 Immunocytochemically detectable MT and p53 have been found more commonly in comedo DCIS of the breast with high-grade cytology. The aim of this study is to confirm these findings and to investigate the relationship between MT and p53 in a single large series of cases of DCIS of the breast. To this end, 127 cases of DCIS were classified histologically according to architecture, cytonuclear differentiation (grade), presence and extent of intraduct necrosis, and using the Van Nuys system. Sections were immunostained for p53 and MT (E9) using established techniques, and the extent and intensity of staining were assessed semi-quantitively. The results confirmed that there was generally more MT and p53 positivity in poorly differentiated (grade 3) DCIS with extensive necrosis and that MT expression was greater in grade 2 lesions than p53 expression. However, overall there was no statistically significant correlation between p53 and MT staining. The results indicate that MT and p53 overexpression may arise from independent mechanisms in early breast neoplasia. Received: 3 July 1996 / Accepted: 5 November 1996  相似文献

为探讨金属硫蛋白(MT)对豚鼠乳头肌缺血再灌注损伤所致心律失常的影响,利用标准玻璃微电极技术,采用缺氧及复氧豚鼠乳头肌模型,模拟体内缺血再灌注损伤,观察不同浓度MT对豚鼠乳头肌电生理特性的影响。结果显示低浓度的MT(0.002mmol/L)对正常及缺氧和复氧豚鼠乳头肌的动作电位(AP)有关参数及自律性均无影响;中等浓度的MT(0.02mmol/L)仅使正常乳头肌的AP复极达50%时程(APD50)缩短24%(P<0.05),但使缺氧乳头肌的AP复极达20%时程(APD20)、APD50和AP复极达90%时程(APD90)分别缩短68%、56%和43%(P均<0.01),并使静息电位(RP)、AP幅值(APA)和0相最大上升速率(Vmax)分别增加30%、30%和45%(P均<0.01);高浓度的MT(0.1mmol/L)使正常豚鼠乳头肌的APD20、APD50和APD90分别缩短57%、54%和50%(P均<0.01),并且RP、APA及Vmax分别下降22%(P<0.05)、28%(P<0.01)和29%(P<0.05),而使缺氧豚鼠乳头肌的APD20、APD50和APD90分别延长92%、78%和50%(P均<0.01),对RP、APA及Vmax无明显影响。在复氧期间,0.02mmol/L的MT可使自律性的发生率从77.8%降至55.6%(P<0.05);而0.1mmol/L的MT则使自律性的发生率从77.8%降至22.2%(P  相似文献

目的:观察低强度红激光照射对大鼠脑主动脉金属硫蛋白(metallothionein,MT)生成的影响,探讨激光照射的细胞保护机制。方法:取24只大鼠的胸主动脉,将其制成厚度为0.5mm的薄片,置于含10%胎牛血清Krebs-Henseleit液6孔培养板,经95%O2-5%CO2、37℃孵育。设对照组及3个实验组,对照组不照射激光,3个实验组分别以功率密度为3、6和12J/cm^2的632nm波长的激光照射。采用竞争性RT-PCR法测定胸主动脉内MT的mRNA水平变化,以^109Cd牛血红蛋白饱和法测定MT的含量。结果:3个实验组MT1 mRNA水平分别比对照组增加175%(P>0.05)、418.6%(P<0.05)和668.2%(P<0.01);MT2 mRNA水平分别比对照组增加163.9(P>0.05)、615%、(P<0.01)和1087%(P<0.01);相应的MT蛋白含量分别比对照组增加142.8%(P<0.05)、170.8%(P<0.05)和142.1%(P<0.05)。  相似文献

目的:研究金属硫蛋白(MT)对同型半胱氨酸(Hcy)诱导大鼠血管平滑肌细胞(vascularsmoothmusclecells,VSMCs)增殖的影响及其作用机制。方法:以[3H]-TdR掺入法测定VSMCs增殖程度,免疫沉淀法测定VSMCs内丝裂素活化蛋白激酶(MAPK)活性,[109Cd]-血红蛋白饱和法测定MT含量,硫代巴比妥酸法测定丙二醛(MDA)含量,NADH氧化法测定乳酸脱氢酶(LDH)漏出量。结果:Hcy(10-6-10-4mmol/L)呈浓度依赖性的刺激培养大鼠VSMCs[3H]-TdR掺入,0.1mmol/LHcy刺激[3H]-TdR掺入比对照组高4.2倍(P<0.01)。Hcy亦呈浓度依赖性地激活VSMCsMAPK活性、增加细胞MDA的生成和LDH的漏出(P均<0.01)。单独MT孵育,对VSMCs的上述指标均无明显影响(P>0.05)。但MT(10-6-10-4mol/L)呈浓度依赖性抑制100μmol/LHcy的促增殖效应(r=0.98,P<0.01)。MT显著抑制Hcy对VSMCs的MAPK活性、MDA生成和LDH漏出的激活作用(均P<0.01)。以0.5mmol/LZnCl2预孵育6h后,VSMCsMT含量比非诱导细胞高5.7倍(P<0.01),这种内源性MT高表达的细胞,显著抵抗Hcy刺激的-TdR掺入和MAPK激活;抑制Hcy的促细胞MDA生成与LDH漏出效应(均P<0.01)。结论:MT能有效抑制Hcy促大鼠VMSCs增殖作用,其机制可能与MT拮抗Hcy对MAPK的激活和其抗氧化作用有关。  相似文献

