IntroductionThis Consolidated Standards of Reporting Trials randomized clinical trial investigated T helper (Th1, Th2, Th9, Th17, and Tfh) and regulatory T (Treg) cell–type cytokines and their networks in apical periodontitis (AP). We also assessed the effects of calcium hydroxide [Ca(OH)2] intracanal medications (ICMs) on helper T and Treg cell–type cytokines.MethodsTwenty teeth with primary endodontic infection and apical periodontitis were randomly divided into two groups: Ca(OH)2 + saline solution (n = 10) and Ca (OH)2 + 2% chlorhexidine-gel (n = 10). Samples were collected from the periradicular tissue fluid (PTF) before (PTFs1) and after 14 days of ICMs (PTFs2). The Human High Sensitivity T Cell Panel was used to quantify target T-helper (Th)1: interelukin (IL)-2, IL-12, and interferon-gamma (IFN-γ); Th2: IL-4, IL-5, and IL-13; Th9: IL-9; Th17: IL-17; T follicular helper cells (Tfh): IL-21; and Treg-cell-type cytokine: IL-10.ResultsTh1-type cytokines were higher than Th2-type ones, at PTFs1. Positive (+) associations were found among all Th1-type cytokines and all Th2-type cytokines. There were negative (-) correlations between all Th1- and Th2-type cytokines. Size of radiolucent lesions and symptoms (tenderness to percussion and/or pain on palpation) were positively correlated with Th1-type cytokines, IL-17, and IL-21 but negatively correlated with Th2-type cytokines and IL-10 (all, P < .001). Both ICMs increased Th2-type cytokines and IL-10 (P < .05) and decreased Th1-type cytokines, IL-17, and IL-21 (P < .05), with no differences among them (P > .05).ConclusionsComplex T-cell cytokine networks are involved in AP. Both Ca(OH)2 ICMs effectively increased IL-4, IL-5, IL-10, and IL-13 and lowered IL-2, IL-12, IL-17, IL-21, and IFN-γ.  相似文献

The aim of this study was to gain information about the effect of thermal treatment of calcium silicate-based sealers. BioRoot RCS (BR), Total Fill BC Sealer (TFBC), and Total Fill BC Sealer HiFlow (TFHF) were exposed to thermal treatment at 37 °C, 47 °C, 57 °C, 67 °C, 77 °C, 87 °C and 97 °C for 30 s. Heat treatment at 97 °C was performed for 60 and 180 s to simulate inappropriate application of warm obturation techniques. Thereafter, specimens were cooled to 37 °C and physical properties (setting time/flow/film thickness according to ISO 6876) were evaluated. Chemical properties (Fourier-transform infrared spectroscopy) were assessed after incubation of the specimens in an incubator at 37 °C and 100% humidity for 8 weeks. Statistical analysis of physical properties was performed using the Kruskal-Wallis-Test (P = 0.05). The setting time, flow, and film thickness of TFBC and TFHF were not relevantly influenced by thermal treatment. Setting time of BR decreased slightly when temperature of heat application increased from 37 °C to 77 °C (P < 0.05). Further heat treatment of BR above 77 °C led to an immediate setting. FT-IR spectroscopy did not reveal any chemical changes for either sealers. Thermal treatment did not lead to any substantial chemical changes at all temperature levels, while physical properties of BR were compromised by heating. TFBC and TFHF can be considered suitable for warm obturation techniques.  相似文献

材料学的发展对牙髓疾病的治疗有重要意义,聚乳酸?羟基乙酸共聚物[poly(lactic?co?glycolic acid),PLGA]是一种被广泛应用于生物医用材料制备的有机高分子化合物。近年来,作为载药/分子系统和组织再生支架在牙髓疾病治疗中展现出应用前景,本文将对PLGA在牙髓疾病治疗中的应用作一综述,为其进一步开发利用提供参考。文献复习结果表明,PLGA作为药物/分子输送系统主要应用于盖髓材料、根管消毒剂和根尖诱导成形剂的改良。PLGA改良盖髓材料能延长药物作用时间、降低毒性;改良根管消毒剂能实现药物缓释,使药物深入更细微的结构,与致病菌有更广泛的接触;改良根尖诱导成形剂能为根尖诱导提供更便捷的给药方式。PLGA作为组织工程支架主要应用于牙髓再生的研究,通过PLGA物理性能、作用环境的不断优化为种子细胞提供更适宜增殖分化的微环境。如何合理利用PLGA的优势,研制出更适宜根管内应用的材料,还需要进一步的研究。  相似文献

土家族医药学是我国少数民族医药体系中的一个重要分支学科,它植根于湖南、湖北、贵州、重庆等广大土家族地区,有其自身的民族药物理论认识、用药特色与经验,特别是关于用药禁忌方面,形成了土家族传统药配伍禁忌的生药十三反、十四反原则,对数以万计中草药的配伍原则及使用上有一定指导作用。文章针对土家族医药工作者们常遵循的此配伍禁忌,对药物的来源进行考证,并结合性味、功效记载,从药性特别是四性、毒副作用分析等方面对土家医的生药十三反、十四反之相反理论基础作出初步探讨,以期为完善中药的配伍禁忌理论提供借鉴,为传统民族医药的进一步开发与应用研究提供一些切入思路。  相似文献

