BACKGROUND: Blood component irradiation is an accepted method of preventing transfusion-associated GVHD. Previous publications have largely focused on the technical aspects of the irradiation process itself, but relatively little attention has been paid to the details associated with the implementation of a blood irradiation program at the level of a community cancer center. STUDY DESIGN AND METHODS: An observational study was performed, detailing the specific operational, documentation, and quality assurance measures employed in providing a blood component-irradiation service within the institutional context of a community cancer center. RESULTS: The Montgomery Cancer Center irradiated 589 units of blood components in 1998 and 1999 to provide a local blood bank with an alternative for procurement of irradiated blood components while complying with applicable quality assurance and regulatory requirements. CONCLUSION: Blood component irradiation is within the scope of most well-equipped and adequately staffed community cancer centers. Establishment of a blood component irradiation program requires scrupulous physics and dosimetry support, both to ensure the quality of the irradiated component and to satisfy governmental agency regulatory requirements.  相似文献

Microcrystalline cellulose II (MCC II) – a polymorph of commonly used MCC I – was introduced as new pelletization aid in wet-extrusion/spheronization leading to fast disintegrating pellets. Previous investigations suggested that pellet properties were influenced by the fraction of MCC II. Furthermore, it is unknown whether the storage conditions can affect the disintegration behavior. Therefore, the effects of MCC II fraction and the storage conditions on several pellet properties were investigated.MCC II-based pellets were prepared of pure MCC II or binary mixtures containing 10–50% (steps of 10%) MCC II as pelletization aid and theophylline, chloramphenicol or lactose. The pellets were characterized by their aspect ratio, equivalent diameter, water content, tensile strength, porosity as well as shrinking, and disintegration behavior and drug release according to their MCC II fraction. Furthermore, the pellets were stored at different relative humidities (0–97%rh), and the influence on their disintegration and drug release was investigated.With increasing MCC II fraction, the pellets became lager in size, decreased their porosity, and required higher water contents for spheronization. Moreover, the disintegration time increased and the disintegration itself was incomplete. Furthermore, the storage conditions had an impact on the disintegration properties of MCC II-based pellets. The disintegrating was affected irreversibly after storage at high humidity (80–97%rh) resulting in a slow drug release. Therefore, MCC II-based pellets need to be stored below 80%rh to secure a fast disintegration.A better knowledge of the properties of MCC II-based pellets was obtained providing a basis for a successful manufacturing and adequate storage of MCC II-based pellets prepared by extrusion/spheronization.  相似文献

目的观察大气细颗粒物(PM2.5)致大鼠气道黏液纤毛清除系统的损伤作用及标准桃金娘油干预的影响。方法将大鼠分成对照组(N组)、PM2.5暴露组(PM组)、标准桃金娘油干预组(PM+CS组),分别予以吸入净化空气、暴露石家庄室外空气(PM2.5浓度的范围29.90 mg/m^3~99.65 mg/m^3)、标准桃金娘油混悬液300 mg/(kg·d)灌胃。于3和6个月各处死大鼠8只,气管及肺组织行HE和AB-PAS染色;扫描电镜观察纤毛超微结构;计数BALF中细胞及分类;RT-qPCR测定黏蛋白(MUC)5AC mRNA和水通道蛋白(AQP)5 mRNA表达;免疫组化、Western blot测定MUC5AC蛋白表达。结果PM组肺及气管组织炎性损伤及黏液细胞化生明显;气管纤毛黏连、倒伏及缺失、排列稀疏、紊乱,BALF中白细胞总数、中性粒细胞计数和中性粒细胞百分比显著高于N组(P<0.05);MUC5AC mRNA相对表达量显著高于N组(P<0.05);AQP5 mRNA相对表达量显著低于N组(P<0.05);MUC5AC蛋白表达显著高于N组(P<0.05)。与PM组相比,标准桃金娘油可减轻上述损伤作用(P<0.05)。结论标准桃金娘油可通过减轻PM2.5致大鼠的纤毛结构损伤、黏液高分泌和炎性损伤作用,对黏液纤毛清除系统起保护作用。  相似文献

A patient with isolated 3-methylcrotonyl-CoA carboxylase (MCC) deficiency with an unusual clinical presentation is described. The patient presented with clusters of seizures with two or three months disease free interval in the first year of life which then evolved into attacks of status epilepticus after the age of 12 months. MCC deficiency was suspected because of elevated C5-OH-carnitine in tandem mass spectrometry and elevated 3-hydroxy-isovaleric acid in urine organic acid analysis. Deficiency of MCC was confirmed in cultured fibroblasts and mutation analysis revealed a novel mutation in MCCB, p.S39F. Attacks of status epilepticus as a predominant symptom have not been described before in isolated MCC deficiency.  相似文献

目的:研究主成分分析法及聚类分析法在微晶纤维素分类中的应用。方法:测定了来源于5个不同厂家的12种PH101和PH102型微晶纤维素的粉体学等物理性质,采用主成分分析法及聚类分析法对微晶纤维素的分类进行研究。结果:主成分分析和聚类分析的分析结果一致,主成分分析揭示了PH101和PH102型微晶纤维素分类的依据。结论:主成分分析法和聚类分析法可以用于微晶纤维素的分类研究,其结果与传统生产应用中微晶纤维素分类一致。  相似文献
