目的 探讨我国弓形虫Chinese 1优势基因型感染对宿主脑组织铁代谢及脑损伤的影响。方法 将20只C57BL/6(体质量15~17 g)小鼠随机分为对照组和感染组,每组10只。感染组每只小鼠腹腔注射4 000个弓形虫Chinese 1优势基因型TgCtwh3虫株速殖子,对照组小鼠注射等量无菌PBS,饲养6 d后处死小鼠并取其脑组织。采用电感耦合等离子体质谱法(inductively coupled plasma mass spectrometry, ICP⁃MS)检测小鼠脑组织铁元素水平;采用RNA芯片检测两组小鼠脑组织差异基因数目并对功能基因表达情况进行基因本体论(Gene Ontology, GO)功能富集;采用实时荧光定量PCR(fluorescent quantitative real⁃time PCR, qPCR)技术检测小鼠脑组织中弓形虫表面抗原1(Toxoplasma gondii surface antigen 1,TgSAG1)基因及部分锌铁调控蛋白(Zrt⁃ and Irt⁃like protein, ZIP)家族mRNA表达水平;采用光镜和电镜观察小鼠脑组织海马齿轮回(dentate gyrus, DG)超微结构;采用Western blotting检测谷胱甘肽过氧化物酶4(glutathione peroxidase 4, GPx4)蛋白表达水平;采用硫代巴比妥酸(TBA)法检测丙二醛(malondialdehyde, MDA)水平;采用免疫组化检测血管内皮生长因子(vascular endothelial growth factor, VEGF)蛋白表达光密度(optical density, OD)值。结果 光镜下可见感染组小鼠脑组织海马DG区细胞坏死,电镜下见感染组小鼠脑组织海马区出现胞质空泡化、核皱缩坏死、线粒体嵴断裂消融、自噬小体增加等超微结构变化。与对照组相比,感染组小鼠脑组织中铁元素水平上调[(32.92 ± 0.90) µg/g vs.(37.72 ± 1.10) µg/g;t = 3.397, P < 0.01];RNA芯片检测感染组小鼠脑组织发现721个基因上调、276个基因下调,差异表达基因在金属离子结合能力上有明显富集。与对照组相比,感染组小鼠脑组织金属元素转运体ZIP2 mRNA表达水平上调(t = 8.659,P < 0.05)、GPx4表达下降[(1.046 ± 0.025) vs. (0.720 ± 0.101);t = 3.129,P < 0.01])、MDA水平升高[(4.37 ± 0.33) nmol/mgprot vs.(5.93 ± 0.54) nmol/mgprot;t = 2.451,P < 0.05)]、VEGF蛋白平均OD值上调[(0.348 3 ± 0.017 8) vs. (0.490 6 ± 0.010 5);t = 6.641,P < 0.01]。结论 Chinese 1优势基因型弓形虫感染后,小鼠脑组织中铁元素蓄积、抗氧化能力下调、氧化应激水平升高,提示弓形虫感染可影响宿主脑组织铁代谢而导致脑损伤。  相似文献

