Point-of-use (POU) water treatment systems and devices play an essential role in limited access to sanitary safe water resources. The filtering materials applied in POU systems must effectively eliminate contaminants, be readily produced and stable, and avoid secondary contamination of the treated water. We report an innovative, 2D Ti3C2/Al2O3/Ag/Cu nanocomposite-modified filtration material with the application potential for POU water treatment. The material is characterized by improved filtration velocity relative to an unmodified reference material, effective elimination of microorganisms, and self-disinfecting potential, which afforded the collection of 99.6% of bacteria in the filter. The effect was obtained with nanocomposite levels as low as 1%. Surface oxidation of the modified material increased its antimicrobial efficiency. No secondary release of the nanocomposites into the filtrate was observed and confirmed the stability of the material and its suitability for practical application in water treatment.  相似文献

Gene therapy using recombinant adenoviral vectors represents a promising therapeutic tool to prevent vein graft stenosis, the main complication of coronary artery bypass grafting. However, the low transduction efficiency of vascular smooth muscle cells and endothelial cells (EC) is a potential limitation, presumably due to the low levels of functional adenovirus receptor (coxsackie:adenovirus receptor; CAR). Designing vectors specifically targeted to alpha(v) integrins is a strategy that might overcome the poor expression of CAR in vascular smooth muscle cells and EC. RGD, a receptor-binding motif that can interact with alpha(v) integrins, was inserted into the HI loop and at the C-terminus of the adenoviral fiber protein in two separate adenovirus vectors encoding a beta-galactosidase reporter gene. Av1nBgCRGD (C-terminus) and Av1nBgHIRGD (HI loop) were evaluated in EC in culture and in jugular vein organ culture. Transduction of primary rat and rabbit EC with Av1nBgHIRGD was significantly more efficient when compared to Av1nBgCRGD or Av1nBg. Transduction of mouse, rat and rabbit jugular veins in organ culture using Av1nBg showed that adenovirus-mediated gene expression was greatest in rabbit jugular veins compared to rat and mouse veins. Av1nBgHIRGD augmented gene expression approximately four-fold in rabbit jugular veins when compared to Av1nBg. Histochemical analysis showed that numerous EC but few smooth muscle cells were transduced at all vector concentrations. A substantial number of adventitial fibroblasts were transduced only at the highest vector concentrations of Av1nBgHIRGD. These findings demonstrate that integrin-targeted vectors allow for enhanced gene delivery to veins and strengthen the viability of adenoviral-mediated gene transfer of therapeutic transgenes to human veins prior to vein grafting.  相似文献

Genetic instability is thought to be responsible for the numerous genotypic changes that occur during neoplastic transformation and metastatic progression. To explore the role of genetic instability at the level of point mutations during mammary tumor development and malignant progression, we combined transgenic mouse models of mutagenesis detection and oncogenesis. Bitransgenic mice were generated that carried both a bacteriophage lambda transgene to assay mutagenesis and a polyomavirus middle T oncogene, mammary gland-targeted expression of which led to metastatic mammary adenocarcinomas. We developed a novel assay for the detection of mutations in the lambda transgene that selects for phage containing forward mutations only in the lambda cII gene, using an hfl- bacterial host. In addition to the relative ease of direct selection, the sensitivity of this assay for both spontaneous and chemically induced mutations was comparable to the widely used mutational target gene, lambda lacI, making the cII assay an attractive alternative for mutant phage recovery for any lambda-based mouse mutagenesis assay system. The frequencies of lambda cII- mutants were not significantly different in normal mammary epithelium, primary mammary adenocarcinomas, and pulmonary metastases. The cII mutational spectra in these tissues consisted mostly of G/C-->A/T transitions, a large fraction of which occurred at CpG dinucleotides. These data suggest that, in this middle T oncogene model of mammary tumor progression, a significant increase in mutagenesis is not required for tumor development or for metastatic progression.  相似文献

The influence of d-amphetamine on the acquisition of light-contingent bar-pressing was assessed in rats in a two-lever operant task as a function of dose and incentive value of the sensory reinforcer. Relative to saline-injected controls, amphetamine increased the probability of discriminated responding on the active reward-bar at the 0.75 but not at the 2.50 mg/kg dosage level. Further, only at the lower dose was the effect of incentive condition significant, with animals receiving variant light-onset emitting a greater percentage of presses on the active bar than animals receiving invariant light-onset. It was concluded that low doses of amphetamine do not enhance all categories of behavior in the rat in a learning situation, but selectively facilitate only those responses directed by the reinforcement contingencies. In addition, the poor performance observed for animals injected with the higher dosage in the present experiment may have been due to the stimulated perseveration of stereotypic behavior patterns which were incompatible with the task requirements.This experiment was conducted during the first author's tenure as a Predoctoral Trainee in Psychology at the Gerontological Psychology Research Laboratory.  相似文献

The combination of fibre metal laminates (FML) and sandwich structures can significantly increase the performance under impact of FMLs. The goal of this work was to create a material that will combine the superior properties of FMLs and foam sandwich structures in terms of the impact resistance and simultaneously have lower density and fewer disadvantages related to the manufacturing. An extensive impact testing campaign has been done using conventional fibre metal laminates (carbon- and glass-based) and in the proposed fibre foam metal laminates to assess and compare their behaviour. The main difference was observed in the energy absorption mechanisms. The dominant failure mechanism for fibre foam laminates is the formation of delaminations and matrix cracks while in the conventional fibre metal laminate the main failure mode is fibre cracking due to high local stress concentrations. The reduction in the fibre cracking leads to a better after-impact resistance of this type of structure improving the safety of the structures manufactured with these materials.  相似文献
