BACKGROUND:In recent years, the development of stem cell culture and isolation technologies provides new therapeutic choices for fracture healing. OBJECTIVE:To investigate the effect of exogenous bone marrow mesenchymal stem cells on bone fracture healing in traumatic fracture rats and on the migration ability of endogenous bone marrow mesenchymal stem cells. METHODS:Femoral fracture models were made in 48 Wistar rats and then randomized into experimental group and control group (n=24/group). Bone marrow mesenchymal stem cells from another healthy rats were isolated using adherent method and then injected into the rats via the tail vein in the experimental group. Rats in the control group were given the same volume of normal saline. At 2, 3, 4, 8, 12 weeks after injection, we extracted bone marrow mesenchymal stem cells from the femur of rats in the two groups. RT-qPCR was used to detect expression levels of type I collagen and CD44. Transwell method was used to detect cell migration ability. Immunohistochemitry method was employed to detect expression of nerve growth factors in the callus. RESULTS AND CONCLUSION:mRNA levels of type I collagen and CD44 in rat bone marrow mesenchymal stem cells were significantly higher in the experimental group than the control group at 2, 3 and 4 weeks after injection (P < 0.05). Compared with the control group, the higher migration ability of bone marrow mesenchymal stem cells was found in the experimental group at 2 and 3 weeks after injection (P < 0.05) as well as the higher expression of nerve growth factor in the callus in the experimental group at 3, 4, 8, 12 weeks after injection. All these findings suggest that exogenous bone marrow mesenchymal stem cells can improve the migration ability of endogenous bone marrow mesenchymal stem cells and the expression of nerve growth factor in the callus in rats with femoral fracture, thereby promoting fracture healing in rats.  相似文献

BACKGROUND: During internal fixation for complex long bone fractures, it is difficult to position the internal fixator completely to the contours of the bone surface, often resulting in unstable fixation and affecting the quality of the repair. Digital three-dimensional (3D) models were therefore established based on computed tomography (CT) scans that closely reflected the site and degree of fractures, thereby guiding precise positioning of the internal fixator. From a safety point of view, the digital 3D-derived model was created to ensure that the internal fixator could achieve better repairs of complex long bone fractures.  相似文献

目的探讨改良Watson-Jones入路在髋关节后脱位合并PipkinI、II型股骨头骨折微创治疗中的应用。方法 7例髋关节后脱位合并PipkinI、II型股骨头骨折患者,PipkinI型1例、II型6例,全部采用改良Watson-Jones入路切开复位钛质空心螺钉内固定治疗。结果按照Harris评分:90-96分,平均92.5分。结论改良Watson-Jones入路是治疗髋关节后脱位合并PipkinI、II型股骨头骨折的理想手术入路。  相似文献

BACKGROUND: Osteogenesis is a common problem after surgery for femoral neck fracture in elderly patients. Internal fixation for the treatment of femoral neck fracture should be performed to optimize bone remodeling and strengthen fractured bone trabeculae, with the aim of achieving strong fixation from the perspective of biomechanics. Percutaneous internal fixation with cannulated compression screws has become a preferred treatment method of osteoporotic femoral neck fracture in elderly patients, but the insufficient holding power of the screws used in the femoral neck does not lead to strong fixation. An alternative, joint prosthesis, is recommended for the repair of femoral neck fracture in elderly patients. However, its long-term therapeutic effects in the treatment of osteoporotic fracture of proximal femoral neck remain poorly understood in patients with avascular necrosis of the femoral head.  相似文献

BACKGROUND: Matrix metalloproteinases (MMPs) have been shown to break the balance between the production and degradation of extracellular matrix by degrading collagen, gelatin, elastin and other macromolecules, which is the leading cause of bone and cartilage damage in rheumatic arthritis. OBJECTIVE: To determine the expression levels of MMP-2 and MM-9 in the rabbit knee osteoarthritis cartilage, and to analyze whether they are related to the early MRI performance. METHODS: Fifty healthy Japanese white rabbits free from osteoarthritis were enrolled and randomly divided into control group (n=10) and model group (n=40). The anterior and posterior cruciate ligament amputation was adopted to establish the model of osteoarthritis. Subsequently, MRI examination and the expression levels of MMP-2 and MMP-9 were detected at 1, 3, 5 and 7 weeks (n=10 at each time point), respectively. RESULTS AND CONCLUSION: MRI examination showed that more damaged cartilage appeared with time in the rabbit model of osteoarthritis, and significantly different from MRI classification (P < 0.05). The expression levels of MMP-2 and MMP-9 mRNA in the model group were always significantly higher than those in the control group (P < 0.05); MMP-2 level reached the highest at 3 weeks after modeling (P < 0.01), and then presented a slight decline; MMP-9 level peaked at 5 weeks after modeling (P < 0.01) and slightly decreased at 7 weeks. MRI classification of cartage injury and expressions of MMP-2 (R2=0.119, P=0.119) and MMP-9 (R2= 0.466, P=0.466) were shown to have the correlation.