目的 观察温阳解郁颗粒(Wenyang Jieyu granule,WYJY)对皮质酮(Corticosterone,CORT)诱导损伤型小鼠海马神经细胞(TH22 cell)的保护作用,基于脑源性神经营养因子(Brain derived neurotrophic factor, BDNF)/酪氨酸激酶B(Tyrosine kinase B, TrkB)/细胞外信号调节蛋白激酶(Extra cellular regulated protein kinases, ERK)信号通路探讨WYJY保护海马神经细胞的作用机制。方法 体外构建小鼠海马神经细胞皮质酮诱导损伤模型,以不同浓度的WYJY和氟西汀(Fluoxetine,FXT)含药血清作用于模型细胞,细胞增殖-毒性检测(Cell Counting Kit-8, CCK-8)法分析细胞活性,倒置显微镜下观察给药前后细胞形态结构的改变,采用蛋白免疫印迹法(Western Blot)、实时荧光定量PCR(Quantitative real-time PCR, qPCR)法检测神经细胞内凋亡因子(BCL2-Associated X, Bax)、抗凋亡因子(B-cell lymphoma-2, Bcl-2)、BDNF、Trkb、ERK以及丝氨酸/苏氨酸激酶(Phospho-p90RSK, RSK)、环磷腺苷效应元件结合蛋白(cAMP-response element binding protein, CREB)蛋白表达水平以及相关基因的表达水平。结果 在浓度为459.5 μmol·L-1的CORT作用24 h后,HT22细胞的活性抑制率达到50%,在此条件作用下细胞形态结构损伤明显,凋亡程度严重,细胞上清中BDNF的含量显著减少(P<0.05),细胞内凋亡相关因子Bax/Bcl-2的比值明显升高(P<0.01),BDNF、Trkb、ERK、RSK、CREB磷酸化蛋白表达水平和mRNA表达水平明显降低(P<0.01);以5%的浓度为2.85 g·kg-1的WYJY和10%的FXT含药血清作用于受损的HT22细胞后,HT22细胞存活率明显提升(P<0.01),细胞结构的损伤明显改善,细胞凋亡程度减轻,细胞外BDNF的含量显著升高(P<0.05),细胞内Bax/Bcl-2比值显著下调(P<0.01),BDNF、Trkb、ERK、RSK、CREB磷酸化蛋白表达水平和mRNA表达水平显著提升(P<0.05,P<0.01)。结论 温阳解郁颗粒可有效保护高浓度CORT造成的小鼠海马神经细胞损伤。调控BDNF/Trkb/ERK通路,放大CREB信号传导,影响Bcl-2、BDNF水平,可能是其保护海马神经元,发挥抗抑郁疗效的重要机制。  相似文献

Spinster 2 (Spns2) is a transporter that pumps sphingosine-1-phosphate (S1P), a bioactive lipid mediator synthesized in the cytoplasm, out of cells into the inter cellular space. S1P is a signal that modulates cellular behavior during embryonic development, inflammation and tissue repair, etc. A Spns2-null (KO) mouse is born with failure of eyelid closure (eyelid-open-at birth; EOB) and develop corneal fibrosis in adulthood. It remains elusive whether corneal lesion is caused by exposure to keratitis (lagophthalmos) of EOB phenotype or the loss of Spns2 directly perturbs the corneal tissue morphogenesis and intra-eyelid structures. Therefore, we investigated differences between the cornea and ocular adnexa morphogenesis in KO and wild-type (WT) embryos and adults as well.The loss of Spns2 perturbs cornea morphogenesis during embryonic development as early as E16.5 besides EOB phenotype. Histology showed that the corneal stroma was thinner with less extracellular matrix accumulation, e.g., collagen and keratocan in the KO mouse. Epithelial stratification, expression of keratin 12 and formation of desmosomes and hemidesmosomes were also perturbed in these KO corneas. Lacking Spns2 impaired morphogenesis of the Meibomian glands and of orbicularis oculi muscles. KO glands were labeled for ELOVL4 and PPARγ and were Oil-Red O-positive, suggesting KO acinar cells possessed functionality as the glands.This is the first report on the roles of Spns2 in corneal and Meibomian gland morphogenesis. Corneal tissue destruction in an adult KO mouse might be due to not only lagophthalmos but also to an impaired morphogenesis of cornea, Meibomian glands, and orbicularis oculi muscle.  相似文献

