BackgroundIschemia reperfusion (I/R) play an imperative role in the expansion of cardiovascular disease. Sinomenine (SM) has been exhibited to possess antioxidant, anticancer, anti-inflammatory, antiviral and anticarcinogenic properties. The aim of the study was scrutinized the cardioprotective effect of SM against I/R injury in rat.MethodsRat were randomly divided into normal control (NC), I/R control and I/R + SM (5, 10 and 20 mg/kg), respectively. Ventricular arrhythmias, body weight and heart weight were estimated. Antioxidant, inflammatory cytokines, inflammatory mediators and plasmin system indicator were accessed.ResultsPre-treated SM group rats exhibited the reduction in the duration and incidence of ventricular fibrillation, ventricular ectopic beat (VEB) and ventricular tachycardia along with suppression of arrhythmia score during the ischemia (30 and 120 min). SM treated rats significantly (P < 0.001) altered the level of antioxidant parameters. SM treatment significantly (P < 0.001) repressed the level of creatine kinase MB (CK-MB), creatine kinase (CK) and troponin I (Tnl). SM treated rats significantly (P < 0.001) repressed the tissue factor (TF), thromboxane B2 (TXB2), plasminogen activator inhibitor 1 (PAI-1) and plasma fibrinogen (Fbg) and inflammatory cytokines and inflammatory mediators.ConclusionOur result clearly indicated that SM plays anti-arrhythmia effect in I/R injury in the rats via alteration of oxidative stress and inflammatory reaction.  相似文献

Background: The sea cucumber, Bohadschia marmorata, is a marine echinoderm consumed and used as a medication. Extract of this species displays a broad spectrum of bioactivity, such as antifungal, antibacterial, immunomodulatory, and cytotoxic properties. This investigation explored sea cucumber extract for hepatorenal protection against the toxicity of methotrexate (MTX). Methods: Four groups of mice were divided into G1: control, G2: MTX treated, G3: B. marmorata extract-treated daily for 14 days, and G4: B. marmorata extract and MTX treated. Results and Conclusions: Biochemical analysis and histopathological examination of liver tissue showed that administration of MTX increased serum levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT), lowered levels of serum albumin, total protein, Superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH). Administration of B. marmorata extract to MTX- injected mice significantly reversed the increase in serum levels of liver enzymes and induced a significant elevation in serum albumin and total protein levels. SOD, CAT, and GSH levels returned to nearly normal levels. Histopathological examination indicated fewer signs of toxicity in liver and kidney tissues of mice treated with both extract and MTX compared to MTX treatment alone. An extract of B. marmorata will protect mice from hepatorenal toxicity induced by MTX.  相似文献

目的评估CHA2DS2-VASc评分对急性心肌梗死(AMI)患者院内结局事件的预测价值。方法回顾性分析冠心病医疗结果评价和临床转化研究(China PEACE)回顾性急性心肌梗死研究中23728例AMI患者的病历信息,按CHA2DS2-VASc评分分为低(0~3分)、中(4~6分)、高(7~9分)分值组。院内结局包括主要不良心血管事件、死亡、死亡或放弃治疗、再发心肌梗死、缺血性卒中等。采用多因素Cox回归分析CHA2DS2-VASc评分对AMI患者院内结局的影响。通过受试者工作特征(ROC)曲线,评估CHA2DS2-VASc评分对AMI患者院内死亡与死亡或放弃治疗的预测价值。结果入组患者年龄66(56,75岁)岁,女性占30.7%。CHA2DS2-VASc评分高分值组患者院内结局事件发生率更高,基础疾病更多(P值均<0.001);多因素logistic回归中,院内病死率(OR=6.13,95%CI 4.77~7.87,P<0.001)、院内死亡或放弃治疗率(OR=6.43,95%CI 5.16~8.00,P<0.001)、主要心血管事件发生率(OR=4.94,95%CI 4.06~6.01,P<0.001)明显高于其他两组。ROC曲线分析显示,无论院内病死率,还是死亡或放弃治疗率,CHA2DS2-VASc评分与简化版全球急性冠状动脉事件登记(global registry of acute coronary events,mini-GRACE)评分相比差异无统计学意义(ROC曲线下面积:0.699与0.696,P=0.752;0.708与0.713,P=0.489)。结论CHA2DS2-VASc评分是一种有效预测AMI患者院内风险的评估工具,该评分操作简单,预测价值与mini-GRACE评分相当。  相似文献

