目的:检测免疫共刺激分子B7-H4及Fas、FasL在不同宫颈组织中的表达,研究三者的异常表达与宫颈癌免疫逃逸机制的关系。方法:选择陕西省肿瘤医院病理科2014-2018年的石蜡包埋标本162例,采用免疫组织化学方法检测20例正常宫颈、30例宫颈上皮内瘤变(CIN)和112例I期-IV期宫颈癌组织中B7-H4及Fas、FasL的表达水平。结果:B7-H4在正常宫颈组、CIN组、宫颈癌组的阳性表达率分别为0、20.0%、47.32%,两两相比,具有显著性差异(P<0.05)。在112例宫颈癌病例中FIGO临床分期I期至IV期各期别宫颈癌组织B7-H4的阳表达率分别为16.67%、43.18%、70.83%、85.71%,各组间相比差异具有统计学意义。正常对照组Fas的阳性表达率是85.00%,CIN组43.33%,宫颈癌组41.96%,正常组与CIN、正常组与宫颈癌组相比差异均具有统计学意义(P<0.05),CIN组与宫颈癌组相比,无明显差异(P>0.05)。FasL的阳性表达率在正常组为20.00%,CIN组为50.00%,宫颈癌组为56.25%,正常组与CIN、宫颈癌组比较,差异具有统计学意义(P<0.05),CIN组与宫颈癌组比较无显著差异;B7-H4阳性宫颈癌组Fas阳性表达率28.30%明显较B7-H4阴性宫颈癌组Fas阳性表达率54.24%低,差异具有统计学意义(P<0.05);而B7-H4阳性宫颈癌组FasL阳性表达率79.25%明显高于B7-H4阴性宫颈癌组FasL阳性表达率35.59%,差异具有统计学意义(P<0.05)。结论:负性免疫共刺激分子B7-H4和Fas、FasL凋亡蛋白在宫颈癌的发生、进展中起到了重要作用,其协同作用可能为宫颈癌细胞逃逸机体免疫的机制之一。  相似文献

BACKGROUNDDue to the special clinical features and biologic characteristics of adolescent and young adult (AYA) cancers, AYA cancers are different from cancers in children and elderly individuals. However, there are few reports on AYA hepatocellular carcinoma (HCC).AIMTo investigate the overall survival (OS) of AYA (15-39 years) and elderly (40-74 years) patients with HCC.METHODSThe data of all the HCC cases were extracted from the Surveillance, Epidemiology, and End Results database from 2004 to 2015 and were then divided into two groups based on age: AYA group (15-39 years) and older group (40-74 years). Kaplan-Meier curves and log-rank tests were used to compare the OS of the two groups. Propensity score matching (PSM) was employed to analyze the OS difference between the two groups. The Cox proportional hazards regression model was used to perform multivariate analysis to explore the risk factors for OS of HCC patients.RESULTSCompared to elderly cancer patients, AYA patients with HCC had a worse Surveillance, Epidemiology, and End Results stage, including the distant stage (22.1% vs 15.4%, P < 0.001), and a more advanced American Joint Committee on Cancer (AJCC) stage, including AJCC III and IV (49.2% vs 38.3%, P < 0.001), and were more likely to receive surgery (64.5% vs 47.5%, P < 0.001). Before PSM, the AYA group had a longer survival in months (median: 20.00, interquartile range [IQR]: 5.00-62.50) than the older group (median: 15.00, IQR: 4.00-40.00) (P < 0.001). After PSM, the AYA group still had a longer survival in months (median: 21.00, IQR: 5.00-64.50) than the older group (median: 18.00, IQR: 6.00-53.00) (P < 0.001). The Cox proportional hazards regression model showed that advanced age (hazard ratio [HR] = 1.405, 95%CI: 1.218-1.621, P < 0.001) was a risk factor for OS of HCC patients. In the subgroup analysis, the Cox proportional hazards regression model showed that in AJCC I/II HCC patients, advanced age (HR = 1.749, 95%CI: 1.352-2.263, P < 0.001) was a risk factor for OS, while it was not a risk factor in AJCC III/IV HCC patients (HR = 1.186, 95%CI: 0.997-1.410, P = 0.054) before PSM. After PSM, advanced age (HR = 1.891, 95%CI: 1.356-2.637, P < 0.001) was still a risk factor for OS in AJCC I/II HCC patients, but was not a risk factor for OS in AJCC III/IV HCC patients (HR = 1.192, 95%CI: 0.934-1.521, P = 0.157) after PSM.CONCLUSIONAYA patients with HCC have different clinical characteristics from older adults. In different AJCC stages, the two groups of patients have different OS: In AJCC I/II HCC patients, advanced age is a risk factor for OS, but it is not a risk factor for OS in the AJCC III/IV HCC patient group.  相似文献

