Brain imaging has been used to predict language skills during development and neuropathology but its accuracy in predicting language performance in healthy adults has been poorly investigated. To address this shortcoming, we studied the ability to predict reading accuracy and single‐word comprehension scores from rest‐ and task‐based functional magnetic resonance imaging (fMRI) datasets of 424 healthy adults. Using connectome‐based predictive modeling, we identified functional brain networks with >400 edges that predicted language scores and were reproducible in independent data sets. To simplify these complex models we identified the overlapping edges derived from the three task‐fMRI sessions (language, working memory, and motor tasks), and found 12 edges for reading recognition and 11 edges for vocabulary comprehension that accounted for 20% of the variance of these scores, both in the training sample and in the independent sample. The overlapping edges predominantly emanated from language areas within the frontoparietal and default‐mode networks, with a strong precuneus prominence. These findings identify a small subset of edges that accounted for a significant fraction of the variance in language performance that might serve as neuromarkers for neuromodulation interventions to improve language performance or for presurgical planning to minimize language impairments.  相似文献

Proper communication between dorsal caudate (CD) and ventral striatum (VS) is likely to be crucial for on‐time responses and its disruption might result in impulsivity. Here, we used functional magnetic resonance imaging (fMRI) with a sensorimotor reaction time task and positron emission tomography (PET) with [11C]raclopride in 14 healthy controls and 18 cannabis abusers to contrast the modulation of striatal fMRI responses by dopamine receptors (D2/D3R) in CD and VS. In controls, we show that the fMRI signals in VS that occurs concomitantly with on‐time responses showed opposite modulation from D2/D3R in CD (inhibitory) and D2/D3R in VS (stimulatory). In contrast, this modulation was not significant in cannabis abusers. Findings suggest that action speed requires balanced VS‐inhibition from D2/D3R in CD and VS‐facilitation from D2/D3R in VS. Using a 9.4 T MRI instrument, we have obtained images of the mouse brain response to photic stimulation during a period between deep anesthesia and the early stages of arousal. The large image enhancements we observe (often >30%) are consistent with literature results extrapolated to 9.4 T. However, there are also two unusual aspects to our findings. (i) The visual area of the brain responds only to changes in stimulus intensity, suggesting that we directly detect operations of the M visual system pathway. Such a channel has been observed in mice by invasive electrophysiology, and described in detail for primates. (ii) Along with the typical positive response in the area of the occipital portion of the brain containing the visual cortex, another area displays decreased signal intensity upon stimulation.  相似文献

The Zucker rat is used as a model of genetic obesity, and while Zucker rats have been well studied for their reduced sensitivity to leptin signaling and subsequent weight gain, little work has examined their responses to environmental signals that are associated with “hedonic” feeding. This study evaluated the effects of a high‐fat food olfactory cue (bacon) in stimulating nose‐poke food‐seeking behavior on first exposure (novel) and after a period of access for consumption (familiar) in lean and obese Zucker rats at either 4 or 12 months of age, and under ad‐lib fed (unrestricted; U) or chronically food‐restricted (70% of ad‐lib; R) conditions. Baseline nose‐poke levels were comparable amongst all groups. At 4 months of age, only ObU rats displayed increased behavioral activation to familiar food cues. Twelve‐month‐old Ob rats, regardless of diet, exhibited substantially greater food‐seeking behavior when exposed to both the novel and familiar olfactory cues. A strong positive correlation between body weight and nose‐poke entries for the familiar food cue was observed at both ages, while this correlation for the novel food cue was significant in 12–month‐old rats only. Similarly, there were strong positive correlations between food intake and poke entries for the familiar food cue was observed at both ages, while this correlation for the novel food cue was significant in 12–month‐old rats only. Although it is possible that differences in olfactory sensitivity contribute to these behavioral effects, our findings support the interactions between food intake, obesity, and food‐seeking behavior and are consistent with leptin inhibiting the brain's reactivity to food cues and suggest that the enhanced sensitivity to the food cues with leptin deficiency is likely to contribute to overeating and weight gain.
