Toxic tau: structural origins of tau aggregation in Alzheimer's disease

Alzheimer’s disease is characterized by the extracellular accumulation of the amyloidβin the form of amyloid plaques and the intracellular deposition of the microtubule-associated protein tau in the form of neurofibrillary tangles.Most of the Alzheimer’s drugs targeting amyloidβhave been failed in clinical trials.Particularly,tau pathology connects greatly in the pathogenesis of Alzheimer’s disease.Tau protein enhances the stabilization of microtubules that leads to the appropriate function of the neuron.Changes in the quantity or the conformation of tau protein could affect its function as a microtubules stabilizer and some of the processes wherein it is involved.The molecular mechanisms leading to the accumulation of tau are principally signified by numerous posttranslational modifications that change its conformation and structural state.Therefore,aberrant phosphorylation,as well as truncation of tau protein,has come into focus as significant mechanisms that make tau protein in a pathological entity.Furthermore,the shape-shifting nature of tau advocates to comprehend the progression of Alzheimer’s disease precisely.In this review,we emphasize the recent studies about the toxic and shape-shifting nature of tau in the pathogenesis of Alzheimer’s disease.     

中药栀子本草考证

栀子临床应用广泛,且历史悠久,为更好地利用栀子资源,通过梳理历代文献与现代研究,对栀子名称、品种、产地、炮制、功效等方面进行考证,发现栀子命名主要以形态、色泽为依据,一定程度上反映了栀子的特征,并且存在与水栀子混用"黄栀"之名的情况。古代文献中记载的栀子产地最早为南阳,但产量不高,受人口迁移以及生产力发展的影响,栀子的产地主要转移到南方,且以南方产者为佳。在历代本草文献中记载入药用的栀子为山栀子,水栀子不堪入药,但在《伤寒论》等古代方书中有"肥栀子""大栀子"入药用的记载,通过后世对相应方剂的记载,确定二者应为水栀子,为水栀子的临床应用提供了有力依据。之所以与本草文献记载有所差异,应与《雷公炮炙论》和《伤寒论》的年代不同,栀子的产量及采收能力有所差距,且本草书重规范,择优入药,方书注重实用等特点有关,并且根据现代实验研究表明山栀子与水栀子化学成分组成、含量相似,具有相同的利胆作用。此外,系统整理了历代本草与方书中栀子的炮制方法和功效,并进行了分析,为栀子属资源的进一步研究与利用提供了参考和借鉴。     

密盖息治疗49例恶性骨转移瘤疗效观察

目的:观察密盖息(鲑鱼降钙素)的治疗骨转移瘤疼痛和血清钙的临床疗效。方法:单用组用密盖息100IU,肌肉注射,每日2次,共7d。联合化疗组化疗后加密盖息100IU肌肉注射,每日2次,每次7d。治疗结束进行疗效评价。结果:单用组骨痛完全缓解66.7%(12/18)。联合化疗组骨痛完全缓解率87.1%(27/31)。单用组血钙降至正常,占77.8%(14/18),联合化疗组血钙完全降至正常占100%(31/31)。副作用是暂时性的心动过速、皮肤潮红、头晕、恶心呕吐,无需特殊处理,自行缓解。结论:密盖息治疗骨转移瘤有止痛、降低血钙作用良好,副作用小,可以在临床上安全使用。

适应性反应是近年医学研究的热点问题之一,利用适应性反应已作为疾病防治的新战略,其核心和精髓是充分调动和激发机体内源性保护机制,增加机体适应性和抗损伤能力,维持机体自稳态.适应性反应的研究表明了机体自身存在着很强的内源性保护机制,其本质是适应性反应诱导了内源性保护物质(保护性蛋白等)的产生,而用药物等途径则可模拟或增强适应性保护作用.适应性反应与中医学的天人相应、正气学说、扶正固本、藏象学说、治未病思想、适应原中药、针灸经络等理论有诸多联系,对中医药研究有一定的启示,对于揭示中医学的科学内涵和实质、发展和弘扬中医理论及促进中西医结合均具有一定参考意义.     

The nature of breast dense core granules: uranaffin reactivity

In this study breast tissue that contained ultrastructural dense core granules has been examined from 24 patients. The possible neuroendocrine characteristics of the granules were studied using the uranaffin reaction and an ultrastructural argyrophil stain. The tissue included 15 breast carcinomas and nine benign samples. Two types of breast dense core granules can be distinguished on the basis of intracellular distribution patterns and histochemical reactivity. Type I dense core granules are the most common and they were found subluminally distributed in non-pathological tissues and benign lesions as well as carcinomas. They were uranaffin-negative and are unlikely to be of endocrine nature. Type II dense core granules were found only in four carcinomas, where they occurred abundantly throughout the cytoplasm. These granules were uranaffin-positive and argyrophilic and thus exhibited characteristics consistent with neuroendocrine structure.

主动吸入药物烟对大鼠一氧化氮的影响

目的　探讨主动吸入药物烟对大鼠NO的影响。方法　制备大鼠主动吸烟模型 ,采用硝酸还原酶法测定各组大鼠血清、肺、心肌及脑组织的NO含量。结果　非药物烟组大鼠血清、肺、心肌及脑组织中NO含量明显低于非给烟对照组 ,药物烟高、中剂量组NO含量明显高于非药物烟组 ,接近非给烟对照组。结论　吸烟对大鼠血液、肺、心肌及脑组织有不同程度的损伤作用 ,使NO含量降低 ;药物烟具有一定的保护和防止NO含量降低的作用。