Antiseptics used in endodontics for disinfection purposes include root canal dressings and irrigants. Osmotic shock is known to cause the alteration of microbial cell viability and might have a role in the mechanism of action of root canal antiseptics. The aim of this review was to determine the role of osmolarity on the performance of antiseptics in root canal treatment. A literature search using the Medline electronic database was conducted up to 30 May 2013 using the following search terms and combinations: ‘osmolarity AND root canal or endodontic or antiseptic or irrigation or irrigant or medication or dressing or biofilm; osmolality AND root canal or endodontic or antiseptic or irrigation or irrigant or medication or dressing or biofilm; osmotic AND root canal or endodontic or antiseptic or irrigation or irrigant or medication or dressing or biofilm; osmosis AND root canal or endodontic or antiseptic or irrigation or irrigant or medication or dressing or biofilm; sodium chloride AND root canal or endodontic or antiseptic or irrigation or irrigant or medication or dressing or biofilm’. Publications were included if the effects of osmolarity on the clinical performance of antiseptics in root canal treatment were stated, if preparations with different osmolarities values were compared and if they were published in English. A hand search of articles published online, ‘in press’ and ‘early view’, and in the reference list of the included papers was carried out following the same criteria. A total of 3274 publications were identified using the database, and three were included in the review. The evidence available in endodontics suggests a possible role for hyperosmotic root canal medicaments as disinfectants, and that there is no influence of osmolarity on the tissue dissolution capacity of sodium hypochlorite. There are insufficient data to obtain a sound conclusion regarding the role of hypo‐osmosis in root canal disinfection, or osmosis in any further desirable ability.  相似文献

Introduction

The aim of this study was to characterize the response of mouse subcutaneous tissue to triple antibiotic paste (TAP) using conventional light microscopy and real-time PCR (qRT-PCR).

Methods

Polyethylene tubes containing TAP or calcium hydroxide (CH) (ie, the control group) were implanted in mouse subcutaneous tissue. Animals that received empty tubes or no tubes were used as additional controls. After periods of 7, 21, and 63 days postimplantation, the specimens were removed and subjected to histologic processing. The number of inflammatory cells and vessels, vessel areas, vascular density, and relative percentage of collagen were evaluated. Gene expression of proinflammatory (interleukin-1 beta, tumor necrosis factor alpha, and interleukin 17) and anti-inflammatory (transforming growth factor beta) cytokines and angiogenic factors (vascular endothelial growth factor and hypoxia-inducible factor-1 alpha) was quantified by 7 and 21 days postimplantation. Results were analyzed using the Student t test, analysis of variance, and the Tukey test (α = 0.05).

Results

TAP induced an exuberant inflammatory and angiogenic response, with higher numbers of inflammatory cells, higher vascular area and density, and lower relative percentage of collagen compared with CH. In general, the expression of genes involved in inflammation and angiogenesis was higher in the TAP group compared with animals that received CH or empty tubes.

Conclusions

The response of mouse subcutaneous tissue to TAP was characterized by exuberant and persistent inflammatory and angiogenic responses with no repair and high gene expression of biomarkers associated with inflammation and angiogenesis.  相似文献

This study investigated the effect of ultrasonic activation of intracanal‐heated sodium hypochlorite (NaOCl) on its dentinal tubular penetration and root canal cleanliness in vitro. In experiment 1, mandibular premolars were randomly allocated to three groups (n = 8): group A, ultrasonic activation; group B, ultrasonic activation of intracanal‐heated NaOCl and group C, syringe‐and‐needle irrigation. Penetration of the fluorescent‐labelled NaOCl was investigated using light microscopy. In experiment 2, mandibular premolars were randomly allocated to group B or C (n = 10), for histological analysis of the remaining pulp tissue and debris. Data were statistically analysed using Kruskal–Wallis and Mann–Whitney tests (P = 0.05). The highest penetration of NaOCl was observed in group B, followed by group A (P < 0.05). Group B showed significantly less amount of debris than group C (P < 0.05). Dentinal tubule penetration of NaOCl and root canal cleanliness were significantly improved by ultrasonic activation of intracanal‐heated NaOCl.  相似文献

目的:观察腰硬联合麻醉下剖宫产术中体位变化对产妇血流动力学的影响。方法将40例ASAⅠ~Ⅱ级拟行剖宫产术的产妇分为A、B两组,每组20例,于剖宫产术中分别采用平卧位和左侧斜卧15°位。分别于入室后5 min、腰硬联合麻醉时侧卧位后3 min、麻醉后3 min、切皮即刻、胎儿娩出后3 min、手术结束时点记录产妇心输出量、每搏量、心率、平均动脉压、总外周阻力、胸腔液体含量。结果两组产妇总输液量、升压药用量、心率、胸腔液体含量无明显差异;心输出量、每搏量组内各时比较点差异有统计学意义(P<0.05),但组间比较差异无统计学意义(P>0.05);麻醉后A组血压、总外周阻力下降较B组明显(P<0.05)。结论腰硬联合麻醉剖宫产术围术期血流动力学波动较大,主要发生在麻醉后及胎儿娩出前后,需要及时准确的监测以指导治疗。术中采取左侧斜卧位,配合恰当的容量治疗及麻醉管理,可减少低血压的发生。  相似文献
