目的 开发满足ISO15189认可要求的临床实验室试剂管理系统,为试剂的精细化管理提供对策。方法 本系统采用B/S系统架构,软件功能由JAVA语言实现,MySQL 5.7为应用数据库,Redis 3.2.100为缓存数据库。软件后台底层架构使用SpringMVC+SpringJDBC架构技术,通过SnakerFlow进行流程控制管理。结果 建立了临床实验室标准化试剂管理系统,实现了供应商管理、物资管理、采购管理、库存管理、预警提醒、温湿度实时监控、库存盘点等功能。结论 该系统不仅满足试剂使用的各种需求,而且紧扣ISO15189认可条款,为试剂的标准化管理提供高效的解决方案。  相似文献

目的:建立注射用三磷酸胞苷二钠(CTP-Na2)有关物质检验方法。方法:采用高效液相色谱法,以C18为填充柱,以含反相离子对试剂的磷酸盐缓冲液为流动相[0.02 mol·L^-1磷酸盐缓冲液-乙腈(90∶10)];流速:1.0 mL·min^-1;检测波长:271 nm;柱温:35℃,采用校正因子计算有关物质。结果:CTP-Na 2在0.093~1.860 mg·mL^-1范围内,溶液的浓度与峰面积的线性关系良好,r=0.9997;平均回收率为99.6%(RSD=1.5%,n=9),CTP-Na2对照品溶液在25 h内的稳定性良好(RSD=0.7%);CTP-Na2及主要降解杂质二磷酸胞苷二钠(CDP-Na2)、单磷酸胞苷二钠(CMP-Na2)的方法定量限分别为4.1、2.1、2.8 ng,检出限分别为0.82、0.16、0.22 ng。结论:现行质量标准中有关物质项存在诸多问题,新建的检测方法操作简便、灵敏度高,可用于质量控制。  相似文献

目的:建立地黄不同炮制品的薄层色谱鉴别方法。方法:以D-果糖、梓醇、蔗糖、棉子糖、水苏糖、蜜二糖及甘露三糖为对照品,考察提取溶剂(水,20%甲醇,50%甲醇,80%甲醇),展开剂(正丁醇-甲醇-三氯甲烷-冰乙酸-水,乙酸乙酯-吡啶-冰乙酸-水,正丁醇-冰乙酸-水),显色剂(苯胺-二苯胺-磷酸溶液,茚三酮溶液),点样量(2,4,6μL),检视条件(日光,日光底部灯,365 nm和254 nm)对薄层色谱分析的影响,确定生地黄及熟地黄饮片的供试品溶液制备方法和最佳薄层色谱条件。结果:采用薄层色谱高效硅胶G板,以正丁醇-甲醇-三氯甲烷-冰乙酸-水(13∶5∶5∶1∶2)为展开剂展开,喷以苯胺-二苯胺-磷酸溶液,110℃下加热显色,于日光底部灯下进行检视,所得地黄不同炮制品薄层色谱分离效果和显色效果较佳,斑点清晰且特征性好。结论:该薄层色谱鉴别方法操作简便易行,定性特征明显、结果直观,可有效鉴别地黄不同炮制品,并可为熟地黄的炮制终点确定提供实验依据。  相似文献

BackgroundThe role of preoperative laboratory values for risk stratification following joint arthroplasty is currently ambiguous. In order to improve upon existing risk stratification within joint arthroplasty, this study sought to define novel phenotypes of total hip or total knee arthroplasty patients based entirely on preoperative laboratory measures. These phenotypes (“clusters”) were compared to elucidate statistically and clinically significant differences in outcomes.MethodsA total of 134,252 patients were gathered from the National Surgical Quality Improvement Program database between 2005 and 2015. “K-means” with 3 clusters was applied using 9 preoperative laboratory values: sodium, blood urea nitrogen (BUN), creatinine, albumin, bilirubin, white blood cell count, hematocrit, platelet count, and international normalized ratio of prothrombin values (INR). Outcome measures included 30-day readmissions, severe adverse events, and discharge to nonhome.ResultsCluster 2 was characterized by elevated preoperative BUN, creatinine, and INR and demonstrated almost twice the rate of adverse events (3.52% vs 2.20% and 2.22%), 30-day readmissions (6.39% vs 3.31% and 3.71%), and discharge to nonhome (47.97% vs 30.50% and 35.85%). Cluster 3 was characterized by a slightly higher risk of discharge to nonhome than cluster 1 and was overwhelmingly female (79.5% female, 35.8% discharge to nonhome). Cluster 1 represents the lowest-risk subgroup, experiencing the lowest rates of readmissions, adverse events, and discharge to nonhome.ConclusionPreoperative laboratory values, namely BUN, creatinine, and INR, are useful in identifying patients at risk of adverse outcomes. This analysis supports the existing surgical literature pushing for preoperative hydration as a targeted intervention to expedite recovery.  相似文献

