上皮性卵巢肿瘤组织中MMP2和MMP9的表达及临床意义   总被引:1,自引:0,他引:1  
目的研究基质金属蛋白酶(MMP2和MMP9)在上皮性卵巢肿瘤组织中的表达及临床意义。方法应用免疫组化EnVision法检测38例上皮性卵巢癌(A组)、20例交界性上皮性卵巢肿瘤(B组)、20例良性上皮性卵巢肿瘤(C组)和20例正常卵巢组织(D组)中MMP2和MMP9的表达和预后。结果 MMP2和MMP9在上皮性卵巢癌中的阳性率分别为94.7%和92.1%,显著高于良性卵巢肿瘤(40%)和正常卵巢(40%和50%),A组中的强阳性率(73.7%和78.9%)显著高于B组(27.2%和31.8%)、C组(5%)和D组(10%)(P<0.05);晚期上皮性卵巢癌中的强阳性率(94.4%和88.9%)显著高于早期的强阳性率(40%和45%)(P<0.05);中低分化(G2~G3级)上皮性卵巢癌中的强阳性率(53.3%和40%)显著高于高分化(G1级)的强阳性率(12.5%)(P<0.05);有淋巴转移的上皮性卵巢癌中的强阳性率(94.7%和89.5%)显著高于无淋巴转移组的强阳性率(68.4%和73.7%)(P<0.05)。结论 MMP2和MMP9可能参与卵巢的正常生理功能,它的异常高表达在上皮性卵巢癌的浸润、转移和交界性上皮性卵巢肿瘤的复发中起了重要作用。  相似文献

Aim of study

Placenta extract features as a composition of ointments used for skin beautification, dermatological diseases and skin dryness. However, little evidence has been cited about its underlying mechanisms of action by which it exerts a beneficial role in dermatological diseases in vivo. In this study, we intended to test the effect of topical application of porcine placenta extract in mouse model of contact hypersensitivity and elucidate its mechanism of action.

Materials and methods

To test the in vitro effect of porcine placenta extract, RAW 264.7 cells were cocultured with porcine placenta extract and stimulated with LPS (1 μg/ml) and the expression of inflammatory mediator TNF-α was estimated by RT-PCR at the mRNA level and by intracellular staining at the protein level. To further test in vivo efficacy, porcine placenta extract was topically applied to the mice with experimental skin hypersensitivity. For in vivo studies placenta extract in gel form was topically applied to ear of DNCB-induced contact hypersensitivity mouse model everyday for 2 weeks and progression of the disease was estimated by following criteria: (a) ear thickness, (b) serum IgE level by ELISA, (c) histological examination of ear tissue by H&E staining and (d) cytokine profile of total cells and CD4+ T cells by real time PCR.

Results

Topical application of porcine placenta extract on mouse ears with contact hypersensitivity decreased the severity and progression of the disease manifested by reducing ear swelling, inflammation and edema. Histological evaluation showed that placenta extract treatment reduced lymphocyte infiltration in the ear tissues. Protective effect of placenta extract is also associated with down-regulation of serum IgE level and inflammatory cytokine production (IL-1β, IFN-γ, TNF-α, IL-4, IL-12 and IL-17) in total lymph node cells and CD4+ T cells.

Conclusions

Our data indicate that protective effect of porcine placenta extract in contact hypersensitivity is mediated by inhibition of the inflammatory responses and IgE production, suggesting a potential therapeutic application of porcine placenta extract to modulate skin inflammation.  相似文献

目的检测宫颈癌患者血清中细胞因子IL-17的表达水平,探讨IL-17的变化在宫颈癌临床监测中的意义。方法收集51例宫颈癌患者外周血标本,酶联免疫吸附试验(ELISA)检测血浆IL-17的表达。结果宫颈癌患者外周血IL-17的表达水平均明显高于正常对照组(P<0.05),宫颈癌浸润组与正常对照组相比,IL-17明显升高(P<0.05)。结论宫颈癌患者血浆IL-17明显升高,提示Th17细胞与宫颈癌的发生发展可能存在着一定的关系。  相似文献

Ethnopharmacological relevance

Clematichinenoside AR (AR) has been defined as a major active ingredient of triterpenoid saponins extracted from Clematidis Radix et Rhizoma, which is a traditional Chinese herbal medicine that has long been used in the treatment of rheumatoid arthritis (RA). To further explore the mechanism of AR in the treatment of RA, we investigated whether its immunomodulatory effects are related to Treg-mediated suppression derived from Peyer?s patches (PPs) in adjuvant induced arthritis (AIA) rat model.

