .01), and iPPO relative to leg lean mass, in which terbutaline had a negative effect compared with placebo. Furthermore, content of electron transport chain complex I‐V decreased by 11% (P < .05) for terbutaline compared with placebo. Accordingly, chronic treatment with the selective beta2‐adrenergic agonist terbutaline may negatively affect O2max and iPPO in relative terms, but not in absolute. 相似文献

Human and equine rabies immunoglobulins are currently available for passive immunization against rabies. However, these are hampered by the limited supply and some drawbacks. Advances in antibody engineering have led to overcome issues of clinical applications and to improve the protective efficacy. In the present study, we report the generation of a trivalent single-chain Fv (scFv50AD1-Fd), that recognizes the rabies virus glycoprotein, genetically fused to the trimerization domain of the bacteriophage T4 fibritin, termed ‘foldon’ (Fd). scFv50AD1-Fd was expressed as soluble recombinant protein in bacterial periplasmic space and purified through affinity chromatography. The molecular integrity and stability were analyzed by polyacrylamide gradient-gel electrophoresis, size-exclusion chromatography and incubation in human sera. The antigen-binding properties of the trimeric scFv were analyzed by direct and competitive-ELISA. Its apparent affinity constant was estimated at 1.4 ± 0.25 × 109 M−1 and was 75-fold higher than its monovalent scFv (1.9 ± 0.68 × 107 M−1). The scFv50AD1-Fd neutralized rabies virus in a standard in vitro and in vivo neutralization assay. We showed a high neutralization activity up to 75-fold compared with monovalent format and the WHO standard serum. The gain in avidity resulting from multivalency along with an improved biological activity makes the trivalent scFv50AD1-Fd construct an important reagent for rabies protection. The antibody engineering approach presented here may serve as a strategy for designing a new generation of anti-rabies for passive immunotherapy. 相似文献

West syndrome is the most frequent cause of epilepsy in Down syndrome. West syndrome is often associated with poor long-term prognosis in most of children. We report a girl with West syndrome associated with Down syndrome which occurred at 8 months of age for repetitive flexor spasms and electroencephalography (EEG) showed hypsarrhythmia. She had Down syndrome facies, microcephaly, psychomotor development delay and axial hypotonia. Computed tomography of the brain was normal. Her karyotype was 47, XX, +21. Phenobarbital therapy was immediately effective with good clinical control of seizures, while the EEG monitored after one month was unchanged. At 2 years of age, the patient had hypertonic status epilepticus following a lung infection. The EEG showed a persistence of hypsarrhythmia. Sodium valproate and hydrocortisone therapy was effective with good seizure control but her psychomotor development was severely impaired. After a follow-up of 7 years, the patient presents growth retardation, microcephaly, severe psychomotor development delay, generalized hypotonia and tetraparesis. Knowledge of West syndrome in Down syndrome allows the early detection and prompt management of this neurological complication in order to optimize psychomotor development and improve the quality of life of these children. 相似文献

Acrylic resins are widely used in the fabrication of denture bases and have been shown to be cytotoxic as a result of substances that leach from the resin. The primary eluate is residual monomer. Numerous reports suggest that residual monomer may be responsible for mucosal irritation and sensitization of tissues. This information is important, not only to assess the biologic effects of such materials, but also to enable a comparison among the different polymerization methods, thus assisting the clinician in selecting a material with minimal cytotoxicity. Although thoracic duct injury is an uncommon complication of neck dissection, it can have a significant physiological and immunological impact on the patient's recovery as well as having more serious effects. We report a novel technique of using cyanoacrylate tissue glue for managing thoracic duct damage when this is noted at the time of neck dissection surgery.We have used this technique in four patients with no post-operative chyle leak or added complications. 相似文献

Objectives
To investigate the effect of cementation, cement type and vent-holes on the marginal and internal fit of Zirconia (Zr) Copings.Materials and methods
Extracted premolars (N?=?100) were mounted in resin and prepared for Zr crowns. Samples were randomly divided into 5 groups (n?=?20); A: No Cementation; B: Cementation with Glass-Ionomer (GI); C: GI?+?Vent-Holes; D: Cementation with Resin Cement (RC); E: RC?+?Vent-Holes. The copings were fabricated using semi sintered Zr Blocks with a standardized cement space of 20?µm. Groups C & E were designed to have 0.5?mm of Vent-Holes on the Buccal and Lingual Cusp Tips. The copings were tried, cemented, thermocycled, re-embedded in resin and cross sectioned into two halves. The copings were examined with 3D-Digital Microscope (HIROX, KH-7700, Tokyo, Japan) at 50-200X. The gaps were recorded at 9 predetermined points.Results
The non-cemented groups showed statistically better fit compared to the cemented groups P?<?0.05 except vs Group E (RC plus vent holes). No significance was found between the cemented groups P?>?0.05. Copings with vent-holes showed statistically better fit than copings without holes P?<?0.05.Conclusion
Marginal and Internal gap values designed in the software programs differed than the actual values measured for the non-cemented copings. Cementation process also influenced the fit of the Zr copings, the resin cement being the more accurate. The addition of vent-holes on the occlusal surface improved the fit. 相似文献10.

This review summarises all head and neck oncology and related papers published in the British Journal of Oral and Maxillofacial Surgery (BJOMS) during the period 2009-2010. A similar review, covering the years 2007-2008 was published in the Journal in 2009 and was well received by readers, as evidenced by the high number of downloads from the BJOMS website. In this article, we include all head and neck oncology-related articles published in the print version of the journal during 2009/10 and concentrate on summarising the full length article papers. Compared to the previous 2-year period, there has been a 25% increase in number of articles published in this sub-specialty. 相似文献
