腺嘌呤致大鼠雄性不育的实验研究   总被引:14,自引:0,他引:14  
目的:探讨腺嘌呤致大鼠雄性不育的可能机制。方法:用含0.5%的腺嘌呤饲料饲喂大鼠30d,在实验第30天活杀取睾丸组织,常规连续病理切片。观察睾丸组织的病理改变情况;免疫组化检测睾丸曲细精管TGF-β1蛋白表达水平,脱氧核苷酸末端转移酶缺口末端标记法(TUNEL)检测生精细胞凋亡。结果:实验组大部分曲细精管退行性变性,各级精细胞变性,数量减少;睾丸间质细胞和Sertoli细胞均可见TGF-β1蛋白表达,而对照组只有睾丸间质细胞可见TGF-β1蛋白表达,且实验组TGF-β1蛋白表达强度较对照组为高(P<0.01);睾丸生精细胞凋亡细胞百分率较对照组明显增多(P<0.01)。结论:腺嘌呤诱导的大鼠雄性不育可能与TGF-β1的抑制生精作用及促进生精细胞凋亡有关。  相似文献

目的观察威草胶囊对尿酸性肾病大鼠肾组织中结缔组织生长因子(connective tissue growth fac-tor,CTGF)和肝细胞生长因子(hepatocyte growth factor,HGF)表达的影响,探讨其对尿酸性肾病的治疗作用及其机制。方法将50只SD雄性大鼠随机分为正常对照组、模型组、别嘌呤醇组、威草胶囊小剂量组和威草胶囊大剂量组5组。用酵母和腺嘌呤合用进行模型复制,模型复制成功后分别给予9.0 g/L氯化钠注射液、别嘌呤醇和威草胶囊灌胃。治疗前及治疗4周末,分别检测各组大鼠血中尿素氮(blood urea nitro-gen,BUN)、肌酐(creatinine,Cr)、尿酸(uric acid,UA)水平,采用免疫组织化学技术检测大鼠肾组织CTGF和HGF的表达情况。采用苏木精-伊红染色观察各组大鼠肾组织的病理变化。结果与正常对照组比较,模型组血UA、BUN、Cr均明显升高(P〈0.01),模型组肾组织中CTGF表达水平明显升高,HGF表达水平明显降低;与模型组比较,威草胶囊大、小剂量组和别嘌呤醇组大鼠血UA、BUN、Cr水平及肾组织中CTGF的表达水平明显降低(P〈0.01),威草胶囊大、小剂量组和别嘌呤醇组肾组织中HGF的表达水平明显升高(P〈0.01);威草胶囊大、小剂量组和别嘌呤醇组之间上述指标比较,差异均无显著性(P〉0.05)。病理检测显示,大、小剂量威草胶囊和别嘌呤醇可减轻肾间质尿酸盐结晶沉积和炎性细胞浸润,以及肾小管纤维化程度。结论威草胶囊能够改善尿酸性肾病大鼠模型的肾功能,其机制可能与其降低血尿酸,减少尿酸盐结晶在肾小管的沉积,以及减少CTGF和增加HGF在尿酸性肾病模型大鼠肾组织中的表达,阻止肾间质纤维化的发展等有关。  相似文献

 目的:观察全反式维甲酸和低于临床用药浓度不同剂量的化疗药物吉西他滨、顺铂等及两者联合处理A549人肺腺癌细胞株,以了解不同药物、不同剂量及不同作用时间对A549细胞增殖、细胞周期的影响。方法:用MTT法测定不同药物组相对于对照组的A549细胞增殖抑制率,并用流式细胞学方法检测各药物组及对照组的细胞周期分布。结果:维甲酸联合应用低浓度的顺铂和吉西他滨可显著抑制A549细胞的增殖,随着化疗药物的浓度的增加,抑制率也一并增加,并在达到同样增殖抑制率的效果下降低顺铂和吉西他滨的药物浓度。结论:维甲酸与低浓度化疗药物联合应用有希望显著减少药物的剂量,降低化疗药物的毒性,增强疗效。  相似文献

目的研究肠致病性大肠杆菌(EPEC)毒力因子EspF转染人肠上皮细胞(HIC)株时,血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和IL-8水平的动态变化及与HIC凋亡的关系。方法以Annexin V FITC/PI双染流式细胞仪检测HIC的凋亡率,并用酶联免疫吸附测定法检测转染0、6、12、24、48 h细胞培养上清液中TNF-α、IL-6和IL-8浓度的变化。结果 EspF转染HIC后TNF-α、IL-6和IL-8活性均显著增加,HIC凋亡率在转染后逐渐增高,HIC凋亡率与TNF-α、IL-6和IL-8的水平呈正相关。结论 EspF可激活多种炎症介质和诱导HIC凋亡,EspF转染HIC后TNF-α、IL-6和IL-8水平的升高在EPEC致病过程中起重要作用。  相似文献

目的:探讨不同残存尿量水平患者接受高通量或低通量透析获益差异。 方法:采用回顾性病例对照设计。选取沈阳军区总医院血液净化中心第一血液透析室维持性血液透析患者。按24小时残存尿量多少分组(尿量<300ml或≥300ml),比较尿毒症毒素水平差异。再按照是否使用高通量透析器进行亚组分析,比较高低通透析对不同残存尿量水平患者的临床指标影响及差异。 结果:对比发现残存尿量较多患者倾向于更低的透析频次(P=0.031),体重指数(P=0.015)显著更高,血清肌酐 (P=0.033)、胱抑素C (P=0.002)、β2微球蛋白 (P=0.000)、铁蛋白(P=0.012)水平显著更低。对于残存尿量较少患者:使用高通量透析器者β2微球蛋白水平(P=0.000)及铁蛋白水平(P=0.043)显著较低,但甲状旁腺激素水平(P=0.023)及体重水平 (P=0.049) 显著较高。对于残存尿量较多患者:使用高低通量透析β2微球蛋白水平(P=0.439)与甲状旁腺激素(P=0.190)水平无显著差异。 结论:使用高通量透析可获得更好的β2微球蛋白清除效果。高通量透析有助于维持更好的体重及体重指数,高通量透析对于不同残存尿量水平患者的生物学效应存在差异。残存尿量较少的患者使用高通量透析获益更多。  相似文献

Objective: To study the therapeutic effect of Weicao Capsule (威草胶囊, WCC) on gout. Methods: Two hundred gout patients were assigned to two groups. The treated group was treated with WCC and the control group was treated with Tongfengding Capsule (痛风定胶囊). Both groups were given the respective treatments orally 3 times a day, 2 capsules each time with 2 weeks as one course and all patients received 2 successive courses of treatment. Changes of blood β2- microglobulin (13 2-M), hemoglobin (Hb), 24 h urinary protein (24 h UP), pH value of urine and blood uric acid (BUA) as well as kidney function were observed. Results: After treatment, level of β 2-M got lowered significantly, Hb and 24 h UP, blood urea nitrogen, serum creatinine and the clearance rate of creatinine, as well as blood lipids all improved obviously in the treated group (all P〈0.01), while these parameters remained unchanged in the control group (P〉0.05). The pH value of urine was improved in both groups showing an insignificant difference between them (P〉0.05). BUA was decreased in both groups with a decrease to a larger extent in the treated group (P〈0.01). The total effective rate was 87% in the treated group, which was superior to that in the control group (62%, P〈0.05). Conclusion: WCC has a favorable therapeutic effect on gout and its mechanism of action for improving renal function and reducing urinary protein could be related with the lowering of blood β 2-M, BUA and lipids.  相似文献
