In clear cell renal cell carcinoma (ccRCC), glycolysis is enhanced mainly because of the increased expression of key enzymes in glycolysis. Hence, the discovery of new molecular biomarkers for glycolysis may help guide and establish a precise system of diagnosis and treatment for ccRCC. Expression profiles of 1079 tumor samples of ccRCC patients (including 311 patients treated with everolimus or nivolumab) were downloaded from public databases. Proteomic profiles of 232 ccRCC samples were obtained from Fudan University Shanghai Cancer Center (FUSCC). Biological changes, tumor microenvironment and prognostic differences were explored between samples with various glycolysis characteristics. There were significant differences in CD8+ effector T cells, epithelial-to-mesenchymal transition and pan-fibroblast TGFb between the Low and High glyScore groups. The tumor mutation burden of the Low glyScore group was lower than that of the High glyScore group. And higher glyScore was significantly associated with worse overall survival (OS) in 768 ccRCC patients (P < .0001). External validation in FUSCC cohort also indicated that glyScore was of strong ability for predicting OS (P < .05). GlyScore may serve as a biomarker for predicting everolimus response in ccRCC patients due to its significant associations with progression-free survival (PFS). And glyScore may also predict overall survival in patients treated with nivolumab. We calculated the glyScore in ccRCC and the defined glyScore was of strong ability for predicting OS. Whether ultra-processed food consumption is associated with the risk of pancreatic cancer has not been determined. We performed a prospective study to fill this gap. A population-based cohort of 98 265 American adults was identified from the Prostate, Lung, Colorectal and Ovarian Cancer Screening Trial. Ultra-processed foods were defined by the NOVA classification. Cox regression was used to estimate hazard ratios (HRs) for pancreatic cancer incidence. Subgroup analysis was performed to identify the potential effect modifiers. During a mean follow-up of 8.86 years, 387 pancreatic cancer cases occurred. High consumption of ultra-processed foods was found to be associated with an increased risk of pancreatic cancer (fully adjusted HRquartile 4 vs 1:1.49; 95% confidence interval [CI]: 1.07-2.07; Ptrend = .021) in a linear dose-response manner (Pnonlinearity = .075). Subgroup analysis further found that the positive association of ultra-processed food consumption with the risk of pancreatic cancer was more pronounced in subjects aged <65 years (HRquartile 4 vs 1:2.17; 95% CI: 1.14-4.15) than in those aged ≥65 years (HRquartile 4 vs 1:1.32; 95% CI: 0.88-1.94), though the interaction test failed to achieve the statistical significance (Pinteraction = .061). A high risk of developing second primary malignancy (SPM) has been reported among head and neck cancer patients. Here, we aimed to statistically quantify the impact of SPM development on the survival of head and neck cancer patients. Our study was conducted using the Surveillance, Epidemiology and End Results database to collect the data of 48 316 patients who received curative surgical resection for initial primary head and neck squamous cell carcinoma (IP-HNSCC) in 1975 to 2019. SPM diagnosis was treated as a time-varying covariate and multivariable Cox regression analysis was conducted to estimate the association between SPM development and survival, overall or by the subsite of IP-HNSCC. Of the included patients, 11 238 patients (23.3%) developed SPM during the follow-up period. A significant reduction in survival was observed among patients with SPM (hazard ratio [HR] for overall survival, 3.30; 95% confidence interval [CI]: 3.20-3.41). The impact of SPM development on reduced survival was more significant in patients with localized IP-HNSCC vs regional IP-HNSCC (HROS, 3.41; 95% CI: 3.24-3.6 vs HROS, 3.18; 95% CI: 3.05-3.31; P for interaction <.001). The survival impact of SPM development was more evident in younger patients than in older patients. SPM in lung and bronchus was associated with the most pronounced reduction in survival, overall and across all subsites of HNSCC. Our results indicated that SPM development led to a significant reduction in survival. A greater survival benefit may be achieved through intensive surveillance for SPM in lung and bronchus targeting younger patients and those with localized HNSCC. 相似文献

目的分析重症肺炎(痰热壅肺型)合并Ⅱ型呼吸衰竭应用喜炎平联合自拟通腑益肺汤治疗效果情况。方法选取本院2019年7月至2020年7月诊治的100例重症肺炎(痰热壅肺型)合并Ⅱ型呼吸衰竭患者资料,按随机数表分两组,对照组给予喜炎平,研究组给予喜炎平联合自拟通腑益肺汤,分析两组临床效果、肺功能及炎症因子指标情况。结果研究组总有效率(98.00%)比对照组(82.00%)高(P<0.01,χ;=7.1111),且研究组TLC[(5.15±0.75)L]、FEV;[(2.91±0.41)L]及MMV[(107.45±11.52)L]等肺功能高于对照组[(4.43±0.69)L、(2.31±0.43)L及(96.61±9.28)L;P<0.01,t=4.9957、5.1816];研究组患者IL-6[(25.38±5.18)ng/L]、PCT[(0.93±0.18)ng/L]与PAF[(9329.83±1136.28)U/L]等炎症因子低于对照组[(43.52±7.30)ng/L、(1.28±0.26)ng/L、(11851.96±1532.36)U/L;P<0.01,t=14.3300、8.0498、9.3486]。结论重症肺炎(痰热壅肺型)合并Ⅱ型呼吸衰竭应用喜炎平联合自拟通腑益肺汤治疗,可改善肺功能情况,降低机体炎症因子指标,临床用药效果显著。 相似文献

目的基于磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)通路探究丙泊酚对七氟烷诱导大鼠神经细胞凋亡的保护作用。方法2020年1月—2021年1月于湖北省恩施土家族苗族自治州中心医院实验室进行实验,选取健康、清洁级Wistar雄性大鼠30只,随机数字表法分为对照组、七氟烷组、七氟烷+丙泊酚组(联合组),各10只。七氟烷组吸入1.5%七氟烷干预,联合组在七氟烷组基础上加丙泊酚150 mg/kg腹腔注射,对照组给予等体积的生理盐水干预,均连续干预2周。干预后各组大鼠行Morris水迷宫实验,观察大鼠海马神经细胞凋亡情况,检测氧化应激相关指标、凋亡相关因子及PI3K/Akt通路蛋白表达水平。结果逃避潜伏期、海马神经细胞凋亡率比较,七氟烷组>联合组>对照组(F/P=17.038/0.001、36.834/0.001),穿越平台次数、目标象限停留时间占比比较,七氟烷组<联合组<对照组(F/P=8.913/0.001、9.125/0.001)。LDH、MDA、Bax、Caspase-3水平比较,七氟烷组>联合组>对照组(F/P=35.166/0.001、20.773/0.001、43.896/0.001、17.462/0.001),SOD、GSH-Px、Bcl-2水平及PI3K、p-Akt表达比较,七氟烷组<联合组<对照组(F/P=12.092/0.001、16.243/0.001、15.236/0.001、28.850/0.001、28.869/0.001)。结论丙泊酚能改善七氟烷诱导的大鼠认知功能、氧化应激反应,抑制神经细胞凋亡,可能与PI3K/Akt通路有关。 相似文献