目的:探讨二补助育汤对胚胎着床障碍模型小鼠子宫内膜形态及血管生成素-1(Ang-1)mRNA、血管内皮生长因子(VEGF)mRNA的表达和定位的影响。方法:24只ICR雌性小鼠随机分为空白组、模型组、戊酸雌二醇组、二补助育汤组,每组6只,用米非司酮建立胚胎着床障碍动物模型,各组给予相应药物灌胃,妊娠第5天处死小鼠后,检测各组妊娠率、平均着床位点数、子宫内膜Ang-1和VEGF mRNA表达量及其蛋白定位。结果:模型组小鼠平均胚胎着床位点数、Ang-1 mRNA、VEGF mRNA表达量明显低于空白组(均P<0.05);与模型组比较,二补助育汤组平均胚胎着床位点数、Ang-1 mRNA、VEGF mRNA表达量显著提高(均P<0.05)。结论:二补助育汤可提高子宫内膜Ang-1和VEGF蛋白表达量,促进子宫内膜血管生成,从而提高子宫内膜容受性。  相似文献

Mammary epithelial cells (MECs) play an important role against Streptococcus uberis infection which is one of the main causes of bovine mastitis and a potential threat to human health. Toll-like receptors (TLRs) and their mediated signaling pathways are critical in both innate and infection responses, yet their roles in anti-S. uberis infection in MECs remains poorly defined. In this work we investigated the regulatory mechanisms of TLR2 in inflammatory responses, where WT and TLR2−/− mice were euthanized at 15–18 days gestation, and mammary gland tissues were collected aseptically. The mouse MECs (MMECs) were isolated by combined digestion with type I collagenase, hyaluronidase and trypsin. We challenged MMECs with S. uberis and quantified antioxidant capacity as well as reactive oxygen species (ROS), proinflammatory cytokines and cell damage at different times. The loss of TLR2 function in MMECs results in more serious cell damage, increased cell adhesion, and significantly decreased ROS and mitochondrial ROS (mROS) with bactericidal function in response to S. uberis infection. Moreover, it was observed that the antioxidant capacity declined, and the production of TLR2-mediated cytokines (except CXC ligand 15) also were reduced. We demonstrated that TLR2 can mediate cellular anti-infective processes in MMECs by regulating the production of ROS and mROS and the secretion of cytokines. The results suggest an unpredicted role of TLR2 in MMECs in response to S. uberis infection.  相似文献

Adenovirus infections are a major cause of epidemic keratoconjunctivitis (EKC), which can lead to corneal subepithelial infiltrates and multifocal corneal opacity. In the current study, we investigated the use of an E1/E3-deleted adenovirus serotype 5 (Ad5) vector as a vaccine administered intramuscularly (IM) or intranasally (IN) against subsequent challenges with a luciferase-expressing Ad5 (Ad5-Luci) vector via eyedrop. We evaluated the adaptive immune response to Ad5 vector vaccination and confirmed a robust polyfunctional CD8 T cell response in splenic cells. Neutralizing Ad5 antibodies were also measured in the sera of vaccinated mice as well as Ad5 antibody in the eye wash solutions. Upon challenge with Ad5-Luci vector 8 weeks post the primary immunization, transduction was significantly reduced by > 70% in the vaccinated mice, which was slightly better in IM- vs. that in IN-vaccinated animals. Resistance to subsequent challenge was observed 10 months post primary IM vaccination, with sustained reduction up to 60% in the Ad5-Luci vector transduction. Passive immunization of naive mice with antisera from IM to vaccinated mice subsequently challenged with the Ad5-Luci vector resulted in approximately 40% loss in transduction efficiency. Furthermore, the mice that received IM immunization with or without CD8 T cell depletion showed > 40% and 70% reductions, respectively, in Ad8 genomic copies after Ad8 topical challenge. We conclude that Ad-vector vaccination successfully induced an adaptive immune response that prevented subsequent Ad transduction in the cornea and conjunctiva-associated tissues in a mouse model of adenovirus keratoconjunctivitis, and that both cellular and humoral immunity play an important role in preventing Ad transduction.  相似文献