Long non-coding RNAs (lncRNAs) have been identified as crucial regulators in the tumorigenesis and progression of hepatocellular carcinoma (HCC). Recently, long intergenic non-protein coding RNA 205 (LINC00205) has been identified as a prognostic biomarker in HCC. However, the biological role of LINC0205 and its potential molecular mechanism are poorly investigated. Here, we found that the expression of LINC00205 was dramatically up-regulated in HCC tissues compared to adjacent nontumor tissues. Furthermore, the level of LINC00205 in both Hep3B and Huh7 cells was prominently higher than that in normal hepatic cell line LO2. Notably, the high expression of LINC00205 was strongly correlated with tumor size ≥5 cm, venous infiltration and advanced tumor stages. Functionally, LINC00205 knockdown obviously repressed the proliferation, migration and invasion of Hep3B and Huh7 cells in vitro. An inverse correlation between LINC00205 and miR-122-5p was detected in HCC tissues. Interestingly, LINC00205 knockdown increased the level of miR-122-5p in both Hep3B and Huh7 cells. Mechanistically, luciferase reporter assay demonstrated LINC00205 acted as a competing endogenous RNA (ceRNA) by directly interacting with miR-122-5p. More importantly, miR-122-5p overexpression significantly restrained the proliferation, migration and invasion of HCC cells. Collectively, our study provides solid evidence to support the oncogenic role of LINC00205 in HCC, which may be benefit for the improvement of HCC therapy.  相似文献

MicroRNAs (miRNAs) have been found to be aberrantly expressed and exert essential roles in the tumorigenesis and progression of gastric cancer (GC). miR-301b-3p has been recognized as a cancer-related miRNA in lung cancer, bladder cancer and hepatocellular carcinoma. However, the function of miR-301b-3p in GC progression and its underlying mechanism have not been studied yet. In this study, we found that miR-301b-3p expression was up-regulated in GC tissues compared to adjacent noncancerous tissues. Furthermore, the elevated levels of miR-301b-3p were detected in GC cell lines (SGC-7901, AGS, MKN-45 and MGC-803) as compared with GES-1 cells. Interestingly, GC tissues from patients with tumor size ≥ 5 cm and advanced tumor stages showed obvious higher levels of miR-301b-3p compared to matched controls. Functionally, miR-301b-3p knockdown prominently inhibited cell proliferation, and induced cell cycle arrest at G1 phase and apoptosis in MGC-803 cells. Meanwhile, ectopic expression of miR-301b-3p conversely regulated these biological behaviors of MKN-45 cells. Next, we found that miR-301b-3p knockdown increased, whereas miR-301b-3p overexpression reduced the expression of zinc finger and BTB domain containing 4 (ZBTB4) in GC cells. Accordingly, luciferase reporter assay identified ZBTB4 as a direct target of miR-301b-3p. ZBTB4 overexpression markedly restrained the growth of MGC-803 cells. More importantly, ZBTB4 silencing partially reversed miR-301b-3p knockdown-induced tumor suppressive effects on MGC-803 cells. In conclusion, we firstly revealed that miR-301-3p was highly expressed in GC and contributed to tumor progression via attenuating ZBTB4, which might provide a novel molecular-targeted strategy for GC treatment.  相似文献