目的评价益气通脉汤治疗孤立性冠状动脉肌桥心绞痛气虚血瘀证患者的临床疗效。方法将符合入选标准的2016年4月-2020年1月首都医科大学附属北京中医医院64例孤立性冠状动脉肌桥心绞痛气虚血瘀证患者,采用随机数字表法分为2组,每组32人。对照组口服盐酸地尔硫卓片,治疗组在对照组基础上服用自拟益气通脉汤。2组均连续治疗8周。分别于治疗前后进行中医证候评分,采用西雅图心绞痛量表评估患者生活质量及机体功能状态,观察治疗前后平板运动心电图试验情况,记录运动诱发心绞痛需要的时间及平板运动后引起心电图中ST段动态演变的时间;评价心绞痛疗效及中医证候疗效。结果治疗组心绞痛疗效总有效率为84.4%(27/32)、对照组为53.1%(17/32),2组比较差异有统计学意义(χ2=8.09,P<0.05)。治疗后,治疗组西雅图心绞痛量表中躯体活动受限程度[(69.24±14.21)分比(59.42±11.71)分,Z=-2.61]、心绞痛稳定状态[(82.25±21.24)分比(69.11±19.52)分,Z=-2.64]、心绞痛发作情况[(80.24±18.31)分比(69.11±15.54)分,Z=-2.63]、治疗满意程度[(86.16±19.23)分比(61.19±17.35)分,Z=-2.22]、疾病认知程度[(74.41±21.13)分比(60.43±19.42)分,Z=-2.40]评分高于对照组(P<0.05);治疗组运动诱发心绞痛时间[(476.15±62.15)s比(399.38±78.42)s,Z=-2.08]、运动后ST段下移1 mm时间[(394.54±75.61)s比(309.64±81.62)s,Z=-2.40]较对照组延迟(P<0.05)。治疗组中医证候疗效总有效率为93.8%(30/32)、对照组为65.6%(21/32),2组比较差异有统计学意义(χ2=7.96,P<0.05)。治疗组治疗后中医证候积分[(25.15±6.15)分比(36.38±10.42)分,Z=-2.56]低于对照组(P<0.05)。治疗期间,2组均未见明显不良反应。结论益气通脉汤可有效改善孤立性冠状动脉肌桥心绞痛气虚血瘀证患者的临床症状,减少心绞痛发作,延迟运动诱发心绞痛时间,提高临床疗效。  相似文献

Introduction and objectivesThe index of microcirculatory resistance (IMR) measured after primary percutaneous coronary intervention (PCI) for ST-segment elevation myocardial infarction (STEMI) is associated with microvascular obstruction (MVO) and adverse clinical events. To evaluate MVO after successful primary PCI for STEMI without pressure wires or hyperemic agents, we investigated the feasibility and usefulness of functional angiography-derived IMR (angio-IMR).MethodsThe current study included a total of 285 STEMI patients who underwent primary PCI and cardiac magnetic resonance (CMR). Angio-IMR of the culprit vessel after successful primary PCI was calculated using commercial software. MVO, infarct size, and myocardial salvage index were assessed using CMR, which was obtained a median of 3.0 days [interquartile range, 3.0-5.0] after primary PCI.ResultsAmong the total population, 154 patients (54.0%) showed elevated angio-IMR (> 40 U) in the culprit vessel. MVO was significantly more prevalent in patients with angio-IMR > 40 U than in those with angio-IMR ≤ 40 U (88.3% vs 32.1%, P < .001). Infarct size, extent of MVO, and area at risk were significantly larger in patients with angio-IMR > 40 U than in those with angio-IMR ≤ 40 U (P < .001 for all). Angio-IMR showed a significantly higher discriminatory ability for the presence of MVO than thrombolysis in myocardial infarction flow grade or myocardial blush grade (area under the curve: 0.821, 0.504, and 0.496, respectively, P < .001).ConclusionsAngio-IMR was significantly associated with CMR-derived infarct size, extent of MVO, and area at risk. An elevated angio-IMR (> 40 U) after primary PCI for STEMI was highly predictive of the presence of MVO in CMR.This trial was registered at ClnicalTrialsgov (Identifier: NCT04828681).  相似文献

von Willebrand factor (VWF) plays a crucial role in hemostasis and thrombosis. VWF is involved in platelet attachment to the subendothelium, serving as a carrier protein for coagulation factor VIII. In this study, myocardial tissues from deceased patients with ischemic heart disease and a mouse model of acute myocardial infarction were subjected to immunohistochemistry to determine VWF expression. We examined 28 neutral formalin-fixed, paraffin-embedded myocardial tissue samples obtained from the autopsies of patients who were diagnosed with ischemic heart disease within 48 h postmortem. Most myocardial cells were negative for VWF, although some cells showed nonspecific positivity. Elevated VWF expression was observed around myocardial cells undergoing remodeling, suggesting that endothelial proliferation occurred at these sites. In contrast, completely fibrotic myocardial foci did not show upregulated VWF expression. Positivity in fibrin deposition and hemorrhagic sites was observed. The same VWF expression characteristics as those observed in the human samples were observed in the mouse model. VWF immunostaining as an endothelial marker may be a useful supplementation to conventional staining techniques that are currently used in the diagnosis of ischemic heart disease in terms of examining the timing of myocardial remodeling in detail and highlighting the remodeling process.  相似文献
