Prenatal exposure to polybrominated diphenyl ethers (PBDEs) have been reported to impair executive function in children, but little is known whether childhood PBDE exposures play a role. Using the Health Outcomes and Measures of the Environment (HOME) Study, a prospective birth cohort in the greater Cincinnati area, we investigated the association between repeated measures of PBDEs during childhood and executive function at 8 years in 208 children and whether effect modification by child sex was present. We used child serum collected at 1, 2, 3, 5, and 8 years to measure PBDEs. The Behavior Rating Inventory of Executive Function was completed by parents to assess executive function at 8 years. We used multiple informant models to examine childhood PBDEs during several exposure windows. Null associations were observed between early childhood PBDEs and executive function. However, we observed significant adverse associations between a 10-fold increase in concurrent concentrations of BDE-28 (β = 4.6, 95% CI 0.5, 8.7) and BDE-153 (β = 4.8, 95% CI 0.8, 8.8) with behavioral regulation. In addition, PBDEs at 8 years were significantly associated with poorer emotional and impulse control. No associations were noted between childhood PBDEs and metacognition or global executive function. However, child sex significantly modified the associations, with significantly poorer executive function among males with higher concurrent BDE-153, and null associations in females. Our study findings suggest that concurrent PBDE exposures during childhood may be associated with poorer executive function, specifically behavior regulation. Males may also be more sensitive to adverse associations of concurrent PBDEs on executive function.  相似文献

Polychlorinated biphenyls (PCBs) are typical persistent organic pollutants that can interfere with multiple organ systems of humans. Previously, we concluded that persistent exposure to low doses of PCB118 could severely damage the thyroidal structure, dramatically decrease the concentration of serum thyroid hormones and inhibit the pivotal gene expressions such as sodium/iodide symporter (NIS) and thyroglobulin (Tg). To explore the molecular mechanisms of thyrocyte dysfunction induced by 2,3′,4,4′,5‐pentachlorobiphenyl (PCB118), monolayer cultured human thyroid epithelial cells (HTECs) were treated with PCB118 or dimethyl sulfoxide (DMSO) as a control. Our results indicated that relatively higher concentrations of PCB118 could induce a loss in the viability of HTEC. In cultures with concentrations of PCB118 from 0.025 to 25 nM, which did not affect cell viability or apoptosis, concentrations of Tg and thyroxine (T4) were significantly decreased compared with those in the controls. In addition, mRNA and protein levels of Akt were increased significantly in the PCB118‐treated groups, whereas FoxO3a expression did not show particular variation. Furthermore, exposure to PCB118 was associated with a significant increase of the protein levels of p‐Akt and p‐FoxO3a, and these effects were blocked by LY294002. In contrast, mRNA and protein expression levels of NIS were decreased significantly, and this effect was blocked by LY294002. Unlike control cells, a cytoplasmic shift of FoxO3a was observed in the PCB118‐treated group. Our research suggests that PCB118 may induce thyrocyte dysfunction through the Akt/FoxO3a/NIS signalling pathway, which provides potential new insights for finding interventions to counteract the damage to the human body caused by PCBs. Although the contamination of wooden art objects with pesticides is well known, to the authors' knowledge no attempt has yet been made to investigate the eventual presence of other toxic compounds that have been produced during the degradation of pesticides or that may be present in the technical formulations. Here, the authors report on the presence of polychlorinated biphenyls (PCBs) and polychlorinated terphenyls (PCTs) in scrapings from wooden antique art objects, namely printing blocks, sculptures, and masks. These antiques belong to 2 fine art museums in Belgium—Antwerp's Ethnographic Museum and the Plantin-Moretus Museum. It is documented that these art objects were treated with pesticides in the 1950s. In addition, 2-heptachlorodibenzo-p-dioxin (HpCDD) isomers and octachlorodibenzo-p-dioxin (OCDD) were also identified. The presence of these toxic compounds in these antiques requires a better understanding of safety for the persons (conservators, museum employees, restorers, and visitors) coming in contact with these objects.  相似文献

