目的:探讨鼻咽癌患者血清中miR-141-3p及miR-155-3p的表达水平及临床意义。方法:选取2017年01月至2020年09月我院收治的95例鼻咽癌患者和45例健康对照组作为研究对象,采用实时荧光定量PCR法检测miR-141-3p及miR-155-3p表达水平。应用受试者工作特征(receiver operating characteristic, ROC)曲线分析血清中miR-141-3p及miR-155-3p表达水平对鼻咽癌的诊断价值。Pearson相关分析分析鼻咽癌患者血清中miR-141-3p与miR-155-3p表达水平的相关性。结果:鼻咽癌组血清中miR-141-3p(3.25±1.28 vs 0.64±0.17)及miR-155-3p(1.47±0.83 vs 0.35±0.08)表达水平均明显高于对照组(P均<0.001)。鼻咽癌患者血清中miR-141-3p及miR-155-3p表达水平升高与临床分期高、分化程度低、淋巴结转移及远处转移相关(P均<0.001)。ROC曲线显示,血清中miR-141-3p及miR-155-3p表达水平诊断鼻咽癌的最佳截断值分别为2.18、0.97,两项联合诊断鼻咽癌的曲线下面积(0.940,95%CI:0.877~0.998)最大,其敏感度和特异度为97.0%和86.2%。相关分析显示,鼻咽癌患者血清中miR-141-3p与miR-155-3p表达水平呈正相关(r=0.853,P<0.001)。结论:鼻咽癌患者血清中miR-141-3p及miR-155-3p表达水平明显升高,有望作为鼻咽癌诊断的潜在标志物。 相似文献
miR-150 has been demonstrated to inhibit tumor progression in various human cancers, including colorectal
cancer, ovarian cancer, and thyroid cancer. However, the role of miR-150 in melanoma remains to be determined. In this study, we found that miR-150 was underexpressed in melanoma tissues and cell lines. Through
transfection of miR-150 mimics, we found that miR-150 significantly inhibited the proliferation, migration,
and invasion of melanoma cells. In mechanism, we found that MYB was a target of miR-150 in melanoma
cells. Overexpression of miR-150 significantly inhibited mRNA and protein levels of MYB in melanoma cells.
Moreover, there was an inverse correlation between the expression of miR-150 and MYB in melanoma tissues.
We also showed that MYB was upregulated in melanoma tissues and cell lines. Through functional experiments, we found that restoration of MYB in miR-150-overexpressed melanoma cells rescued the proliferation, migration, and invasion. Therefore, our findings demonstrated that miR-150 suppressed the proliferation,

Renal carcinoma greatly threatens human health, but the involved molecular mechanisms are far from complete
understanding. As a master oncogene driving the initiation of many other cancers, ZBTB7 has not been established to be associated with renal cancer. Our data revealed that ZBTB7 is highly expressed in renal carcinoma
specimens and cell lines, compared with normal cells. The silencing of ZBTB7 suppressed the proliferation
and invasion of renal cancer cells. ZBTB7 overexpression rendered normal cells with higher proliferation
rates and invasiveness. An animal study further confirmed the role of ZBTB7 in the growth of renal carcinoma. Moreover, miR-137 was identified to negatively regulate the expression of ZBTB7, and its abundance is
inversely correlated with that of ZBTB7 in renal carcinoma specimens and cell lines. ZBTB7 overexpression
may be induced by miR-137 downregulation. Interestingly, ZBTB7 can also suppress miR-137 expression by
binding to its recognition site within the miR-137 promoter region. Taken together, we identified an autoregulatory loop consisting of ZBTB7 and miR-137 in gastric cancers, and targeting this pathway may be an effective

Objective: To investigate the changes in serum miR-124 levels in patients with acute cerebral infarction (ACI) and elucidate the underlying mechanism by a dynamic monitor.
Methods: Fifty-four patients with ACI and 51 healthy controls were included in our study. Baseline characteristics and blood samples were collected for further analysis. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to measure the serum miR-124 levels. The dual-luciferase reporter assay was used to evaluate the effect of miR-124 on iASPP, a protein that inhibits apoptosis stimulating proteins in the p53 family.
Results: Compared with normal controls, the miR-124 levels in the ACI group rapidly decreased at phase 1 (within 24?h after ischemia) (p?<?0.001) and then gradually increased at phase 2 (48?~?72?h after ischemia) (p?<?0.001) and phase 3 (the 7th day after ischemia) (p?<?0.001). The dual-luciferase reporter assay showed that miR-124 down-regulates iASPP expression in 293T cells.
Conclusion: The miR-124 levels are down-regulated in ACI patients. The dynamic changes of miR-124 might provide a possible method for the detection of ischemic stroke.

Reactive Red 195 was detected from industrial waste samples electrochemically on graphene modified glassy carbon electrode (GCE), using both bare and surface changed GCE at different pH media from 1.0 to 13.0. The optimum pH was determined to be 4.0. RR 195 exhibited good linear responds at pH 4.0 on both electrode surfaces. Other parameters, such as accumulation potential, accumulation time, initial scan potential, pulse height, pulse width, and potential scan increment and scan rate are optimized and calibration plot was also derived on different concentrations of the dye. The stripping voltammetric behavior of dye exhibits very low limit of detection on graphene coated electrode (30?ppb). The adsorption of compound on GCE and graphene coated GCE are confirmed using atomic force microscopy studies. 相似文献
