We studied expression and functional activity of tumor cell P-gp in children with various leukemia variants and analyzed its prognostic role in acute lymphoblastic leukemia. Functional activity of P-gp increased in acute myeloid leukemia, relapses of acute lymphoblastic leukemia (in comparison with primary disease), and in a group of patients in whom no remission was attained. Human bone marrow MSC cultured in neurogenic medium containing EGF and FGFb demonstrated alteration of the phenotype and expression of neuronal precursor/early neuron markers nestin and NSE. Signals of expression of neuronal and oligodendroglial markers MAP-2, dm-20, and MBP were detected after prolongation of incubation in neurogenic medium to 2 weeks. Cells with neuronal morphology were immunopositive to early neuronal marker β-III-tubulin. Replacement of neurogenic medium for α-MEM with 10% fetal calf serum induced reversion of the phenotype to that typical for human MSC. This indicates high plasticity of the phenotype and expression profile of neuronal markers in MSC cultured under neurogenic conditions or possibility of dedifferentiation of MSC reaching the stage of neuronal precursors/early neurons.  相似文献

We studied accumulation of porphyrin photosensitizers chlorine e6, hematoporphyrin, and their derivatives by different lymphocyte subpopulations. The intensity of staining of B lymphocytes and natural killer cells with photosensitizers was higher compared to T lymphocytes. T cell subpopulation differed by their ability to bind photosensitizers. Relative accumulation of dimethyl esters of chlorine e6 and hematoporphyrin in cells surpassed that of nonesterified porphyrins. The contents of CD8+, CD4+CD8+, CD3+HLA-DR+, CD+INF-γ+ T cells, and natural killers (CD16+56+) and NK/T cells (CD16+56+CD3+) increase after 7-day culturing in the presence of interleukin-2. The number of apoptotic cells and cells in S-, and G2+M phases of the cell cycle also increased. Interleukin-6 predominantly induced proliferation of CD3+HLA-DR+ T cells and G2+M mitotic cells. Comparison of the lymphocytic IL-2 (lIL-2, lymphokinin) and recombinant IL-2 (rIL-2) showed that lIL-2 is characterized by a more pronounced stimulating effect of low doses on antibody production in mice and on human immunoglobulin production. High doses of lIL-2, in contrast to rIL-2, do not cause hyperstimulation of antibody production or immunoglobulin production. Human lymphokine-activated killer (LAK) cells, induced by rIL-2in vitro during 7 but not 3 days, enhance the production of human immunoglobulins. Monocytes activated by rIL-2 suppress the stimulating effect of LAK cells on immunoglobulin production.
