大鼠急性心肌损伤E选择素和细胞因子的变化   总被引:5,自引:1,他引:5  
目的观察异丙肾上腺素导致大鼠急性心肌损伤早期E选择素和细胞因子TNF-α、IL-1βmRNA的变化,进一步探讨E选择素和细胞因子TNF-α、IL-1β在异丙肾上腺素导致大鼠急性心肌损伤发生发展中的作用。方法将大鼠随机分为对照组(35只)和实验组(35只),实验组给予异丙肾上腺素(isoproterenol,Iso),对照组给予生理盐水,分别在给药后4、8、16、24、48h各取7只大鼠心脏,光镜下观察心肌形态结构病理改变,应用逆转录-聚合酶链反应(RT-PCR)检测心肌组织E选择素、TNF-α和IL-1βmRNA的表达。结果实验组4hE选择素mRNA表达与对照组比较差异无统计学意义(P>0.05),实验组E选择素mRNA表达8h开始升高,24h达高峰,48h恢复正常;实验组TNF-αmRNA表达4h已经升高,16h达高峰,24h到48h逐渐下降;实验组IL-1βmRNA表达4h已经升高,8h达高峰,16h到48h逐渐下降。结论E选择素和细胞因子TNF-α、IL-1β在大鼠急性心肌损伤的发生中具有重要意义。  相似文献

目的研究二苯乙烯苷(TSG)对H2O2诱导损伤的人脐静脉内皮细胞P选择素和E选择素表达的影响。方法运用逆转录聚合酶链反应(RT-PCR)和酶联免疫吸附试验(ELISA)分别检测P选择素和E选择素的mRNA和蛋白表达。结果 200μmol/L的H2O2作用人脐静脉内皮细胞24 h,P选择素和E选择素的mRNA和蛋白表达水平均明显上调,与空白对照组比较,差异具有显著性(P<0.01),但TSG预处理人脐静脉内皮细胞4 h后再加200μmol/L H2O2作用24 h,H2O2诱导的内皮细胞P选择素和E选择素的mRNA和蛋白表达水平降低,与H2O2处理组比较,差异有显著性(P<0.05)。结论 TSG通过降低黏附分子P选择素和E选择素的表达可保护内皮细胞免受氧化应激损伤,影响动脉粥样硬化进程。  相似文献

In the present paper, the acute and subchronical inflammatory processes of the vaginal epithelial were studied in mice experimentally infected with two Trichomonas vaginalis strains of different pathogenicity, by means of histological and immunological methods. There was an increase in the stratified epithelium layers as well as edema produced by the increase of vascularization in the propia submucosa and infiltration of leukocytes. The proliferation of the vaginal epithelium favors the settlement and persistence of the parasitic infection. All of the findings corresponded with signs of a systemic disease being observed in the animals, including significant weight loss and also intestinal invasion. The entire inflammatory process has been corroborated by studies of adhesion molecules such as E-Selectin, VCAM-1 and PECAM-1. A correlation between the time of appearance and the perseverance of the inflammatory process with E-Selectin and VCAM-1 expression was observed, but not with PECAM-1. The strain with a higher pathogenicity was able to invade deep vaginal tissues and thus, parasites could not be detected by vaginal washings. This may be an important cause of diagnosis and treatment failure. Also, by the different localization of trichomonads, it appeared that the battle between host and parasite took place in different areas dependent upon the characteristics of the strain.  相似文献

目的研究在人脐静脉内皮细胞中3-羟-3-甲基戊二酰辅酶A(HMG-CoA)还原酶对CD40/CD40L系统介导的粘附分子表达的影响,及对淋巴细胞与内皮细胞的粘附功能的影响。方法采用人脐静脉内皮细胞株ECV-304,CD40L干预,使之与其表面的CD40作用,用不同浓度辛伐他汀或辛伐他汀(5μmol.L-1)+甲羟戊酸干预,流式细胞术和RT-PCR检测干预前后E选择素(E-Selectin)的表达,流式细胞术检测CD40L介导的淋巴细胞与内皮细胞的粘附功能。结果不同浓度辛伐他汀可下调CD40L介导的E-Selectin的表达,并呈一定的剂量依赖性。甲羟戊酸(400μmol.L-1)抑制了辛伐他汀(5μmol.L-1)对E-Selectin表达的下调作用。辛伐他汀可降低CD40L介导的淋巴细胞与内皮细胞的粘附功能。结论他汀类药物的抗炎作用与抑制CD40-CD40L系统有关,其机制是通过抑制HMG-CoA还原酶而发挥作用的。  相似文献

