目的:研究全蝎(Scorpio)蛋白药效组分对HepG2细胞的体外抑制作用,为建立全蝎质量生物效应评价方法和标准奠定基础。方法:采用改良MTT法、流式细胞法研究全蝎蛋白药效组分对HepG2细胞毒和细胞周期抑制的作用。结果:抑制癌细胞生长的浓度大于或等于37 mg.mL-1,抑制率与浓度呈正比,相关系数为0.9157,IC50为244 mg.mL-1,显效时间0~48 h;全蝎蛋白药效组分浓度大于或等于9.25 mg.mL-1时,能显著提高HepG2细胞的凋亡率。结论:全蝎蛋白药效组分具有促进HepG2细胞凋亡、抑制增殖的作用,其生物效应指标为9.25~175 mg.mL-1,可作为全蝎质量生物效应评价指标之一。  相似文献

目的建立动物中药材全蝎的鉴定分析新方法。方法用X射线衍射Fourier图谱分析技术对湖北、陕西、山东等不同产地的10个全蝎样品进行分析研究,获取全蝎中药材的对照粉末X射线衍射Fourier指纹图谱。结果除7#样品外,其他9个样品衍射图谱的几何拓扑图形基本一致。9个全蝎样品(除7#)共有11个特征标记峰。结论X射线衍射Fourier指纹图谱鉴定法可用于动物中药材全蝎样品的分析鉴定和质量控制。  相似文献

One hundred three primary total knee arthroplasties using the Scorpio posterior-stabilized total knee system were studied. Clinical, functional, and radiographic assessments were performed preoperatively and at 7 weeks, 6 months, and annually postoperatively. Standard knee scoring systems were used for clinical and functional evaluation. Radiographic assessment was characterized according to standard zones. Presence of anterior knee pain and patellar clunk was also noted. Mean follow-up was 5.25 years. At latest postoperative follow-up, follow-up was 89%. Knee Society scores were 91.3 clinical and 73.0 functional. Mean range of motion increased from a mean preoperative level of 96.5 degrees to 124.5 degrees at latest-year follow-up. There were 4 reports of anterior knee pain. One femoral and 1 tibial radiolucency were found. The Scorpio posterior-stabilized design (Stryker Orthopaedics, Mahwah, NJ) showed favorable midterm results compared with other posterior-stabilized designs.  相似文献

Animal toxins and venoms have recently been developed as cancer treatments possessing tumor cell growth-inhibitory, antiangiogenesis, and proapoptotic effects. Endometriosis is a common benign gynecological disorder in reproductive-age women, and no definite treatment for this disorder is without severe side effects. As endometriosis and malignant tumors share similar characteristics (progressive, invasive, estrogen-dependent growth, and recurrence), animal toxins and venoms are thought to be effective against endometriosis. The objective of this study was to outline studies using toxic animal-based medicinal materials (TMM) as endometriosis treatment and to explore its clinical applicability. Preclinical and clinical studies using TMM were searched for in four databases from inception to October 2020. A total of 20 studies of TMM on endometriosis were included. In eight clinical studies, herbal medicines containing TMM were effective in relieving symptoms of endometriosis, with no side effects. In twelve experimental studies, the main therapeutic mechanisms of TMM against endometriosis were proapoptotic, antiangiogenesis, estrogen level-reducing, and possible anti-inflammatory effects. TMM are thus considered promising sources for the development of an effective treatment method for endometriosis. Further studies are needed to clarify the therapeutic mechanism of TMM against endometriosis and to provide sufficient grounds for clinical application.  相似文献

The effect of the venom of the Chactoid family of scorpions on blood pressure was scantly investigated and was addressed in the present study using the venom of the Israeli scorpion, Scorpio maurus palmatus. Blood pressure in rats was monitored via cannulated femoral artery, while venom and toxins were introduced into femoral vein. Venom injection elicited a biphasic effect, expressed first by a fast and transient hypotensive response, which lasted up to 10 min, followed by a hypertensive response, which lasted up to one hour. It was found that these effects resulted from different venom components. Phospholipase A2 produced the hypotensive effect, while a non-enzymatic neurotoxic polypeptide fraction produced the hypertensive effect. Surprisingly, the main neurotoxic polypeptide to mice had no effect on blood pressure. In vitro experiments indicated that the hypertensive factors caused histamine release from the peritoneal mast cells, but this effect is assumed to be not relevant to their in vivo effect. In spite of the cytotoxic activity of phospholipase A2, it did not release histamine. These findings suggest that the effects of venom and isolated fractions on blood pressure parameters are mediated by different mechanisms, which deserve further pharmacological investigation.  相似文献

目的:建立全蝎、蜈蚣、土鳖虫、僵蚕等4种药材中总磷脂含量测定方法。方法:采用Folch试剂超声提取,钼蓝试剂显色,分光光度法测定10个不同地区的全蝎等4种药材中总磷脂的含量。结果:样品中总磷脂含量以磷计的回归方程Y=0.0238X-0.01(r=0.9994),线性范围在2.38~7.14μg,RSD=0.53%,平均回收率为99.9(%n=5)。结论:采用钼蓝比色法测定全蝎等4种药材中10个不同地区商品中总磷脂含量,方法简便,方法学结果理想,可作为该4种药材质量控制的方法。  相似文献

目的:观察全蝎蜈蚣对胶原免疫性关节炎(Collagen-Induced Arthritis,CIA)大鼠外周血CD4+CD25+FoxP3+Treg细胞的调节作用。方法:Wistar大鼠60只,随机分成6组,分别为正常对照组、模型对照组、全蝎蜈蚣高(0.4 g/kg)、中(0.2 g/kg)、低剂量(0.1 g/kg)组和Ⅱ型胶原蛋白(60μg/kg)治疗组(CⅡ组),二型胶原蛋白诱导大鼠关节炎,并采用流式细胞术检测大鼠外周血CD4+CD25+FoxP3+Treg细胞及酶联免疫吸附试验检测血清中IL-2表达水平。结果:与模型组比较,全蝎蜈蚣高、低剂量组大鼠外周血CD4+CD25+T细胞及CD4+CD25+FoxP3+Treg细胞水平显著升高,且中、低剂量组血清IL-2水平显著下降。结论:全蝎蜈蚣治疗关节炎大鼠可能是通过上调外周CD4+CD25+FoxP3+Treg细胞表达水平以恢复或重建免疫耐受而实现的。  相似文献

目的:研究全蝎(Scorpio)蛋白药效组分对Bel7402肿瘤细胞凋亡的影响,为建立全蝎与临床疗效对应的生物效应质量评价方法奠定基础。方法:蛋白质提取和冷冻干燥的方法获得全蝎蛋白药效组分,采用改良MTT法、流式细胞法研究全蝎蛋白药效组分对Bel7402细胞毒和细胞周期抑制的作用。结果:抑制癌细胞生长的浓度大于或等于37 g.L-1,剂量与效应呈正比,相关系数为0.912 5,IC50为241 g.L-1,显效时间0~48 h;全蝎蛋白药效组分浓度大于或等于9.25 g.L-1时,能显著提高Bel7402细胞的凋亡率。结论:全蝎蛋白药效组分具有促进Bel7402细胞凋亡、抑制增殖的作用,其生物效应指标为:9.25~175 g.L-1,可作为全蝎生物效应质量评价的指标之一。  相似文献
