目的验证内皮细胞和平滑肌细胞(EC/SMC)移植可通过促进血管生成和骨再生治疗股骨头坏死。方法先建立无水乙醇诱导兔股骨头坏死模型,将制备的组织工程聚合物凝胶注射到骨坏死区域。分为未干预组、细胞/凝胶组、微球/凝胶组、细胞/微球/凝胶组,干预后第2、4、6周,通过影像学(X线和MRI)和组织病理学(HE染色)检测股骨头坏死后血管和骨组织再生。使用Image-Pro Plus图像分析软件对新生血管密度进行分析。结果 X线、MRI和组织病理学检测均显示骨坏死面积缩小,新生毛细血管和新生骨修复坏死区域。图像软件定量分析细胞/微球/凝胶组的新生血管密度明显高于其它各组。结论骨坏死区域形成稳定和功能成熟的血管网证明EC/SMC在治疗性血管再生的作用。局部注射组织工程化凝胶提供了治疗早期股骨头坏死的新方法。  相似文献

Abstract

Nonwoven fabrics prepared via an electrospinning method, so-called electrospun fibermats, are expected to be promising scaffold materials for bone tissue engineering. In the present work, poly(L-lactic acid) (PLLA) fibermats, consisting of fibers with diameters ranging from 1 to 10 μm, were prepared by electrospinning. Mouse osteoblast-like cells (MC3T3-E1) were seeded on the fibermats with various fiber diameters (10, 5 and 2 μm; they are denoted by samples A, B and C, respectively) and cultured in two different directions in order to compare the migration behaviours into the scaffold of the normal condition and the anti-gravity condition. The cells in/on the fibermats were observed by laser confocal microscopy to estimate the cellular migration ability into them. When the MC3T3-E1 cells were cultured in the normal direction, the thickness of their layer increased to approx. 90 μm in the sample A, consisting of 10-μm fibers after 13 days of culture, while that in the sample C, consisting of 2-μm fibers, did not increase. When the MC3T3-E1 cells were cultured in the anti-gravity condition, the thickness of the cell layer in the sample A increased to approx. 60 μm. These results mean that the MC3T3-E1 cells migrated into the inside of sample A in either the normal direction or the anti-gravity one. The cellular proliferation showed no significant difference among the fibermats with three different fiber diameters; MC3T3-E1 cells on the fibermat with 2 μm fiber diameter grew two-dimensionally, while they grew three-dimensionally in the fibermat with 10 μm fiber diameter.  相似文献

目的:制备载乳铁蛋白壳聚糖微球/nHA/Co复合材料作为组织工程材料拟修复骨缺损。方法:通过乳化交联法法制备载乳铁蛋白壳聚糖微球,化学沉淀法制备纳米羟基磷灰石,改良酸酶法提取Ⅰ型胶原,应用京尼平交联制备复合材料,SEM检测材料的微观结构,测定壳聚糖微球对乳铁蛋白的载药率和包封率。结果:复合材料及各组成成份扫描电镜观察均具有较好的微观形态,壳聚糖微球对乳铁蛋白的载药率为(1.06±0.06)%,包封率为(88.2±2.9)%。结论:载乳铁蛋白壳聚糖微球/nHA/Co复合材料具有较好的微观形态。  相似文献

The goal of reconstruction of the alveolar cleft in patients with cleft lip and palate is to improve the quality of tissue, the structural stability, and increase the volume of bone. This study is a systematic review with meta-analysis of volumetric bony filling using autogenous bone and various tissue-engineered bone substitutes. We made an electronic search on MEDLINE, EMBASE, SCOPUS, WEB OF SCIENCE, “grey” publications (materials and research produced by organisations outside traditional channels for commercial or academic publishing and distribution), and relevant cross references according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Studies that reported the outcomes of volumetric grafting were included in the meta-analysis. Of 1276 studies, 26 were included in the meta-analysis. Pooled analysis of 25 studies that used autogenous bone showed a significant reduction in the volume of the cleft equivalent to 62.0% bone fill (95% CI 54.3 to 69.6), in contrast to 10 studies that used a tissue-engineered material and reported bone filling of 68.7% (95% CI 54.5 to 82.8). The estimated sizes of pooled effects across studies showed that there was no significant difference between the two major intervention groups (p value 0.901). Our statistical analysis showed that autogenous bone grafts did not differ significantly from tissue-engineered materials in their ability to fill clefts.Systematic review registration: International Prospective Register of Systematic Reviews, PROSPERO (CRD42017065045).  相似文献

The major problem in skin grafting is that tissue-engineered skin grafts after their transplantation are initially entirely dependent on diffusion. Since this process is slow and inefficient, nutrients, growth factors, and oxygen will insufficiently be supplied and the regenerating graft will undergo a physiological crisis, resulting in scar-like dermal structures and shrinkage. The tissue-engineering of a vascular network in human dermo-epidermal skin substitutes (DESS) is a promising approach to overcome this limitation. Here we report, for the first time, on the use of the adipose stromal vascular fraction (SVF)-derived endothelial cell population to tissue-engineer DESS containing a highly efficient capillary plexus. To develop vascular networks in vitro, we employed optimized 3D fibrin or collagen type I hydrogel systems. Upon transplantation onto immune-deficient rats, these pre-formed vascular networks anastomosed to the recipient's vasculature within only four days. As a consequence, the neo-epidermis efficiently established tissue homeostasis, the dermis underwent almost no contraction, and showed sustained epidermal coverage in vivo. Overall, the here described rapid and efficient perfusion of SVF-based skin grafts opens new perspectives for the treatment of hitherto unmet clinical needs in burn/plastic surgery and dermatology.  相似文献

