视网膜母细胞瘤p53、c-myc及增殖细胞核抗原表达的研究   总被引:9,自引:0,他引:9  
目的 探讨P53、c-myc及增殖细胞核抗原在视网膜母细胞瘤中的表达及其与Rb分化和蔼和视神经浸润的关系。方法 应用抗人P53、c-myc及PCNA的单克隆抗体,采用锭霉卵白素-生物素免疫组化法,对31例Rb常规石蜡标本行p53、c-myc及PCNA表达的测定。结果 p53、c-myc及PCNA在Rb中的表达阳性率分别为51.6%、454.2%及67.7%,且均与Rb的分化程度有关。PCNA的表达  相似文献

一氧化氮与青光眼患者眼压升高关系的研究   总被引:7,自引:0,他引:7  
目的探讨一氧化氮(nitricoxide,NO)在青光眼患者眼压升高中的作用.方法对急性闭角型青光眼患者16例,老年性白内障患者29例,正常志愿者30例,应用高效液相色谱分析仪检测其血浆和房水中NO的含量,并以NO2和NO3之和表示.结果正常对照组血浆中NO的平均含量为(2969.37±862.85)ng/L;青光眼组血浆中NO的平均含量为(1390.78±302.95)ng/L,较对照组明显降低(t=2.853,P<0.01);白内障组血浆中NO的平均含量为(4234.89±1495.23)ng/L,较对照组明显升高.对照组房水中NO平均含量为(1405.83±602.70)ng/L;青光眼组房水中NO的平均含量为(2052.03±694.74)ng/L,与对照组比较差异无显著性(t=1.862,P>0.05);白内障组房水中NO的平均含量为(2454.67±655.13)ng/L,较对照组明显升高.房水/血浆NO含量比值对照组为47.3%,白内障组为58.0%,二者的比值差异无显著性;而青光眼组为144.0%.结论血浆、房水中NO含量与急性闭角型青光眼眼压升高有关.  相似文献

Prolactin and prolactin receptors in the lacrimal gland.   总被引:1,自引:0,他引:1  
Light and electron microscopic immunocytochemistry, in situ hybridization and Dot Blot analysis revealed intracellular localization of prolactin-like molecules and prolactin mRNA in epithelial cells of the lacrimal glands of rabbits. There was also positive immunostaining for prolactin receptors on acinar cells and some interstitial cells. On Western blots of homogenates of whole lacrimal gland, isolated lacrimal acinar cells, isolated lacrimal interstitial cells and peripheral blood lymphocytes, prolactin antibody consistently labeled protein bands migrating at approximately 36 and 50 kD. These data confirm that lacrimal gland acinar cells produce endogenous prolactin-like molecules, but also express prolactin receptors. Since prolactin immunoreactivity has been detected in tear fluid and we found no accumulations of immunogold label in endocytic or transport vesicles, we hypothesize that the prolactin-like molecules in tear fluid originate primarily from synthesis within the acinar cells. We hypothesize further that prolactin from pituitary and other non-acinar cell origin has a modulating influence on acinar cell activity as well as immune function in the lacrimal gland, and that some of the prolactin-like molecules produced by the acinar cells contribute to these functions by autocrine/paracrine mechanisms.  相似文献

We investigated the effects of four-muscle tenotomy on saccadic characteristics in infantile nystagmus syndrome (INS) and acquired pendular nystagmus (APN). Eye movements of 10 subjects with INS and one with APN were recorded using infrared reflection, magnetic search coil, or high-speed digital video. The expanded nystagmus acuity function (NAFX) quantified tenotomy-induced foveation changes in the INS. Saccadic characteristics and peak-to-peak nystagmus amplitudes were measured. Novel statistical tests were performed on the saccadic data. Six out of the 10 INS subjects showed no changes in saccadic duration, peak velocity, acceleration, or trajectory. In the other four, the differences were less than in peak-to-peak amplitudes (from 14.6% to 39.5%) and NAFX (from 22.2% to 162.4%). The APN subject also showed no changes despite a 50% decrease in peak-to-peak amplitude and a 34% increase in NAFX. The "small-signal" changes (peak-to-peak nystagmus amplitude and NAFX) were found to far exceed any "large-signal" changes (saccadic). Tenotomy successfully reduced INS and APN, enabling higher visual acuity without adversely affecting saccadic characteristics. These findings support the peripheral, small-signal gain reduction (via proprioceptive tension control) hypothesis. Current linear plant models, limited to normal steady-state muscle tension levels, cannot explain the effects of the tenotomy.  相似文献

PURPOSE: To determine the effect of alpha-crystallin binding on lens membrane lipid characteristics and the stability of Ca2+ -ATPase activity when challenged with H2O2 or elevated temperatures. METHODS: Alpha-Crystallin binding to muscle sarcoplasmic reticulum membranes was quantified using a centrifugation protocol. Alpha-Crystallin binding to lens epithelial lipids was measured by a fluorescence energy transfer technique. Lipid phase transition temperature and lipid order was measured using fluorescence spectroscopy. Ca2+ -ATPase activity was measured using classical biochemical assays. RESULTS: The main phase transition temperatures of multilamellar vesicles composed of sphingomyelin or lipids extracted from bovine lens were 40 degrees C and 20 degrees C, respectively. In the presence of saturating amounts of alpha-crystallin, the phase transition temperature and lipid order of both sphingomyelin and lens lipid membranes remained almost the same as that without alpha-crystallin. The interaction of alpha-crystallin and lipid is likely to be restricted to the membrane surface. The binding of alpha-crystallin did not influence the oxidative or thermal inactivation of the Ca2+ -ATPase pump. CONCLUSION: Alpha-Crystallin-lens membrane binding does not protect the Ca2+ -ATPase pump from thermal derangement or oxidation by H2O2.  相似文献