African swine fever (ASF) is a devastating disease, which is causing huge economic losses in China. Therefore, it is urgent to provide a rapid, highly specific and sensitive diagnostic method for the detection of African swine fever virus (ASFV), the ASF infectious agent. In this study, a novel quantitative real‐time polymerase chain reaction (qPCR) assay with lyophilized powder reagents (LPR), targeting the major structural protein p72 gene, was established for the detection of ASFV. This assay had many advantages, such as saving time and money, good sensitivity and repeatability. The sensitivity of this assay was 100 copies/μl of ASFV plasmid templates, and the assay showed 10‐fold greater sensitivity than a qPCR assay recommended by OIE. Furthermore, specificity analysis showed that qPCR with LPR for ASFV had no cross‐reactivity with other important swine pathogens. In clinical diagnoses of 218 blood samples of domestic pigs in China, the positive rate of the diagnosis of ASFV by qPCR with the LPR and commercial kit reached 80.73% (176/218) and 76.61% (167/218) respectively. The coincidence rate between the two assays is 92.20% (201/218), and kappa value is 0.768 (p < .0001) by SPSS analysis. The overall agreement between the two assays was 95.87% (209/218). Further Pearson correlation and linear regression analysis showed a significant correlation between the two assays with an R2 value of 0.9438. The entire procedure, from specimen processing to result reporting, can be completed within 2 hr. Our results demonstrated that the qPCR‐LPR assay is a good laboratory diagnostic tool for sensitive and efficient detection of ASFV.  相似文献

文题释义:玻璃化冷冻保存:是利用这些高浓度的低温保护剂组合成玻璃化冻存液,通过与水分子发生强烈的水合作用,增加溶液黏性,降低冰晶形成速度,从而使细胞在快速降温或复温过程中得以保护。 玻璃化:对于非晶高分子,当高分子通过降温从高弹态转变为玻璃态,或者通过升温从玻璃态转变为高弹态的过程称之为玻璃化转变,发生玻璃化转变的温度叫玻璃化转变温度。对于结晶高分子,玻璃化转变是指其非晶部分所发生的由高弹态向玻璃态(或者玻璃态向高弹态)的转变。因此,玻璃化转变是高分子中普遍存在的现象。但是玻璃化转变现象并不局限于高分子,一些小分子化合物也存在玻璃化转变。 背景:玻璃化冷冻保存是一种应用前途广阔的低温冷冻方法,通过使用高浓度的玻璃化冻存试剂将生物材料进行玻璃态转变,从而实现活性保存。 目的:就玻璃化冻存的生物学原理及玻璃化冻存试剂的分类,卵巢、皮肤与角膜等医学组织标本的玻璃化冻存进行综述。 方法:以“tissue;vitrification;cryopreservation”为英文检索词;“组织;玻璃化;冷冻保存”为中文检索词,检索1994年1月至2019年10月 PubMed 数据库及万方医学网相关文献。按照纳入与排除标准筛选后,对最终纳入的45篇文献进行归纳总结。 结果与结论:玻璃化冻存可以防止细胞内外冰晶形成,避免了冰晶给细胞带来的多种损伤,有效保留了细胞的生物活性与基本功能。玻璃化冻存试剂主要分为渗透性和非渗透性2种,其操作简便、高效,唯一的缺点是高浓度的冻存试剂对细胞具有一定的毒性损伤。为了降低对组织整体损伤风险,可以混合使用多种低毒冻存试剂。目前玻璃化冻存技术已经成功应用于多种细胞,但组织冻存的技术难题尚未完全解决。 ORCID: 0000-0003-0140-9935(张源) 中国组织工程研究杂志出版内容重点:组织构建;骨细胞;软骨细胞;细胞培养;成纤维细胞;血管内皮细胞;骨质疏松;组织工程  相似文献

