本实验探讨了内源性速激肽是否参与白三烯C4(LTC4)的气道效应.LTC4(0.5μgkg-1,iv)可增高豚鼠肺内压(IPP)和气道内依文思蓝渗出。速激肽NK-1受体拮抗剂CP-96345{(2S,3S)-顺式-2-(二苯甲基)-N-[(2-甲氧苯)-甲基]-1-杂氮双环[2.2.2]辛烷-3-胺}1mgkg-1,iv,可减弱LTC4诱导的依文思蓝渗出;NK-2受体拮抗剂SR-48968{(S)-N-甲基-N-[4-(4-乙酰氨基-4-苯基哌啶)-2-(3,4-二氯苯基)丁基]苯甲酰胺},1mgkg-1,iv,可抑制IPP的增高.白三烯拮抗剂ONO-1078(0.03mgkg-1,iv)可阻断这两种反应.结果说明内源性速激肽增强LTC4的气道作用,其中NK-1受体介导微血管渗漏,NK-2受体介导支气管收缩.  相似文献

Ethnopharmacological relevance

Roots of Asparagus racemosus Willd (Shatavari in vernacular) are widely used in Ayurveda as Rasayana for immunostimulation, galactogogue as also in treatment of conditions like ulcers and cancer. Various studies have indicated immunomodulatory properties of Shatavari root extracts and formulations.

Aim of the study

To study the effect of standardized Asparagus racemosus root aqueous extract (ARE) on systemic Th1/Th2 immunity of SRBC sensitized animals.

Materials and methods

We used HPTLC to quantify steroidal saponins (Shatavarin IV, Immunoside®) and flow cytometry to study effects of ARE on Th1/Th2 immunity. SRBC specific antibody titres and DTH responses were also monitored as markers of Th2 and Th1 responses, respectively. We also studied lymphocyte proliferation. Cyclosporin, cyclophosphamide and levamisole were used as controls.

Results

Treatment with ARE (100 mg/(kg b.w. p.o.)) resulted in significant increase of CD3+ and CD4/CD8+ percentages suggesting its effect on T cell activation. ARE treated animals showed significant up-regulation of Th1 (IL-2, IFN-g) and Th2 (IL-4) cytokines suggesting its mixed Th1/Th2 adjuvant activity. Consistent to this, ARE also showed higher antibody titres and DTH responses. ARE, in combination with LPS, Con A or SRBC, produced a significant proliferation suggesting effect on activated lymphocytes.

Conclusion

The study suggests mixed Th1/Th2 activity of ARE supports its immunoadjuvant potential.  相似文献

The objective of this study was to determine whether the development of tolerance to CP 55,940, a potent cannabinoid agonist, was due to changes in the receptor or second messenger system. ICR mice treated with CP 55,940 (2 mg/kg) twice a day for 6 and one-half days developed a high degree of tolerance to the pharmacological effects of CP 55,940. The ability of CP 55,940 to produce motor hypoactivity, hypothermia and immobility was reduced 163-, 97- and 19-fold, respectively. Evaluation of 3H-CP 55,940 binding to rat brain membranes indicated no difference in receptor affinity between the vehicle- and CP 55,940-treated animals. However, these binding studies revealed a 50% decrease in receptor number in the cerebellum of the CP 55,940-tolerant mice. Although cAMP is generally considered to be the second messenger for cannabinoid receptors, little difference was observed in the inhibitory effects of CP 55,940 on adenylyl cyclase activity in cerebellum between vehicle and drug-treated mice. However, there was an increase in receptor mRNA which suggests a compensation for receptor loss. There are several possible explanations for these results. There may be sufficient spare receptors such that CP 55,940-tolerant mice are capable of producing a maximal effect on the second messenger system. On the other hand, one could conclude that cannabinoid receptor down-regulation does not account for the development of tolerance to all of the effects of CP 55,940 in mice.  相似文献