Materials and methods

AR (8, 16, 32 mg/kg) was orally administered daily from Day 18 to Day 31 after immunization. The effect of AR on AIA rats was evaluated by hind paw swelling and histopathological examination. Percentages of CD4+CD25+Foxp3+ T regulatory cells were determined by flow cytometry. Levels of IL-10, TGF-β1, IL-17A and TNF-α were measured by ELISA. Expressions of Foxp3 and RORγ in synovium were detected using immunohistochemical analysis.

Results

AR treatment significantly reduced paw swelling of AIA rats, and histopathological analysis confirmed it could suppress severity of established arthritis. AR treatment upregulated the percentages of CD4+CD25+Foxp3+ Treg cells among CD4+ T cells in PPs lymphocytes, and increased the levels of IL-10 and TGF-β1 secreted from ConA-activated PPs lymphocytes, whereas decreased the levels of IL-17 A and TNF-α. Similar tendency of circulating CD4+CD25+Foxp3+ Treg cells percentages and serum cytokine levels were observed. Moreover, AR decreased the expression levels of Foxp3 and RORγ in joint synovial membrane.

Conclusions

In conclusion, these results suggested AR has a potent protective effect on the progression of AIA, probably by augmenting CD4+CD25+Foxp3+ Treg cells in PPs to induce immunosuppression, and modulating the balance between Treg cells and Th17 cells systemically. These findings may help to develop AR as a potent immunosuppressive agent for the treatment of RA.  相似文献

目的:研究上中下通用痛风汤对尿酸钠诱导大鼠急性痛风性关节炎(GA)的治疗作用及对Th17细胞的影响。方法:取 60 只大鼠,随机分成 6 组,对照组、模型组、吲哚美辛组、上中下通用痛风汤高、中、低剂量组(20,10,5 g·kg-1),每组 10 只。上中下通用痛风汤组ig给药,1次/d,连续7 d,吲哚美辛(5 mg·kg-1)组第7天开始给药,末次给药1 h后,按Coderre经典方法,除空白组外,其余5 组采用尿酸钠晶体溶液踝关节注射建立急性痛风性关节炎模型。以上中下通用痛风汤与吲哚美辛对比治疗,观察大鼠步态,测定关节肿胀度,流式细胞术检测外周血Th17细胞含量,ELISA方法检测血清白介素(IL)-17,IL-6水平。结果:上中下通用痛风汤高、中、低剂量在致炎后各时间点均能不同程度的抑制尿酸钠所致大鼠踝关节疼痛,减轻肿胀程度,降低步态评分,降低外周血Th17细胞含量。高剂量组Th17细胞含量(1.65±0.25)%明显低于模型组(3.25±0.96)%,相关炎性因子IL-17含量(0.54±0.09)μg·L-1明显低于模型组(0.69±0.07)μg·L-1,IL-6含量亦明显低于模型组,差异有显著性(P<0.05)。结论:上中下通用痛风汤对尿酸钠所致急性痛风性关节炎症有较好的抗炎镇痛作用,其作用机制可能与降低Th17细胞及炎症因子IL-17,IL-6水平有关。  相似文献

目的:探讨实验性变应性鼻炎(llergic rhinitis,AR)发病机制中白介素(IL)-6、IL-17和免疫球蛋白E(IgE)水平的变化及其意义,分析消风宣窍汤对这3种细胞因子的调节作用。方法:用卵蛋白(OVA)通过腹腔注射结合滴鼻的方式建立AR豚鼠模型,造模周期共21 d,将60只Hartley豚鼠随机分为正常组、模型组、消风宣窍汤高、中、低剂量组(42,21,10.5 g·kg-1)、氯雷他定组(1.34 mg·kg-1)。用被动皮肤过敏试验(PCA)检验豚鼠模型制备情况,ig治疗1周,ELISA法检测各组血清中免疫球蛋白(IgE)和IL-6,IL-17的水平。结果:与正常组比较,模型组IgE和IL-6,IL-17水平明显升高(P<0.01);与模型组比较,消风宣窍汤高、中剂量组IgE水平显著降低(P<0.01),IL-6,IL-17水平降低(P<0.05或P<0.01);低剂量组IgE水平降低(P<0.05);氯雷他定组IgE水平降低(P<0.05),IL-6,IL-17水平显著降低(P<0.01)。结论:消风宣窍汤降低AR的IgE水平的机制可能是通过降低IgE和IL-6,IL-17水平实现的,并且两者呈正相关关系,提示IL-6和IL-17在AR发病机制中有重要作用。  相似文献