Objective:To evaluate the effects of diabetic ketoacidosis (DKA) on neurocognitive functions in children and adolescents presenting with new-onset type 1 diabetes.Methods:Newly diagnosed patients were divided into two groups: those with DKA and those without DKA (non-DKA). Following metabolic stabilization, the patients took a mini-mental status exam prior to undergoing a baseline battery of cognitive tests that evaluated visual and verbal cognitive tasks. Follow-up testing was performed 8-12 weeks after diagnosis. Patients completed an IQ test at follow-up.Results:There was no statistical difference between the DKA and non-DKA groups neither in alertness at baseline testing nor in an IQ test at follow-up. The DKA group had significantly lower baseline scores than the non-DKA group for the visual cognitive tasks of design recognition, design memory and the composite visual memory index (VMI). At follow-up, Design Recognition remained statistically lower in the DKA group, but the design memory and the VMI tasks returned to statistical parity between the two groups. No significant differences were found in verbal cognitive tasks at baseline or follow-up between the two groups. Direct correlations were present for the admission CO2 and the visual cognitive tasks of VMI, design memory and design recognition. Direct correlations were also present for admission pH and VMI, design memory and picture memory.Conclusion:Pediatric patients presenting with newly diagnosed type 1 diabetes and severe but uncomplicated DKA showed a definite trend for lower cognitive functioning when compared to the age-matched patients without DKA.  相似文献

Purpose: Islamic Ramadan is the month of fasting, in which intake of food and drink is restricted from sunrise until sunset. The objective of the present study was to find out the effects of religious fasting on posterior ocular structures.

Materials and methods: In this prospective study, 34 eyes of 34 healthy volunteers with a mean age of 34.09?±?7.20?years were enrolled. Volunteers with any systemic disorder and eyes with pathology or previous surgery were excluded. One week before Ramadan (non-fasting period) and during Ramadan (fasting period) at the same hours (at 08:00 and 16:00?h), choroidal, macular, and retinal nerve fibre layer (RNFL) thicknesses were measured by spectral domain optical coherence tomography. Results were compared using paired sample t-test, and a p value <0.05 was accepted as statistically significant.

Results: The comparison of 16:00-h measurements significantly revealed lower values during fasting period when compared non-fasting period for choroidal thickness (non-fasting and fasting, respectively; subfoveal: 299.26?±?41.3 and 280.03?±?38.75 p?p?p?=?0.001) and paracentral macular thickness (superior: p?=?0.002, inferior: p?=?0.010, temporal: p?=?0.013, and nasal: p?=?0.016). By contrast, no significant differences were found in the central macular thickness between the fasting and non-fasting periods (p?=?0.735). Also, no statistically significant difference was noted for RNFL thickness at the different periods and time points.

Conclusion: Our results reveal that Islamic religious fasting is associated with statistically significant alterations in choroidal and paracentral macular thickness in healthy volunteers. However, more detailed investigations should be designed to evaluate whether fasting has a pivotal influence on pathological conditions.  相似文献

The purpose of this study was to investigate the phase behavior of cyclophosphamide (CPA) during various stages of lyophilization, with special emphasis on obtaining crystalline CPA monohydrate (CPA-MH) in the lyophilized product. Subambient differential scanning calorimetry and low-temperature X-ray diffractometry (LTXRD) were used to study the phase behavior of CPA solution (3.7% w/v). In situ lyophilization in LTXRD chamber was used to monitor the phase transitions occurring during the drying stages. Finally, the implications of these findings were confirmed by freeze-drying the aqueous solution in a laboratory-scale freeze-dryer. The results suggested that CPA remains amorphous during freeze concentration, with a Tg' of ?50°C. However, its crystallization as CPA-MH can be induced by annealing the frozen solution between ?5°C and ?10°C. In situ lyophilization in LTXRD showed that the CPA-MH crystallized during annealing, rapidly dehydrated during primary drying, thereby causing structural collapse. The dehydration of CPA-MH can be prevented by lowering the escaping tendency of water molecules from the crystal lattice of CPA-MH by maintaining the chamber pressure to 300, 400, or 500 mTorr. This study highlights the relationship of process parameters used during lyophilization with the solid form of lyophilized CPA.  相似文献