ObjectiveNeuropsychiatric lupus (NPSLE), a manifestation of the autoimmune disease systemic lupus erythematosus (SLE), is characterized by psychiatric symptoms including anxiety and depression and upregulated autoantibodies. The B6.Nba2 spontaneous mouse model develops SLE, but has not previously been tested for NPSLE.MethodsWe investigated the NPSLE phenotype in male and female B6.Nba2 mice (n = 12 each) and age- and sex-matched B6 controls (n = 10 each) via behavioral assessments for anxiety, depression, and memory deficits. Serum anti-dsDNA, anti-nRNP, anti-DWEYS peptide reactive IgG autoantibody levels and soluble TWEAK levels were determined by ELISA. Hippocampal regions were stained for activated microglia and neurons.ResultsBoth male and female B6.Nba2 mice showed elevated anti-dsDNA IgG, anti-nRNP IgG and anti-DWEYS reactive antibodies, elevated serum soluble TWEAK levels, and a strong anxiety and depression phenotype (p < 0.05–0.0001). Male B6.Nba2 mice developed this phenotype at a slightly older age than females. Female B6.Nba2 mice displayed reduced numbers of neurons in the hippocampal region compared to female B6 controls (p < 0.05).ConclusionThe B6.Nba2 mouse model recapitulates many known NPSLE phenotypes, making it a promising model to investigate the development of NPSLE in the context of SLE.  相似文献

The successful licensure of vaccines for biodefense is contingent upon the availability of well-established correlates of protection (CoP) in at least two animal species that can be applied to humans, without the need to assess efficacy in the clinic. In this report we describe a multivariate model that combines pre-challenge serum antibody endpoint titers (EPT) and values derived from an epitope profiling immune-competition capture (EPICC) assay as a predictor in mice of vaccine-mediated immunity against ricin toxin (RT), a Category B biothreat. EPICC is a modified competition ELISA in which serum samples from vaccinated mice were assessed for their ability to inhibit the capture of soluble, biotinylated (b)-RT by a panel of immobilized monoclonal antibodies (mAbs) directed against four immunodominant toxin-neutralizing regions on the enzymatic A chain (RTA) of RT. In a test cohort of mice (n = 40) vaccinated with suboptimal doses of the RTA subunit vaccine, RiVax®, we identified two mAbs, PB10 and SyH7, which had EPICC inhibition values in pre-challenge serum samples that correlated with survival following a challenge with 5 × LD50 of RT administered by intraperitoneal (IP) injection. Analysis of a larger cohort of mice (n = 645) revealed that a multivariate model combining endpoint titers and EPICC values for PB10 and SyH7 as predictive variables had significantly higher statistical power than any one of the independent variables alone. Establishing the correlates of vaccine-mediated protection in mice represents an important steppingstone in the development of RiVax® as a medical countermeasure under the United States Food and Drug Administration’s “Animal Rule.”  相似文献

目的探讨同种氨基酸形成多肽的毒性和应用前景。方法用固相合成法合成天冬氨酸和赖氨酸的2~10肽单链与双链化合物,通过小鼠尾静脉将这些化合物注射到小鼠肝脏内,观察它们对小鼠的致死作用(LD50)。结果当天冬氨酸单链2~10肽的浓度依次达到0.15、0.1、0.09、0.03、0.0065、0.03、0.034、0.035和0.04 mol/L时,小鼠的死亡率达到50%。当赖氨酸单链2~10肽的浓度依次达到0.28、0.1、0.047、0.0225、0.0028、0.00166、0.0015、0.0011和0.00075 mol/L时,小鼠的死亡率达到50%。当天冬氨酸-赖氨酸双链2~10肽的浓度依次得到0.5、0.05、0.017、0.014、0.009、0.006、0.004、0.004、0.004、0.004 mol/L时,小鼠的死亡率达到50%。结论单链多肽的毒性大于双链多肽。在2~6肽范围内,天冬氨酸单链多肽随着氨基酸数目增加毒性逐渐增加;相反,在7~10肽范围内,随着氨基酸数目的增加毒性逐渐降低。在2~10肽范围内,赖氨酸单链多肽随着氨基酸数目增加毒性逐渐增加。在2~6肽范围内,天冬氨酸-赖氨酸双链多肽随着氨基酸数目增加毒性逐渐增加;但在7~10肽范围内,其毒性相似。  相似文献