背景:流式细胞仪分离法和免疫磁珠分离法对细胞活性影响较大,密度梯度离心法虽然能够获得纯度高的单核细胞,但由于多次离心可造成细胞的大量流失且对细胞活性有一定的影响使其应用值得商榷。目的:采用全骨髓贴壁法分离兔骨髓间充质干细胞进行成骨诱导分化及鉴定。方法:采用全骨髓贴壁法体外分离培养兔骨髓间充质干细胞,倒置显微镜下观察细胞形态学特征。在成骨诱导剂作用下,通过碱性磷酸酶染色试剂盒行碱性磷酸酶染色,I型胶原免疫细胞化学染色,VonKossa法及茜素红进行矿化结节染色以及电镜下检测兔骨髓间充质干细胞成骨诱导后的形态结构。结果与结论:经诱导后细胞出现与成骨细胞相似的形态学特征,碱性磷酸酶染色阳性,I型胶原免疫细胞化学染色,Von-Kossa法及茜素红矿化结节染色阳性。表明经成骨诱导剂诱导后全骨髓贴壁法体外分离纯化培养的兔骨髓间充质干细胞能向成骨细胞方向分化增殖。  相似文献

目的:研究豚鼠骨髓间充质肝细胞与纳米羟基磷灰石复合多孔陶瓷材料体外复合培养的结合程度,及构建组织工程人工听骨的可行性分析。方法:利用MSCs和其它细胞不同的生长特性,采用细胞差速贴壁法,倒置相差荧光显微镜下逐日观察各代MSCs形态及生长情况。使用流式细胞仪检测CD29、CD45、CD44进行间充质细胞表面标记物的检测。将MSCs与纳米羟基磷灰石复合多孔陶瓷复合培养,经过各个时间点的扫描电镜检测,观察MSCs与此种支架材料的复合情况。结果:经数次换液和连续传代培养,红细胞等杂质逐渐减少,可得到形态较均一的MSCs,其增殖速度和细胞活性良好。流式细胞仪CD29表达率可达92%、CD44表达率70%。MSCs在与支架材料复合培养到第3天时可稳定的生长在材料表面,第5-7天可将材料表面全部覆盖,7天后开始从材料表面脱落。结论:细胞差速贴壁法经济可靠、操作简单、易于掌握,是体外分离培养MSCs的可行的实验方法;以此法获得的豚鼠MSCs生物学形状稳定,纯度高,增殖能力强。MSCs在与支架材料复合培养结合程度高,可用来构建组织工程人工听骨,为进一步的动物实验奠定基础。  相似文献

 PURPOSE:To evaluate the clinical outcomes of tissue-engineered epithelium transplantation for severe ocular surface burns. METHODS:This was a retrospective observational case series. From October 2005 to May 2011, 19 eyes of 19 patients with grade IV to VI ocular surface burns (Dua Classification) were treated by autologous transplantation of corneal stem cells cultivated on a fibrin gel membrane, with a mean follow-up of 16.2 months (range 12-36 months). Postoperative corneal surface stability, visual acuity (VA), corneal opacity, and neovascularization were evaluated. RESULTS:No corneal perforations occurred and the entire corneal surface was free from epithelial defects in all eyes. At the final follow-up visit, VA in 17 eyes was improved after surgery, with 6 eyes achieving a VA of 20/100 or better. Corneal vascularization was significantly reduced in 17 (89.5%) eyes.Corneal opacity was also improved in 12 (63.2%) eyes. All donor eyes remained healthy. CONCLUSION:Tissue-engineered epithelium transplantation can promote rapid reepithelialization of the ocular surface, inhibit corneal neovascularization, and improve vision for patients with severe ocular surface burns.  相似文献

[目的]研制Ⅰ型胶原(collagen type Ⅰ,CI)海绵/血管内皮生长因子(vascular endothelial growth factor,VEGF)缓释生物支架材料,了解该种材料中的VEGF体外缓释效果.为骨肌腱组织工程寻求一种新型复合支架材料.[方法]从CI海绵自身的力学、孔径、紫外最大吸收光谱、孔隙率、含水率、傅立叶红外光谱及与VEGF的组织相容性等方面研究VEGF胶原缓释系统支架材料的特性,利用生物学方法将VEGF负载于CI,制备具有VEGF缓释效果的复合支架.[结果]CI海绵能够成功的将VEGF包裹起来,可以达到明显的缓释作用,与VEGF有良好的组织相容性.[结论] CI海绵有理想的孔径和合理的生物力学及与VEGF良好的生物学相容性结构性质,VEGF可以从CI - VEGF缓释材料中缓慢释放,有明显的缓释效果,是一种具有良好材料特性的组织工程材料.  相似文献

We present an injection form of tissue-engineering construct on the basis of autogenous human chondroblasts for regeneration of the cartilaginous tissue. A material meeting the requirements for the creation of the injection form of an ITC construct (injectable chondrocyte transplantation) was used as the matrix carrier. The developed construct on the basis of autogenous human chondroblasts and homogenized hemostatic gelatin sponge is now at the stage of complex experimental testing.
