Age-related cognitive decline is the major cause of concern due to its 70% more incidence than dementia cases worldwide. Moreover, aging is also the major risk factor of Alzheimer's disease (AD), associated with progressive memory loss. Approx. 13 million people will have Alzheimer-related memory decline by 2050. Learning and memory is the fundamental process of brain functions. However, the mechanism for the same is still under investigation. Thus, it is critical to understand the process of memory consolidation in the brain and extrapolate its understanding to the memory decline mechanism. Research on learning and memory has identified several molecular signatures such as Protein kinase M zeta (PKMζ), Calcium/calmodulin-dependent protein kinase II (CaMKII), Brain-derived neurotrophic factor (BDNF), cAMP-response element binding protein (CREB) and Activity-regulated cytoskeleton-associated protein (Arc) crucial for the maintenance and stabilization of long-term memory in the brain. Interestingly, memory decline in AD has also been linked to the abnormality in expressing these memory-related molecular signatures. Hence, in the present consolidated review, we explored the role of these memory-related molecular signatures in long-term memory consolidation. Additionally, the effect of amyloid-beta toxicity on these molecular signatures is discussed in detail.  相似文献

目的:探讨消化道肿瘤中同源重组修复相关基因(homologous recombination repair related gene,HRR)突变的发生情况及临床意义。方法:共92例消化道肿瘤患者,79例患者进行了血液标本HRR检测,53例患者进行了组织标本HRR检测,40例患者同时行血液和组织的HRR基因检测,收集患者基因检测结果及临床相关资料。结果:在79例患者血液标本检测中发现10例(12.6%)有临床意义HRR突变,在53例患者组织标本检测中发现9例(17.0%)有临床意义HRR突变。40例同时行血液和组织的HRR基因检测患者中常见的有临床意义HRR突变为CDK12突变4例(10.0%)、ATM突变3例(7.5%)、BRCA1突变2例(5.0%)。13例有临床意义HRR突变患者中常见共存突变为TP53突变10例(76.9%)、APC突变5例(38.5%)、PIK3CA突变4例(30.8%)。40例患者中13例患者血液和/或组织中有临床意义HRR突变,27例患者血液和组织中均无任何临床意义HRR突变且两组相比,有临床意义HRR突变组肿瘤突变负荷(tumor mutational burden,TMB)为6.17(2.24~11.52),而未携带HRR突变组TMB为0.4(0~3.75),差异有统计学意义(P<0.05)。40例患者组织检测中7例HRR有临床意义的突变,33例无HRR突变,血液检测中10例HRR有临床意义的突变,30例无HRR突变,一致性检验的Kappa值为0.333(P=0.031)。结论:携带有临床意义HRR突变的消化道肿瘤患者TMB更高,血液和组织检测HRR突变有较好的一致性。  相似文献

Zedoary tumeric (Curcumae Rhizoma, Ezhu in Chinese) has a long history of application and has great potential in the treatment of liver cancer. The anti liver cancer effect of zedoary tumeric depends on the combined action of multiple pharmacodynamic substances. In order to clarify the specific mechanism of zedoary tumeric against liver cancer, this paper first analyzes the mechanism of its single pharmacodynamic substance against liver cancer, and then verifies the joint anti liver cancer mechanism of its "pharmacodynamic group". By searching the research on the anti hepatoma effect of active components of zedoary tumeric in recent years, we found that pharmacodynamic substances, including curcumol, zedoarondiol, curcumenol, curzerenone, curdione, curcumin, germacrone, β-elemene, can act on multi-target and multi-channel to play an anti hepatoma role. For example, curcumin can regulate miR, GLO1, CD133, VEGF, YAP, LIN28B, GPR81, HCAR-1, P53 and PI3K/Akt/mTOR, HSP70/TLR4 and NF-κB. Wnt/TGF/EMT, Nrf2/Keap1, JAK/STAT and other pathways play an anti hepatoma role. Network pharmacological analysis showed that the core targets of the "pharmacodynamic group" for anti-life cancer are AKT1, EGFR, MAPK8, etc, and the core pathways are neuroactive live receiver interaction, nitrogen metabolism, HIF-1 signaling pathway, etc. At the same time, by comparing and analyzing the relationship between the specific mechanisms of pharmacodynamic substance and "pharmacodynamic group", it is found that they have great reference significance in target, pathway, biological function, determination of core pharmacodynamic components, formation of core target protein interaction, in-depth research of single pharmacodynamic substance, increasing curative effect and so on. By analyzing the internal mechanism of zedoary tumeric pharmacodynamic substance and "pharmacodynamic group" in the treatment of liver cancer, this paper intends to provide some ideas and references for the deeper pharmacological research of zedoary tumeric and the relationship between pharmacodynamic substance and "pharmacodynamic group".  相似文献