中央监护系统能够实现对病人的生理或生化参数连续、长时间、自动、实时监测,并经分析、处理后实现多类别自动报警、自动记录及对诊断结果进行综合判断等。传统的多参数生理监护系统主要是通过各种专用电缆来实现网络化,在增加产品成本的同时也加大了施工的难度。本文在分析了无线收发芯片XE1201的工作原理基础上,重点介绍了应用其开发无线收发模块的原理和方法,并利用此模块构建了无线中央监护系统,继而对整个系统的性能进行了测试。该系统已在实验室中实现,并对心电(ECG)、无创血压、血氧饱和度(SPO2)等多生理参数进行实时监护,将信息提供给医院内的网络,以供访问使用。  相似文献

胃癌组织中KAI1、nm23及P53的表达及其临床意义   总被引:5,自引:4,他引:1  
目的:探讨正常胃黏膜、不典型增生胃黏膜及癌组织中KAI1、nm23及P53蛋白的表达.方法:应用SP法免疫组化检测22例正常胃黏膜,65例不典型增生胃黏膜及74N胃癌组织中的KAI1、nm23及P53蛋白的表达.结果:正常胃黏膜、不典型增生胃黏膜及胃癌组织中,KAI1和nm23阳性率呈降低趋势,组间差异性有统计学意义(x2=20.885, P<0.001;x2=29.133,P<0.05):P53蛋白阳性表达率呈增加趋势,组间差异性有统计学意义(x2=21.954,P<0.001).Fisher精确概率检验显示:在胃癌组中不同的浸润深度、有无淋巴结转移和脉管侵犯组内KAI1、nm23及 P53组阳性表达率的差异性有统计学意义(x2 =20.885,P<0.001;x2=29.133,P<0.05;x2= 21.954,P<0.001);而在年龄、性别组间的差异性无统计学意义.Spearman等级相关分析显示 KAI1与nm23表达呈正相关(r=0.859,P<0.05); KAI1与P53表达呈负相关(r=-0.859,P<0.05), nm23与P53表达呈负相关(r=-0.874,P<0.05) 结论:抑癌基因KAI1与nm23的缺失以及P53 蛋白的过表达可能是胃癌发生、发展及浸润和转移的重要原因之一.  相似文献

To investigate recruitment of slow-twitch (ST) and fast-twitch (FT) muscle fibres, as well as the involvement of the various quadriceps femoris muscle portions during repeated, intense, one-legged knee-extensor exercise, 12 healthy male subjects performed two 3-min exercise bouts at ~110% maximum thigh O2 consumption (EX1 and EX2) separated by 6 min rest. Single-fibre metabolites were determined in successive muscle biopsies obtained from the vastus lateralis muscle (n=6) and intra-muscular temperatures were continuously measured at six quadriceps muscle sites (n=6). Creatine phosphate (CP) had decreased (P<0.05) by 27, 73 and 88% in ST fibres and 25, 71 and 89% in FT fibres after 15 and 180 s of EX1 and after 180 s of EX2, respectively. CP was below resting mean–1 SD in 15, 46, 84 and 100% of the ST fibres and 9, 48, 85 and 100% of the FT fibres at rest, after 15 and 180 s of EX1 and after 180 s of EX2, respectively. A significant muscle temperature increase (Tm) occurred within 2–4 s at all quadriceps muscle sites. Tm varied less than 10% between sites during EX1, but was 23% higher (P<0.05) in the vastus lateralis than in the rectus femoris muscle during EX2. Tm in the vastus lateralis was 101 and 109% of the mean quadriceps value during EX1 and EX2, respectively. We conclude that both fibre types and all quadriceps muscle portions are recruited at the onset of intense knee-extensor exercise, that essentially all quadriceps muscle fibres are activated during repeated intense exercise and that metabolic measurements in the vastus lateralis muscle provide a good indication of the whole-quadriceps muscle metabolism during repeated, intense, one-legged knee-extensor exercise.  相似文献

