Age-related cognitive decline is the major cause of concern due to its 70% more incidence than dementia cases worldwide. Moreover, aging is also the major risk factor of Alzheimer's disease (AD), associated with progressive memory loss. Approx. 13 million people will have Alzheimer-related memory decline by 2050. Learning and memory is the fundamental process of brain functions. However, the mechanism for the same is still under investigation. Thus, it is critical to understand the process of memory consolidation in the brain and extrapolate its understanding to the memory decline mechanism. Research on learning and memory has identified several molecular signatures such as Protein kinase M zeta (PKMζ), Calcium/calmodulin-dependent protein kinase II (CaMKII), Brain-derived neurotrophic factor (BDNF), cAMP-response element binding protein (CREB) and Activity-regulated cytoskeleton-associated protein (Arc) crucial for the maintenance and stabilization of long-term memory in the brain. Interestingly, memory decline in AD has also been linked to the abnormality in expressing these memory-related molecular signatures. Hence, in the present consolidated review, we explored the role of these memory-related molecular signatures in long-term memory consolidation. Additionally, the effect of amyloid-beta toxicity on these molecular signatures is discussed in detail.  相似文献

Increased life expectancy creates the prospect of prolonged marital intimacy. However, mature couples face many challenges — physiological, cultural, and familial — and they must be able to mobilize their individual and relational resources to maintain good levels of marital and sexual satisfaction. The aim of this study is to highlight various factors that contribute to the conjugal and sexual satisfaction of older couples, and to discuss the stressing of these factors in a crisis context. 534 participants aged between 18 and 74 years old answered several questionnaires measuring their degree of sexual and conjugal satisfaction as well as the areas of strength and fragility in the relational dynamics of their couple. In addition, five couples also accepted a semi-structured interview on their relationship dynamics and their vision of the ageing couple. The results highlight the major impact of several variables contributing to an increase in relational and sexual satisfaction within couples (e.g. health concerns, conflict management, fondness…). Furthermore, a comparison between the scores of the over-51s and the younger ones highlights the areas of strength of older partners. These different factors will be discussed. Finally, we will propone therapeutic advices aimed at supporting the areas of strength of mature couples in order to accompany them towards long-term relational and sexual fulfillment.  相似文献

Low- and middle-income countries (LMICs) bear the greatest burden of cardiovascular disease (CVD) worldwide. Emerging evidence, mostly from industrialized countries, suggest that neighborhood characteristics influence cardiovascular health. This study presents results from 27,797 participants living in six LMICs on the association of perceived neighborhood social cohesion, the degree of connectedness among neighbors, with 10-year risk of CVD. We observed that greater perceived neighborhood social cohesion was associated with lower odds of high (>20%) 10-year CVD risk (OR = 0.67, 95%CI: 0.53–0.86). These novel findings, suggest that interventions to enhance social cohesion may have beneficial effects on cardiovascular health in LMICs.  相似文献

Monocytes play important roles in anti-microbial and anti-viral responses and chronic inflammatory diseases. Monocytes' functions are altered by aging. We investigated age-changes in calcium (Ca2+) response to CCL2 and LPS in human monocytes. CCL2 and LPS induced a slow increase of the cytosolic Ca2+ level, with a maximum response at ~360 s and ~300 s, respectively, in monocytes of young and older adults. No difference was observed in the magnitude and in the Ca2+ kinetic with both stimuli. Furthermore, store-operated Ca2+ entry and plasma membrane expression of ORAI1 showed no difference between both groups. In summary, monocytes from older adults maintained the capacity to mobilize calcium as their counterparts in young adults suggesting that the mechanisms underlying the dysfunctions in monocytes in aging might not involve alterations in Ca2+ flow through the plasma membrane.  相似文献

IntroductionLong-term care hospitals (LTCHs) are at a high risk for the inflow and spread of antimicrobial resistance (AMR) pathogens. However, owing to limited laboratory resources, little is known about the extent to which AMR organisms are endemic.MethodsWe performed active surveillance for carbapenem-resistant Enterobacteriaceae (CRE) and extended-spectrum beta-lactamase-producing Enterobacteriaceae (ESBL-E) in newly admitted patients at Marugame Medical Center, a nearly 200-bedded LTCH located in Kagawa, Japan. From August to December 2021, we tested stool samples from patients wearing diapers and confirmed the genetic variants using specific PCR assays. We also collected clinical variables and compared them between AMR carriers and non-carriers.ResultsStool samples were collected from 75 patients, with a median age of 84 years. CRE strain was not detected, but 37 strains of ESBL-E were isolated from 32 patients (42.7%). During the study period, 4.9% of in-hospital patients (37 per 756 patients) were identified to be ESBL-E carriers in the routine microbiological processing, suggesting that active surveillance detected approximately 9-fold more ESBL-E carriers. The blaCTX-M-9 group was the most common (38.5%), followed by the blaTEM (26.9%). The clinical backgrounds of the ESBL-E non-carriers and carriers were not significantly different.ConclusionOur active screening demonstrated that nearly half of the patients hospitalized or transferred to a Japanese LTCH were colonized with ESBL-E. We highlight the enforcement of universal basic infection prevention techniques at LTCHs where patients carrying AMR pathogens gather.  相似文献

BackgroundThe presence of fatigue has been shown to modify running biomechanics. Throughout a run individuals become more fatigued, and the effectiveness of the musculoskeletal protective mechanism can diminish. Older adults are at an elevated risk for sustaining an overuse running related injury. This can be partially explained by changes in the musculoskeletal system and load attenuation.Research questionThe purpose was to compare post-fatigue running mechanics between older and younger runners.MethodsThirty runners (15 young, 15 older) between the ages of 18–65 participated in this study. All participants ran at least 15 miles/week. Running kinematics were captured using a 10-camera motion capture system while participants ran over a 10-m runway with force platforms collecting kinetic data under two conditions: C1: rested state at a controlled pace of 3.5 m/s ( ± 5%); C2: post-exertional protocol where pace was not controlled, rather it was monitored based on heartrate and RPE representative of somewhat-hard to hard intensity exercise. Prior to C2, participants underwent an exertional protocol that consisted of a maximal exercise test to induce fatigue and a required cool-down. A 2 (state of fatigue) x 2 (age) MANOVA was run to test for the effects of fatigue and age and their interactions. Results: No state of fatigue x age interaction was observed. A main effect of age for peak knee extension moment (Y > O; p = 0.01), maximum knee power (Y > O; p = 0.04), maximum hip power (O >Y; p = 0.04), and peak vertical ground reaction force (Y > O; p = 0.007). Regardless of age, participants exhibited decreased knee ROM (p = 0.007) and greater hip extension moment (p < 0.001) in C2 compared to C1.ConclusionWhile different in knee and hip mechanics overall, the subtle differences observed demonstrate that older runners exhibit comparable gait adaptions post-fatigue to younger volume-matched runners.  相似文献