脾胃之气的充足和清气的正常升发对人体生命活动具有决定意义,饮食、劳倦、情志、外感对脾胃的损伤往往影响中焦脾胃清阳的生成及升发,脾胃病以本虚为主,多由虚致实,虚实兼有,病机关键为升降失常,脾气不升。肝对人体气机具有升宣调畅作用,脾气的升清有赖于肝气升发促进。在补中益气的同时,要补肝益气。同时,肾精气充盈,元气方能充沛,肾阳又主气化,肾精不封、肾气不固者必致肾气下陷。治疗须温肾壮阳、升阳举陷。脾胃中气不足,气机郁滞而化火,并脾之清气下流而生泄泻,相火乘其土位更加耗伤中焦清阳之气。段老师特别注重脾气生长、升发,只有清阳升发,脾气上升,元气才能充沛,阴火才会收敛潜藏。治疗时运用辛甘之药以补益中气,借升阳风药以助肝胆之用,补其中而升其阳,升浮变通,使生长之气健旺。段老师善于运用甘温之品,温补中气,升发清阳。  相似文献

The advent of immune checkpoint inhibitors has improved survival in some types of cancer and brought promising prospects to cancer immunotherapy. Despite their clinical benefits, significant off‐target toxicities resulting from the immune system activation have been observed, namely immune‐related adverse events (irAEs), which pose to clinicians a new challenge of optimal management. With steroids being the mainstay of current management of irAEs, immunosuppressive agents are especially indicated for severe or steroid‐refractory cases, based on current immunopathophysiological knowledge and on extrapolations of treatment options for primary autoimmune disorders. This review focuses on the status and recent clinical progress of immunosuppressive agents in the management of severe and steroid‐refractory irAEs.  相似文献

BackgroundPrenatal exposure to perfluoroalkyl substances (PFASs) has been associated with impaired immune and respiratory health during childhood but the evidence is inconsistent and limited for lung function. We studied the association between prenatal PFASs exposure and immune and respiratory health, including lung function, up to age 7 years in the Spanish INMA birth cohort study.MethodsWe assessed four PFASs in maternal plasma samples collected during the 1st trimester of pregnancy (years: 2003–2008): perfluorohexane sulfonate (PFHxS), perfluorooctane sulfonate (PFOS), perfluorooctanoate (PFOA), and perfluorononanoate (PFNA). Mothers reported the occurrence (yes/no) of lower respiratory tract infections, wheezing, asthma, and eczema in the previous 12 months at 1.5 and 4 years of the child (n = 1188) and at 7 years (n = 1071). At ages 4 (n = 503) and 7 (n = 992) years lung function was assessed using spirometry tests.ResultsThe most abundant PFASs were PFOS and PFOA (geometric means: 5.80 and 2.31 ng/mL, respectively). The relative risk of asthma during childhood per each doubling in PFNA concentration was 0.74 (95 CI%: 0.57, 0.96). The relative risk of eczema during childhood per every doubling in PFOS concentration was 0.86 (95 CI%: 0.75, 0.98). Higher PFOA concentrations were associated with lower forced vital capacity and lower forced expiratory volume in 1 s z-scores at 4 years [β (95 CI %): −0.17 (−0.34, −0.01) and −0.13 (−0.29, 0.03), respectively], but not at 7 years.ConclusionThis longitudinal study suggests that different PFASs may affect the developing immune and respiratory systems differently. Prenatal exposure to PFNA and PFOS may be associated with reduced risk of respiratory and immune outcomes, particularly asthma and eczema whereas exposure to PFOA may be associated with reduced lung function in young children. These mixed results need to be replicated in follow-up studies at later ages.  相似文献

[目的]探讨散聚汤加味改善宫颈癌术后放化疗患者的癌因性疲乏和免疫功能的疗效。[方法]筛选在洛阳解放军534医院行宫颈癌手术后患者共82例作为研究对象,按随机数字表法随机分为对照组和观察组各41例。对照组给予同步放疗+TP化疗方案[紫杉醇(TAX)+顺铂(DDP)]。观察组在对照组基础上给予散聚汤加味治疗,每日1剂,每日2次,连续服用12周。比较两组患者癌因性疲乏评分、毒性反应、T淋巴细胞亚群水平以及血清免疫球蛋白(IgA、IgG、IgM)水平。[结果]治疗12周后,观察组患者的癌因性疲乏(情绪、感觉、行为、认知)评分显著低于对照组(P0.01);观察组患者的消化道反应、膀胱刺激发生率明显少于对照组(P0.05);治疗12周后,观察组患者CD3+、CD3+CD4+以及IgA、IgG、IgM水平显著高于对照组,CD3+CD8+明显低于对照组(P0.01)。[结论]散聚汤加味能改善宫颈癌术后放化疗患者的癌因性疲乏,增强免疫功能,且可降低毒副反应。  相似文献