Primary liver cancer or hepatocellular carcinoma (HCC) is one of the most frequent tumors representing the fifth commonest malignancy worldwide and the third cause of mortality from cancer. Currently, the treatments for HCC are not so effective and new strategies are needed for its fight. Chemoprevention, the use of natural or synthetic chemical agents to reverse, suppress or prevent carcinogenesis is considered an important way for confronting HCC. Many of the chemopreventive agents are phytochemicals, namely non-nutritive plant chemicals with protective or disease preventive properties. In this review, we focus on plant polyphenols, one of the most important classes of phytochemicals, their chemopreventive properties against HCC and discuss the molecular mechanisms accounting for this activity.  相似文献

This study deals with the levels of toxic polychlorinated dibenzo‐p‐dioxin and furan congeners (PCDD/Fs) in the livers of piglets affected by infectious diseases using isotope dilution high‐resolution gas chromatography/high‐resolution mass spectrometry (HRGC/HRMS). Seventeen toxic congeners in the liver samples infected with bacterial and viral diseases were compared. For porcine reproductive and respiratory syndrome virus (PRRSV) samples, the North American‐ and European‐type PRRS diseases were observed. This study shows that there are significantly different levels of PCDD/Fs, present, which vary according to the types of diseases as evidenced by our analysis of the piglet liver samples.  相似文献

The aim of this study was to evaluate the association between persistent organic pollutants (POPs) and thyroid hormones in an aging population. Forty-eight women and 66 men, aged 55–74 years and living in upper Hudson River communities completed a questionnaire and provided blood specimens. Serum was analyzed for thyrotropin (thyroid stimulating hormone, TSH), free (fT4) and total thyroxine (T4), total triiodothyronine (T3), and for POPs. POPs included 39 polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane (DDT) and dichlorodiphenyldichloroethylene (DDE) determined by gas chromatography with electron capture detection (GC–ECD), and nine polybrominated diphenyl ethers (PBDEs) determined by high-resolution gas chromatography with high-resolution mass spectrometry detection (HRGC–HRMS). Multivariable linear regression analysis was used to evaluate associations between thyroid hormones and sums of POPs, adjusted for covariates and stratified by sex. Effects were expressed as differences in thyroid hormone levels associated with a doubling in the level of exposure. Among women, DDT + DDE increased T4 by 0.34 μg/dL (P = 0.04) and T3 by 2.78 ng/dL (P = 0.05). Also in women, sums of PCBs in conjunction with PBDEs elicited increases of 24.39–80.85 ng/dL T3 (P < 0.05), and sums of PCBs in conjunction with DDT + DDE elicited increases of 0.18–0.31 μg/dL T4 (P < 0.05). For men estrogenic PCBs were associated with a 19.82 ng/dL T3 decrease (P = 0.003), and the sum of estrogenic PCBs in conjunction with DDT + DDE elicited an 18.02 ng/dL T3 decrease (P = 0.04). Given age-related declines in physiologic reserve, the influence of POPs on thyroid hormones in aging populations may have clinical implications and merits further investigation.  相似文献

[目的]建立新生儿脐带血中多氯联苯(PCBs)的分析方法,并对电子垃圾拆解区新生儿PCBs暴露水平进行评估。[方法]血清样品经正己烷和甲基叔丁基醚混合溶液(1:1)萃取,经干燥得到脂肪并称取其质量。脂肪经正己烷溶解、硫酸净化、酸性硅胶柱净化及浓缩定容后,利用气相色谱-质谱联用仪检测分析PCBs含量。采用EI源电离方式与选择离子检测法(SIM),以外标法进行定量。[结果]加标回收率76.0%~113.0%;方法检出限0.01~0.12ng/g;电子垃圾拆解区新生儿体内28种PCBs的总浓度为338.56 ng/g lipid。[结论该方法检测新生儿脐带血中28种PCBs的同系物,灵敏度高、重复性好、回收率良好,可用于相关样品的检测。广东省汕头贵屿镇电子垃圾拆解区新生儿PCBs已处于较高暴露水平,值得重视。  相似文献