BackgroundOsteoarthritis (OA) is a type of degenerative joint disease affecting millions of individuals worldwide. However, there are currently no great inflammatory treatments available for it. Sophocarpine (SPC), one of the key bioactive compounds derived from Sophora flavescens, has shown remarkable anti-inflammatory effects.MethodsIn this study, we evaluated the effect of SPC on preventing the progression of OA and investigated its molecular target involved. In brief, rat chondrocytes were pretreated with SPC and subsequently stimulated with IL-1β. We found that SPC reduced the production of pro-inflammatory cytokines, such as nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor alpha (TNF-α), and interleukin-6 (IL-6). SPC also inhibited the expression of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) at both the gene and protein level. Moreover, SPC promoted the expression of anabolic factors Sox-9 and aggrecan, while inhibiting the expression of catabolic factors, such as matrix metalloproteinases 13 (MMP-13) and thrombospondin motifs 5 (ADAMTS-5) in rat chondrocytes. Mechanistically, we found that SPC inhibited nuclear factor kappa B (NF-κB) via the phosphatidylinositol 3 kinase (PI3K)/AKT pathway. The beneficial effects of SPC were also observed in vivo using a rat OA model.ConclusionsOur findings indicate that SPC may be a potential novel therapeutic in the treatment of OA.  相似文献

目的:探索HGF/c-met传导通路抑制剂SU11274抑制子宫内膜癌细胞增殖的分子机制。方法:使用不同浓度SU11274(0.5 μmol/L、1.0 μmol/L、1.5 μmol/L)作用ishikawa 和HEC-1B 两种细胞株1小时后,加入40 ng/ml的HGF,继续培养48小时,然后使用RT-PCR检测c-met、Survivin、XIAP水平以及Western blot检测细胞中Survivin、XIAP蛋白表达水平。结果:ishikawa细胞c-met低表达,而HEC-1B细胞c-met明显高表达,HEC-1B细胞中c-met mRNA表达量为ishikawa中的2.5倍。SU11274可以使HEC-1B、ishikawa细胞的Survivin、XIAP蛋白表达下调,呈现剂量依赖性,并且在HEC-1B细胞中,该下调作用明显强于ishikawa细胞(P<0.05)。结论:SU11274可以通过下调HGF/c-met传导通路中Survivin、XIAP的表达来抑制人子宫内膜癌细胞增殖。  相似文献

目的:回顾性分析胸膜间皮瘤患者的临床资料,包括病史、症状、淋巴结及远处脏器转移的规律,并分析不同分期、病理类型及治疗方式的患者生存情况,为该疾病的诊断和治疗提供思路。方法:分析空军军医大学唐都医院2009年4月-2016年9月收治的120例胸膜间皮瘤患者资料,基于第8版TNM分期标准进行肿瘤分期。对不同临床分期、病理类型及治疗方式的患者生存期进行统计分析,应用Kaplan-Meier法进行生存分析。结果:120例患者中有职业暴露史3例(占2.5%)。发病部位为左侧胸膜56例(占46.7%),右侧胸膜63例(占52.5%),双侧胸膜1例。总体的误诊率为30%(36/120),其中误诊为结核32例(26.7%)。以胸痛症状发病55例(占45.8%),胸腔积液107例(占89.2%),两者合并为49例(占40.8%)。确诊方法包括胸膜穿刺诊断53例(占44.2%),胸腔镜诊断41例(占34.2%),开胸探查诊断26例(占21.6%)。所有患者中淋巴结转移(N1~N2)共43例(占35.8%),涉及54个淋巴区;纵隔淋巴结转移42例(占77.8%),纵隔外淋巴区转移12例(占22.2%)。成功随访78例患者,总体中位生存期为15.0个月,1、2、3及5年生存率分别为67.9%、27.7%、16.0%及4.2%。Ⅰ/Ⅱ期患者中位生存期为17.0个月,明显长于Ⅲ/Ⅳ期患者的13.0个月(P<0.05)。上皮型肿瘤患者的中位生存期为17.0个月,与混合型及肉瘤型患者比较有生存优势(P值均<0.05)。手术联合化疗组的患者中位生存期为30.0个月,优于单纯手术、单纯化疗及支持治疗的患者(P值均<0.05)。结论:胸膜间皮瘤的误诊率高,其中大多被误诊为肺结核。恶性胸膜间皮瘤患者主要以胸腔积液症状发病,而且淋巴结转移规律与肺癌有所不同。所以在胸膜间皮瘤的诊断和治疗中,应高度重视这些临床特征,尽量减少误诊,以提高治疗效果。另外,临床分期较早及上皮型病理类型的患者预后相对较好,而且采用手术联合化疗的综合治疗模式可以使胸膜间皮瘤患者获得相对较长的生存期。  相似文献
