枯草杆菌Bacillus sp F26产过氧化氢酶的发酵条件

从内蒙古呼伦贝尔草原的盐碱湖中分离到的一株低度嗜盐嗜碱细菌Bacillus sp F26，能积累高水平过氧化氢酶（CAT）。对Bacillus sp F26发酵产过氧化氢酶的环境与营养条件的研究结果表明，其积累高水平过氧化氢酶的适宜环境条件为：温度37℃，种龄20-22h，接种量5％，装液量50mL／（250mL的摇瓶）。适宜发酵培养基组成（g／L）为：葡萄糖15，牛肉膏10，玉米浆10，酵母膏5，磷酸二氢钾1，氯化镁0．2，氯化钠50，碳酸钠10。采用上述条件进行摇瓶分批发酵实验，发酵20h，过氧化氢酶酶活达到16．32U／mL，细胞干重为4．12g／L。进一步研究发现，在对数生长后期（16h）添加2mmol／L的H2O2可以明显刺激产酶，在5L的发酵罐上进一步以指数速率方式流加H2O2，由于该流加方式可降低H2O2对细胞的毒害作用，过氧化氢酶酶活达到29．89U／mL，与分批发酵相比提高了92．8％。     

Development and optimization of an indirect enzyme-linked immunosorbent assay for the determination of Hexoestrol

An indirect enzyme-linked immunosorbent assay (ELISA) for the detection of hexoestrol (HES) was developed, optimized and validated for the analysis of HES in pork. Many parameters, such as the volume ratio of solution A and solution B, colour developing time, pH value, incubation time, the volume ratio of the standard solution and diluted antiserum, blocking solution, blocking condition, coating solution and coating condition were studied and optimized in the paper. The regression equation of the final inhibition curve is y = - 0.3345x + 1.4955, R²=0.9913. The linear range is 0.1-8.1 ng/ml, and the IC₅₀ is 0.671 ng/ml. The specificity of the assay was evaluated by the cross-reactivity rates of six compounds, of which two structurally related compounds had a relatively higher cross-reactivity rate of 25% and 6%. HES was added into a pork sample at 5 ng/g level and then the sample was extracted. The recovery is between 49.6% and 79.2%, and the variation coefficient is 0.164.     

耐酸性运动发酵单胞菌的筛选和酒精发酵条件的优化

通过酸性平板筛选,在作者所在实验室保藏的运动发酵单胞菌中,得到一株能在pH4.5时进行酒精发酵的菌株,其酒精产量为66.7g/L,与原菌株在pH6.5时的相同.通过单因素实验和正交实验,对该菌在酸性条件下的酒精发酵条件进行了优化.结果表明,在发酵温度为30～35℃,pH4.5,初糖质量浓度150g/L,酵母膏质量浓度4g/L,蛋白胨质量浓度为2g/L时,该菌的酒精产量达到最大,为71.7g/L.     

采用生活垃圾研制烧结砖

该文介绍了用好氧发酵工艺对新鲜生活垃圾进行预处理,用做制砖的原料。制砖工艺采用对辊,双轴搅拌,真空挤出,轮窑烧成等工序,垃圾掺加量为30％～50％,可完全取代内掺材料,节省90％以上的煤炭。     

Polymeric genes MEL8, MEL9 and MEL10 — new members of α-galactosidase gene family in Saccharomyces cerevisiae

Summary We used a combination of genetic hybridization analysis and electrokaryotyping with radioactively labelled MEL1 gene probe hybridization to isolate and identify seven polymeric genes for the fermentation of melibiose in strain CBS 5378 of Saccharomyces cerevisiae (syn. norbensis). Four of the MEL genes, i.e. MEL3, MEL4, MEL6 and MEL7, were allelic to those found in S. cerevisiae strain CBS 4411 (syn. S. oleaginosus) whereas three genes, i.e. MEL8, MEL9 and MEL10 occupied new loci. Electrokaryotyping showed that all seven MEL genes in CBS 5378 were located on different chromosomes. The new MEL8, MEL9 and MEL10 genes were found on chromosomes XV, X/XIV and XII, respectively.

Optimization of parameter values for complex pulse sequences by simulated annealing: Application to 3D MP-RAGE imaging of the brain

A number of pulse sequence techniques, including magnetization-prepared gradient echo (MP-GRE), segmented GRE, and hybrid RARE, employ a relatively large number of variable pulse sequence parameters and acquire the image data during a transient signal evolution. These sequences have recently been proposed and/or used for clinical applications in the brain, spine, liver, and coronary arteries. Thus, the need for a method of deriving optimal pulse sequence parameter values for this class of sequences now exists. Due to the complexity of these sequences, conventional optimization approaches, such as applying differential calculus to signal difference equations, are inadequate. We have developed a general framework for adapting the simulated annealing algorithm to pulse sequence parameter value optimization, and applied this framework to the specific case of optimizing the white matter-gray matter signal difference for a T₁-weighted variable flip angle 3D MP-RAGE sequence. Using our algorithm, the values of 35 sequence parameters, including the magnetization-preparation RF pulse flip angle and delay time, 32 flip angles in the variable flip angle gradient-echo acquisition sequence, and the magnetization recovery time, were derived. Optimized 3D MP-RAGE achieved up to a 130% increase in white matter-gray matter signal difference compared with optimized 3D RF-spoiled FLASH with the same total acquisition time. The simulated annealing approach was effective at deriving optimal parameter values for a specific 3D MP-RAGE imaging objective, and may be useful for other imaging objectives and sequences in this general class.     

聚γ-谷氨酸高产突变株的选育及摇瓶发酵条件

对地衣芽孢杆菌(Bacilluslicheniformis)进行亚硝基胍和60Co诱变,获得一株γ PGA的高产菌株C9.γ PGA质量浓度由9.44g/L提高到19.76g/L,提高了109%.突变株传代10次,质量浓度保持基本稳定.通过正交试验和单因素试验对发酵培养基及发酵条件进行了优化.当发酵培养基中含柠檬酸12g/L、甘油80g/L、L 谷氨酸23g/L、氯化铵7g/L,pH7.0,装液量为50mL/250mL三角瓶,接种体积分数为5%时,37℃摇瓶发酵72h,γ PGA达到23.32g/L.     

刺五加发酵乳饮料的生产工艺

以刺五加嫩叶和鲜牛乳为主要原料，杀茵后接种乳酸茵进行乳酸发酵，在此基础上添加各种辅料制成发酵乳饮料，通过单因素试验及正交试验确定原料的配比、发酵条件及产品的配方，所得产品各项指标符合国家要求，色泽淡绿乳色，酸甜适中，口感细腻，香味浓郁。