目的 :观察Zn2 + 预处理诱导金属硫蛋白 (MT)在心肌细胞的表达及对离体大鼠缺血再灌注损伤心肌的保护作用。方法 :3 2只Sprague Dawley大鼠随机分为对照组及实验组 ,实验组按腹腔注射ZnSO4 到建立再灌注模型的时间分为E1 2h,E2 4h及E4 8h组 ,每组各 8只。检测LDH ,CK、ATP含量及心功能指标 (LVSP与±dp/dtmax) ,用1 0 9Cd/血红素饱和法测量心肌组织中金属硫蛋白的含量。结果 :E2 4h及E4 8h组MT表达量较对照组高 (P <0 .0 1) ,E1 2h组虽有表达 ,但其与对照组间差异无显著性。E2 4h及E4 8h组LDH与CK较其它二组低 ,而ATP较其它二组高 ,心功能指标较其它二组改善。E1 2h组及对照组之间相应各指标 (LDH ,CK、ATP、LVSP与±dp/dtmax)间差异无显著性 (P >0 .0 5 )。结论 :Zn2 + 预处理可诱导MT在心肌细胞的表达而对离体大鼠缺血再灌注损伤心肌具有保护作用。  相似文献

Summary In a 61-year-old woman, who had been exposed for 20 years to cadmium in the production of Ni-Cd batteries, nephrectomy of the contracted kidney was performed. The removed kidney was examined histologically and the cadmium concentration was determined in the cortex (44.97 g g–1) and in the medulla (7.71 g g–1). The homogenates of the renal cortex and medulla were subjected to gel filtration on Sephadex G-75. In the cortex, as well as the medulla, cadmium was predominantly found in the low-molecular (metallothionein) fraction, but in the cortex, Cd content in this fraction was six times higher than in the medulla. The determination of SH groups and proteins in high- and low-molecular fractions indicates an induction of the metallothionein formation primarily in the renal cortex.  相似文献

In order to study the critical concentration of cadmium (Cd) in acute renal dysfunction following Cd, male mice were injected IV with Cd complexed with cysteine. The critical concentration was 10 g Cd/g wet weight in whole kidney and it was the same as that for Cdthionein (Cd-Th), which may suggest that the toxicity of Cd-Th is due to Cd ions liberated from Cd-Th in the kidneys. Renal Cd concentration was at first higher than the critical concentration, but decreased to the critical concentration by 24 h after administration. As an index for renal dysfunction, the uptake of p-aminohippurate (PAH) by renal cortical slices in vitro was sensitive, and showed the different time-course from those of urinary protein and glucose levels. The results suggest the usefulness of PAH uptake as an index. Incidental to the renal dysfunction, renal calcium levels exhibited a marked increase.  相似文献

The mechanisms associated with metallothionein (MT) gene regulation are complex and poorly understood. Only a modest increase in brain MT expression levels is attained by exposure to metals, MT gene transfection, and MT gene knock-in techniques. Accordingly, in the present study, MT null astrocytes isolated from transgenic mice deficient in MT-I and MT-II genes were introduced as a zero background model of MT expression. MT protein levels were determined by western blot analysis. MT proteins in MT-I and MT-II null astrocytes were undetectable. Transient MT-I gene transfection increased the levels of foreign MT expression in MT-I and MT-II null astrocytes by 2.3-fold above basal levels in wild-type astrocytes. Intracellular Na(2)51CrO(4) efflux and D-[2,3-3H]aspartate uptake were studied as indices of acute methylmercury (MeHg) (5 microM) cytotoxicity. In MT-I and MT-II knockout astrocytes MeHg led to significant (p<0.01) increase in Na(2)51CrO(4) efflux and a significant (p<0.05) decrease in the initial rate (1 min) of D-[2, 3-3H]aspartate uptake compared to MT-I and MT-II knockout controls. Transfection of the MT-I gene in MT-I and MT-II null mice significantly (p<0.01) decreased the effect of MeHg on Na(2)51CrO(4) efflux in MT null, as well as wild-type astrocytes. MT-I gene transfection in MT-I and MT-II null astrocytes reversed the inhibitory effect of MeHg on D-[2,3-3H]aspartate uptake, such that initial rates of uptake in MT-I transfected cells in the presence and absence of MeHg (5 microM) were indistinguishable. These results demonstrate that: (1) astrocytes lacking MTs are more sensitive to MeHg than those with basal MT protein levels, (2) the MT-I gene can be overexpressed in MT-I and MT-II null astrocytes by transient MT-I gene transfection, and (3) that foreign MT expression endows astrocytes with increased resistance to MeHg.  相似文献

目的:探讨金属硫蛋白在食管鳞癌中的表达及其与癌细胞增殖、凋亡的关系。方法:采用免疫组化法检测92例食管鳞癌组织中的金属硫蛋白(MT)、增殖细胞核相关抗原(Ki-67抗原)的表达,并用TUNEL法检测细胞凋亡指数。结果:92例食管鳞癌标本中65例MT呈阳性表达,阳性率为70.6%,明显高于正常食管粘膜上皮组织(3/20,P<0.01);MT表达与食管鳞癌患者年龄、性别、浸润深度及病变长度无关,而与组织分化程度、TNM分期、淋巴结转移密切相关,MT与Ki-67抗原表达强度呈正相关,Pearson列联系数=0.227(P<0.05)。随MT表达强度的增加,癌组织凋亡指数逐渐降低。结论:MT表达与食管鳞癌的发生、发展密切相关,MT能促进癌细胞增殖,抑制细胞凋亡,可作为食管鳞癌治疗的新靶点。  相似文献