Maternal serum levels of VCAM-1, ICAM-1 and E-selectin in preeclampsia   总被引:1,自引:0,他引:1  
Endothelial dysfunction is thought to be a central pathogenic feature in preeclampsia on the basis of elevated adhesion molecules. The aim of the present study was to compare the levels of soluble vascular cell adhesion molecule-1 (sVCAM-1), intercellular adhesion molecule-1 (sICAM-1) and E-selectin (sE-selectin) in sera of normal and preeclamptic pregnancies. We studied the serum levels of sVCAM-1, sICAM-1 and sE-selectin in normal pregnant women (n=63), mild preeclampsia (n=33) and severe preeclampsia (n=82). Concentrations of soluble adhesion molecules were determined with enzyme-linked immunoassay (ELISA). Serum concentrations of sVCAM-1 were significantly higher in both mild (p=0.004) and severe preeclampsia (p=0.000) than normal pregnancy. There were also significant differences in sVCAM- 1 levels between mild and severe preeclampsia (p=0.002). sICAM-1 levels of severe preeclampsia were statistically different from those of normal pregnancy (p=0.038). Levels of sE-selectin were elevated in both mild (p=0.011) and severe preeclampsia (p=0.000) compared to normal pregnancy, but no statistical difference between the mild and severe preeclampsia (p=0.345). These results suggest that all three soluble adhesion molecules are increased in severe preeclampsia, and sVCAM-1 among them may be useful in predicting the severity of preeclampsia.  相似文献

目的 探讨糖基化终产物在糖尿病动脉粥样硬化形成中的作用机理。方法 分离正常人脐静脉内皮细胞,将糖基化终产物修饰的人血清白蛋白及未修饰人血清白蛋白与人脐静脉内皮细胞在体外共同培养,用荧光单克隆抗体染色,流式细胞仪定量检测内皮细胞表面选择素E的表达。结果 正常人脐静脉内皮细胞未表达选择素Eo糖基化终产物修饰的人血清白蛋白能以时间和剂量依赖的方式上调血管内皮细胞粘附分子选择素E的表达(P〈0.015),而人血清白蛋白对人脐静脉内皮细胞粘附分子选择素E的表达无影响。结论 糖基化终产物能上调人脐静脉内皮细胞粘附分子的表达,从而促进动脉粥样硬化时单核,巨噬细胞的浸润。  相似文献

目的探讨血清可溶性TNF-α、IL-1β、转化生长因子-β1(TGF-1β)、E-选择素(E-sel)和L-选择素(L-sel)在肾综合征出血热(HFRS)发病机制中的作用。方法41例HFRS患者(轻症组22例,重症组19例)纳入研究,按病期采血,分别采用放射免疫法检测TNF-α、IL-1β,ELISA法检测TGF-β1、E-sel和L-sel,自动生化仪检测肝、肾功能和PLT计数。结果从发热期至多尿期,血清IL-1β、TNF-α、E-sel和L-sel水平均升高,其峰值与对照组比较,分别为(0.56±0.10)μg/LH5(0.19±).06)μg/L、(2.41±1.61)μg/L比(0.824-0.16)μg/L、(198.4±29.2)μg/L比(56.4±25.8)μg/L、(1372.5±137.3)μg/L比(1089.9±137.9)μg/L,差异有统计学意义(P值均〈0.01)。动态观察,上述细胞因子和粘附分子的变化曲线与ALT、BUN的变化趋势一致。血清TGF-1β、TGF-β1/IL-1β和TGF-β1/TNF-α比值在病程前三期均显著降低,与PLT的变化趋势相似。结论HFRS急性期血清IL-1β、E-sel、L-sel升高和TGF-β1水平下降是造成失控性全身炎性反应和肝、肾功能损害的重要因素,在HFRS发病机制中起重要作用;合理的对症治疗和适量使用免疫调节剂可改善预后。  相似文献

Exposure to PM10 is associated with cardiovascular effects. We evaluated the effects of PM10 on E-Selectin expression and monocytic cell adhesion in human umbilical vein endothelial cells (HUVECs). HUVEC were exposed to PM10 (5-40 microg/cm2) for 6 h, following which surface E-Selectin expression was detected by fluorescence microscopy and flow cytometry. The effects of total particles, particles treated with polymixin-B to block the effects of endotoxin, and both soluble and insoluble fractions of particles, were assessed. Incubation with PM10 lead to a concentration-related increase of E-Selectin expression (>seven-fold increase at 40 microg/cm2). Particles pre-treated with polymixin-B inhibited E-Selectin expression to a level slightly higher than untreated particles. An increase in fluorescence was also observed with the insoluble fraction, while the soluble fraction had no significant effect. HUVEC exposed to PM10 were also evaluated for adhesivity of monocytic cells (U937). PM10 strongly increased the adhesion of U937 cells to HUVEC. In conclusion, PM10 induces endothelial cell activation, evidenced by enhanced E-Selectin expression. This activation is manifested functionally as an increase in monocytic cell adhesion. Insoluble components as well as endotoxins appear to be responsible for this activity.  相似文献
