三氧化二砷诱导K562细胞凋亡的初步研究＊

To explore the mechanisms of arsenic trioxide(As₂O₃) in treatment of chronic myelocytic leukemia(CML), K₅₆₂ cell line was used as an in vitro model. The effect of As₂O₃ on K₅₆₂ cells was observed by cell proliferation, cell viability, cell morphology,tumor colony formation and agarose gel electrophoresis assay. The results showed that As₂O₃ at a concentration greats or equal 2.5μmol/L could inhibit the cell proliferation and induce the apoptotic morphology and DNA fragmentation of K₅₆₂ Cells. These results suggest that As₂O₃ can induce the apoptosis of K₅₆₂ cells efficiently.