The effect of various immunomodulators on the induction of experimental autoimmune encephalomyelitis (EAE) is evaluated in the Lewis rat. Bordetella pertussis (BP) is the optimal inductor of EAE in this rat strain. Treatment of the animals with BP either before or after or simultaneously with guinea-pig spinal cord preparation (GpSC) resulted in an EAE about two weeks thereafter. Additional injection of living BCG, of CFA, IFA (incomplete Freund's adjuvant) or Vibrio cholerae neuraminidase (VCN) did not augment or mitigate the effect induced by BP or GpSC. Living BCG, IFA, VCN or Corynebacterium parvum (CP) did not induce EAE when given in combination with GpSC but without BP. CFA combined with GpSC only occasionally induced EAE. However, EAE could be induced by the combination of CFA and GpSC or IFA and GpSC in a part of the animals tested if they had been pretreated or simultaneously been injected with living BCG by intravenous route. EAE could not be enhanced by the additional injection of VCN. Surprisingly, most of the animals peracutely died after injection of CFA and BP in combination with GpSC when they had been pretreated with CP. This effect was most pronounced when pretreatment was done on day -4. No acute effect could be seen when CP was given simultaneously to CFA, BP and GpSC. Animals which did not peracutely succumb developed EAE similarly as those in the positive control groups. CP treatment simultaneously with BP but without CFA resulted in a reduction of the EAE specific mortality. 目的 为了研究上田法与Vojta等法对脑瘫儿运动功能改善情况。方法 将 5 8例脑瘫儿 (包括 3岁以上及重症脑瘫儿 )分为年龄相同的两组 ,分别采用上田法与Vojta等法治疗 ,疗程≥ 1个月。 结果 上田组的坐位功能改善较Vojta组明显 (P <0 0 1)。在上田组中 ,3岁以上脑瘫儿治疗前、后总分平均为 (5 7 0 4± 31 92 )分和 (78 5 2± 2 0 83)分 ,重症脑瘫儿治疗前、后总分平均为 (10 5 4± 9 6 1)分和 (31 5 2± 15 0 3)分。而Vojta组中 ,3岁以上脑瘫儿治疗前、后总分平均为 (73 33± 31 86 )分和 (80 37± 2 4 76 )分 ,重症脑瘫儿治疗前、后总分平均为 (2 3 5 5± 2 1 98)分和 (2 9 33± 2 5 2 1)分。结论 上田法对脑瘫儿坐位功能改善优于Vojta等法 ,尤其对 3岁以上脑瘫儿及重症脑瘫儿效果更佳  相似文献

5-Fluorouracil (5-FU) is currently being used as an anticancer drug to reduce tumor bulk in order to increase the operability rate and postoperative survival in patients with cervical cancer, which has been combined with cisplatin (CP) because of its superior activities observed in human carcinoma cells. However, the combined anticancer effect of 5-FU and CP in cervical carcinoma cells is poorly understood. Therefore, we conducted a study to investigate whether anticancer drugs 5-FU and CP may exhibit the combined antiproliferative effect in cervical carcinoma cells. Using proteomics analysis, including two-dimensional gel electrophoresis and matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF-MS), we investigated the antiproliferative effect-related proteins after treatment with 5-FU and/or CP. Our experiments showed that the combination of 5-FU and CP engaged both the apoptotic pathways: the membrane death receptor-mediated apoptosis pathway and the mitochondrial apoptotic pathway. Moreover, the combination of 5-FU and CP resulted in remarkable increasing susceptibility to apoptosis. We suggest that the combination of 5-FU and CP suppresses the growth of cervical carcinoma cells by synergistic effect with the induction of apoptosis. In vitro synergistic effect of 5-FU and CP supports the basis of the clinical application of the combination chemotherapy to the patients with cervical cancer.  相似文献
