Background
N-Acetylglucosaminyltransferase V (GnT-V), an enzyme that catalyzes the β1-6 branching of N-acetylglucosamine on asparagine-linked oligosaccharides of cellular proteins, enhances the malignant behaviors of carcinoma cells in experimental models. The aim of this study was to determine clinical significance of GnT-V expression in human pT2 gallbladder carcinoma with simple in vitro experiments.Methods
Ninety patients with pT2 gallbladder carcinoma were included for this study. The in vitro and in vivo biological effects of GnT-V were investigated using gallbladder carcinoma cells with variable GnT-V expression levels induced by a small interfering RNA.Results
Of the 90 cases, 57 showed positive staining and the remaining 33 demonstrated negative staining, the subcellular localization in the 57 cases was classified into the granular-type in 31 cases and the diffuse-type in 26 cases. In 76 cases with curative resection, postsurgical survival was significantly poorer in those showing positive staining than in those showing negative staining (P = 0.028). In all of the 76 cases, postsurgical recurrence was significantly more frequent in those showing diffuse-type localization than in those showing negative staining. Experimental analyses demonstrated that the down-regulation of GnT-V expression in gallbladder carcinoma cells induced suppression of cell growth in vitro. The expression levels of GnT-V in the cells were highly correlated with the rapid in vivo growth coupled with the enhanced angiogenesis, and the tendency to form liver metastasis.Conclusions
GnT-V expression in the subserosal layer of pT2 gallbladder carcinoma is correlated with the aggressiveness of the disease. 相似文献4.

A 23-year-old woman was admitted to our hospital with jaundice and hepatic coma. She had taken a weight-loss supplement for one month before admission. Her clinical and laboratory findings were consistent with fulminant hepatic failure and fulfilled the criteria of autoimmune hepatitis. Despite corticosteroid pulse therapy and plasma exchange, her symptoms and laboratory findings deteriorated. Her condition improved after she received a living donor-liver transplant from her sister. Autoimmune hepatitis usually follows a chronic course, but it should be considered a type of fulminant hepatic failure and treated promptly. 相似文献

This study investigates the applicability of the novel antioxidant protein, peroxiredoxin (Prx) I as a marker for tumor status in oral squamous cell carcinoma (SCC). Samples from 53 patients with SCC in the oral cavity were examined by immunohistochemistry. Correlations between the expression level of Prx I and proliferating cell nuclear antigen (PCNA), the clinical features of tumors, and their histopathological classifications were statistically analyzed. Cases exhibiting low Prx I expression level included significantly more with larger tumor mass cases (T-category, P=0.004), positive lymph node metastasis (N-category, P=0.015), advanced stage (P=0.002), and poorly differentiated cells (P=0.020). Shortening the diagnostic delay from the onset of symptoms to the final diagnosis leads to early cancer detection and a reduced incidence of advanced cases. To analyze factors contributing to delays in the diagnosis of oral cancer, information was collected from the medical charts of 152 consecutive patients with oral squamous cell carcinoma, and factors associated with diagnostic delay were examined retrospectively. No characteristic was significantly associated with delay caused by patients. Referral by a non-initial professional, initial visit to a dentist, T1 cancer, and the presence of an ulcerative lesion were significantly associated with delay caused by the initial professionals. Patients with N0 were significantly associated with diagnostic delay caused by the final professional. These results re-emphasize the important role of the initial professional, particularly the dentist, and the diagnostic difficulty posed by ulcerative lesions and small-sized or early-stage oral cancer. 相似文献

This study was carried out to evaluate the clinical characteristics of autofluorescence in oral squamous cell carcinoma (SCC) and analyze the fluorescent substances using high-performance liquid chromatography (HPLC). Fifty of 55 oral SCCs (91%) emitted orange or red fluorescence, which was recorded by fluorescence photography. The intensity of the fluorescence significantly correlated with the T and N categories of the cancers, but did not show statistical difference for the types of clinical appearance and primary sites. Protoporphyrin and coproporphyrin were identified as the fluorescent substance in the SCC samples, and the elution patterns on HPLC revealed some porphyrin compounds as specific to oral cancer. These results suggest that the autofluorescence in oral SCC correlates with the progression of lesions, and that fluorescent substances such as protoporphyrin are produced in association with the cancerous tissue. 相似文献

We evaluated the quality of dynamic magnetic resonance (MR) imaging of the temporomandibular joint (TMJ) in 20 normal volunteers with 40 TMJs. To confirm TMJ, we obtained static proton density weighted images (PDWI) before performing dynamic MR imaging with true-fast imaging in a steady-state precession (true-FISP) sequence. Four sequences of the first 10 volunteers were examined to determine the optimal sequence. The 4 sequences included the integrated parallel acquisition technique (iPAT) and/or fat saturation technique. The optimal sequence was then determined and performed in all 20 volunteers. The quality of imaging was evaluated, especially with respect to the conspicuity of the articular disk, mandibular condyle, articular eminence and lateral pterygoid muscle. One of 3 confidence levels was assigned for this evaluation. Neither iPAT nor fat saturation obtained the best quality imaging. Detection rates in the 20 volunteers were 83% for the articular disk, 95% for the mandibular condyle, 96% for the articular eminence and 7.5% for the lateral pterygoid muscle. We recommend dynamic MR imaging of the TMJ with the true-FISP sequence using neither iPAT nor fat saturation.
